Differentiation of mouse trophoblast stem cells (TSCs) to trophoblast giant cells (TGCs) has been widely used as a model system to study placental development and function. While several differentially expressed genes, including regulators of TSC differentiation, have been identified, a comprehensive analysis of the global expression of genes and splice variants in the two cell types has not been reported.

Here, we report ~ 7800 differentially expressed genes in TGCs compared to TSCs which include regulators of the cell cycle, apoptosis, cytoskeleton, cell mobility, embryo implantation, metabolism, and various signaling pathways. We show that several mitotic proteins, including Aurora A kinase, were downregulated in TGCs and that the activity of Aurora A kinase is required for the maintenance of TSCs. We also identify hitherto undiscovered, cell-type specific alternative splicing events in 31 genes in the two cell types. Finally, we also report 19 novel exons in 12 genes which are expressed in both TSCs and TGCs.

Overall, our results uncover several potential regulators of TSC differentiation and TGC function, thereby providing a valuable resource for developmental and molecular biologists interested in the study of stem cell differentiation and embryonic development.

Many studies have proven effective to clear pathological tau species in animal models of AD, and several clinical trials using conventional immunotherapy with anti-tau native antibodies are currently active. We have previously generated a vectorized scFv derived from the conformation-dependent anti-tau antibody MC1, scFvMC1, and demonstrated that its intracranial injection was able to prevent tau pathology in adult tau mice. Here, we show that, in a prevention paradigm and in two different tau transgenic models (JNPL3 and P301S), a one-time intramuscular injection of AAV1-scFvMC1 generated a long-lasting peripheral source of anti-tau scFvMC1 and significantly reduced insoluble and soluble tau species in the brain. Moreover, our data showed that scFvMC1 was internalized by the microglia, in the absence of overt inflammation. Textual corpora are extremely important for various NLP applications as they provide information necessary for creating, setting and testing those applications and the corresponding tools. They are also crucial for designing reliable methods and reproducible results. Yet, in some areas, such as the medical area, due to confidentiality or to ethical reasons, it is complicated or even impossible to access representative textual data. We propose the CAS corpus built with clinical cases, such as they are reported in the published scientific literature in French.

Currently, the corpus contains 4,900 clinical cases in French, totaling nearly 1.7M word occurrences. Some clinical cases are associated with discussions. A subset of the whole set of cases is enriched with morpho-syntactic (PoS-tagging, lemmatization) and semantic (the UMLS concepts, negation, uncertainty) annotations. The corpus is being continuously enriched with new clinical cases and annotations. The CAS corpus has been compared with similar clinical narratives. When computed on tokenized and lowercase words, the Jaccard index indicates that the similarity between clinical cases and narratives reaches up to 0.9727.

We assume that the CAS corpus can be effectively exploited for the development and testing of NLP tools and methods. Besides, the corpus will be used in NLP challenges and distributed to the research community.

Non-alcoholic fatty liver is one of the most common comorbidities of diabetes. Also MCS had a good therapeutic effect on type 2 diabetes and NAFLD rats by regulating glucose and lipid metabolism. Sixteen potential biomarkers had been identified based on metabolomics which were related to the corresponding pathways including Pantothenate and CoA biosynthesis, fatty acid biosynthesis, citric acid cycle, arginine biosynthesis, tryptophan metabolism, etc.
