Anthocyanins have been reported to have effective chemopreventive activity. Lycium ruthenicum Murray is rich in anthocyanins and exhibits many biological activities. The purpose of this study was to investigate the effects and possible biological mechanism of the main anthocyanin monomer (Pt3G) of Lycium ruthenicum Murray on prostate cancer DU-145 cells. The cell proliferation was detected by methyl thiazolyl tetrazolium assay. The cell apoptosis rates were assessed by flow cytometric analysis and TUNEL assay. The expressions of apoptosis related proteins were evaluated by western blotting. Our data demonstrated that Pt3G inhibited cell proliferation, induced apoptosis and promoted cell cycle arrest at the S phase in a concentration-dependent manner (0, 100, 200 and 400 μg mL-1). Furthermore, it was shown that Pt3G decreased the mitochondrial membrane permeability through regulating the expressions of Bax and Bcl-2. Western blot analysis indicated that Pt3G significantly increased the expression of PTEN and then activated the PI3K/Akt-mediated caspase 3 pathway. In addition, our results also suggested that Pt3G activated the PTEN gene to induce the apoptosis of DU-145 cells by stimulating the overproduction of ROS. To sum up, these results indicate that Pt3G inhibits cell proliferation and induces apoptosis through the ROS/PTEN/PI3K/Akt/caspase 3 signaling pathway in prostate cancer DU-145 cells. Therefore, Pt3G of Lycium ruthenicum Murray may be a potential anti-proliferative agent for the prevention or treatment of prostate cancer.A series of short intrinsically disordered polypeptide conjugated oligonucleotides (IDPOCs) were rationally developed and assembled into well-defined nanospheres. The nanospheres exhibited excellent reversible thermoresponsive regulation of their contraction and expansion. Furthermore, the nanospheres showed biocompatibility, drug encapsulation and effective cellular uptake. The Cu(i)-catalyzed ring-opening cross-coupling of an oxabicyclic olefin with an organoboron reagent provides access to cis-2-aryl-1,2-dihydronaphthalen-1-ol derivatives, in contrast to the exclusive trans-selectivity in related Cu-catalyzed reactions with Grignard reagents. DFT calculations suggest that the reaction possibly proceeds via boronic ester by-product assisted ring-opening as an alternative pathway to the canonical β-oxygen elimination as the rate-determining step. The discharge of excessive metal ions and anions into water bodies leads to the serious pollution of water and environment, which in turn has a certain impact on industry, agriculture, and human life. Because of the unique advantages of luminescent metal-organic frameworks (LMOFs), they have been successfully explored as various fluorescent probes to quickly and effectively detect these pollutants. This perspective not only introduces the design strategy and classification of LMOFs, especially the construction methods of water-stable LMOFs, but also reports the latest progresses in some LMOFs between 2016 and 2020 as well as expounds the mechanisms of LMOFs for detecting anions and cations. Moreover, the luminescence properties of LMOFs are related to the selection of metal ions, the structure of organic ligands, the pore size, and the interaction of guest molecules. Finally, the further development of LMOFs is summarized and prospected in this field.Herein, we report the in situ analysis of human telomerase by a multicolor DNA tetrahedron nanoprobe. The elongated telomeric repeats can hybridize with settled molecular beacons in order, accompanied by sequentially lighted up fluorescence. Imaging telomerase activity, real-time monitoring telomerase action and determining product length distribution in living cells are realized. It detects multiple information of intracellular telomerase and provides deeper insights into the function of telomerase. The present article focuses on fluorescent probes based on Ru(ii)-polypyridyl fragments connected to different chelates, their significant roles in coordination chemistry and their applications in the selective recognition of biorelevant Cu2+ and Fe2+/Fe3+ ions. A critical analysis of the detection of metal ions by both "turn-on" and "turn-off" emission, properties, mechanism and applications of these sensors is presented. Emphasis has also been laid on the efficiency, selectivity and specificity of these sensors towards biologically important Cu2+ and Fe2+/Fe3+ ions.A "two-in-one" strategy was employed to construct 3D-COFs for the first time. Based on this strategy, a 3D-Flu-COF could be readily synthesized in various simplex organic solvents. Endocrinology inhibitor Benefitting from the non-conjugated structure, the 3D-Flu-COF showcased excellent acidichromic sensing performance with good sensitivity, reversibility and naked eye visibility. We use the model system ethanol-dodecane to demonstrate that giant critical fluctuations induced by easily accessible weak centrifugal fields as low as 2000g can be observed above the miscibility gap of a binary liquid mixture. Moreover, several degrees above the phase transition, i.e. in the one-phase region, strong gradients of ethanol concentration occur upon centrifugation. In this case, the standard interpretation of sedimentation equilibrium in the analytical ultracentrifuge (AUC) yields an apparent molar mass of ethanol three orders of magnitude higher than the real value. Notably, these composition gradients have no influence on the distribution gradient of solutes such as dyes like Nile red. The thick opaque interphase formed upon centrifugation does not appear as the commonly observed sharp meniscus, but as a turbidity zone, similar to critical opalescence. This layer is a few millimeters thick and separates two fluids with low compositional gradients. All these effects can be qualitatively understood and explained using the Flory-Huggins solution model coupled to classical density functional theory (DFT). In this domain hetero-phase fluctuations can be triggered by gravity even far from the critical point. Taking into account Jean Perrin's approach to external fields in colloids, a self-consistent definition of the Flory effective volume and an explicit calculation of the total free energy per unit volume is possible.
