This study examined how inclusion of dried distillers' grains with solubles (DDGS) in finishing lamb diets for different periods affects some physiological indicators and meat quality. A total of 40 male lambs were divided into four groups according to feeding regimen during the 120-day finishing period as follows C120 no DDGS included in diet for 120 days; D120 DDGS included in diet for 120 days; D75 no DDGS included in diet for 45 days + DDGS included in diet for 75 days; D45 no DDGS included in diet for 75 days + DDGS included in diet for 45 days. Dietary inclusion rate of DDGS was 27.5%. Fattening performance and rumen parameters were not affected by treatment. Feeding regimens had no significant effect on meat quality except the instrumental tenderness and juiciness score. DDGS fed lambs had higher level of total trans fatty acids and n-6/n-3 ratio in meat. 10t-C181, 11t-C181 and 9c,11t conjugated linoleic acid contents of adipose tissue were higher in all the DDGS groups compared with C120 lambs (p less then .001). These results suggest that soybean meal can be replaced with corn DDGS in lamb diets for up to 120 days during the finishing period with no adverse effects on some physiological response feedlot performance and meat quality.Combination therapy with BRAF and MEK inhibitors (BRAFi/MEKi) have dramatically improved prognosis among patients with BRAF-mutant metastatic melanoma compared with traditional treatment, such as chemotherapy. However, resistance to these targeted agents occurs invariably, thereby limiting their clinical efficacy. Recently, it has been reported that the ligand-independent phosphorylation of erythropoietin-producing hepatocellular receptor A2 (EphA2) at Ser-897 signaling is a driver of BRAF inhibitor resistance in melanoma. A melanoma patient with multiple metastases was treated with dabrafenib plus trametinib therapy and maintained complete remission for more than 2 years. As brain metastasis occurred, we had switched to nivolumab plus ipilimumab therapy. However, new lesions were observed after four cycles of nivolumab plus ipilimumab therapy, she was rechallenged with encorafenib plus binimetinib therapy, and she maintained progression-free status for more than 7 months. We performed immunohistochemical staining of EphA2, phospho-EphA2 (p-EphA2; Ser-897), and epidermal growth factor receptor (EGFR) of melanoma cells before and/or after dabrafenib and trametinib therapy. Immunohistochemical examination showed higher expression of EphA2, p-EphA2, and EGFR in the melanoma cells after dabrafenib plus trametinib therapy as compared with that before therapy. Our results may indicate that EphA2, p-EphA2, and EGFR can be critical factors for resistance and reversible response of BRAFi/MEKi in metastases of melanoma. Our case presents a possible treatment that can help overcome BRAFi/MEKi resistance and improve prognosis of melanoma.
Circular RNA_0004913 (circ_0004913), circular RNA_0008160 (circ_0008160), and circular RNA_0000517 (circ_0000517) are shown to be dysregulated in HCC tissues and cell lines, and also show potential in regulating hepatocellular carcinoma (HCC) pathogenesis. This current study attempted to find possible associations of circ_0004913, circ_0008160, and circ_0000517 with clinical features and prognosis of HCC patients.
A hundred and fifty HCC patients who received hepatectomy were retrospectively reviewed, and their resected specimens including tumor tissues and paired adjacent tissues were obtained, in which the circ_0004913, circ_0008160, and circ_0000517 expressions were detected. Overall survival (OS) data were collected according to the clinical visit records.
Circ_0004913 (p<0.001) and circ_0008160 (p<0.001) were downregulated, while circ_0000517 (p<0.001) was upregulated in tumor tissue compared with paired adjacent tissue. Additionally, tumor circ_0004913 was negatively associated with largest tumor size (p=0.009) and Barcelona clinic liver cancer (BCLC) stage (p=0.020), while tumor circ_0008160 and circ_0000517 were not correlated with any clinicopathological features of HCC patients (all p>0.05). Tumor circ_0004913 high expression was associated with prolonged OS in total HCC patients (p=0.008) and in subgroup of patients with largest tumor size <5cm (p=0.008). Tumor circ_0008160 high expression was correlated with longer OS in patients with BCLC stage B (p=0.026). Univariate Cox's analysis disclosed that tumor circ_0004913 high expression was correlated with longer OS; while after adjustment by multivariate Cox's analysis, it failed to predict OS independently.

Functional development of the fetal cardiac autonomic nervous system (cANS) plays a key role in fetal maturation and can be assessed through fetal heart rate variability (fHRV)-analysis, with each HRV parameter representing different aspects of cANS activity. Current available techniques, however, are unable to assess the fHRV parameters accurately throughout the whole pregnancy. This study aims to test the feasibility of color tissue Doppler imaging (cTDI) as a new ultrasound technique for HRV analysis. Secondly, we explored time trends of fHRV parameters using this technique.
18 healthy singleton fetuses were examined sequentially every 8weeks from 10weeks GA onwards. From each examination, 3 cTDI recordings of the four-chamber view of 10seconds were retrieved to determine accurate beat-to-beat intervals. The fHRV parameters SDNN, RMSSD, SDNN/RMSSD, and pNN10, each representing different functional aspects of the cANS, were measured, and time trends during pregnancy were explored using spline functions within a linear mixed-effects model.
In total, 77% (95% Cl 66-87%) of examinations were feasible for fHRV analysis from the first trimester onwards, which is a great improvement compared to other techniques. The technique is able to determine different maturation rates of the fHRV parameters, showing that cANS function, presumably parasympathetic activity, establishes around 20weeks GA and matures rapidly until 30weeks GA.
This is the first study able to assess cANS function through fHRV analysis from the first trimester onwards. The use of cTDI to determine beat-to-beat intervals seems feasible in just 3 clips of 10seconds, which holds promise for future clinical use in assessing fetal well-being.
