Antimicrobial resistance (AMR) is a major public health threat, and inappropriate antimicrobial use (AMU) in food animal production can contribute to the global burden of AMR in humans. This study was conducted to understand knowledge, attitude, and practice (KAP) of smallholder livestock owners regarding antimicrobial use, residue, and resistance in three agro-ecological zones and production systems in Ethiopia. A cross-sectional study based on structured interviews was conducted. Twenty-one items were used to assess farmers' KAP. Ionomycin order Item response theory (IRT) model and Cronbach's alpha were used to assess the KAP measurement scales. Inferential analyses were used to compare the differences in the practices in terms of the farm and socio-economic characteristics. There was a difference in the type of antimicrobials reported use between agro-ecological zones and production systems. Pastoralists most commonly used antibiotics (86.7%) followed by anthelminthics (70.8%). Overall, tetracyclines (36.4%), aminoglycosiement, supervision, and guidance can also lead to more efficient antimicrobial use by smallholder livestock owners. The study also highlights the need for research into the development of usable tools that measure antibiotic knowledge and attitudes. In this context, alleviating the tightening grain crop supply and ensuring the healthy development of animal husbandry through innovations in protein feedstuff production remain considerable challenges. Moringa oleifera is a miracle tree species with abundant nutrients, high protein biological value, and good feeding effect. As a new protein feedstuff, M. oleifera has great potential in alleviating the feeding crisis. Here, we review available literature regarding the characterization of M. oleifera in the field of animal husbandry in terms of nutrient content, digestion, and absorption characteristics, and feeding effects and present current challenges in using M. oleifera as animal feed. Copyright © 2020 Su and Chen.The present study was designed to evaluate the effects of heat that induces oxidative stress on reproduction organ weight and serum biochemical, testes structure, and function in male guinea pig (Cavia porcellus). Forty-eight male guinea pigs with an average weight of 330.56 ± 23.62 g, aged 3-4 months, were distributed into four groups of 12 animals each. One group (control) was maintained to ambient temperature (20-25°C), while other groups (Groups 2-4) were exposed daily for 6 h, to 32 ± 1°C, 39±1°C, and 46 ± 1°C, respectively. All animals were sacrificed after 60 days' exposure and their reproductive characteristics values were determined. Results revealed a significant decrease (p less then 0.05) of the weight of testes, epididymis, vas deferens, and accessory glands in cavies exposed to the highest temperature investigated (46 ± 1°C), compared to the control animals. There was a significant (p less then 0.05) reduction of serum testosterone and LH levels in all heat stress-exposed groups (≥46 ± 1°C) when compared to the control group. Heat stress significantly (p less then 0.05) decreased sperm mobility, sperm count, and testicular antioxidant enzymes, while increasing testicular malondialdehyde content. However, the serum level of HSP-40 increased in the animals exposed to 39 ± 1°C and decreased when the cavies were exposed to 46 ± 1°C. In conclusion, exposure to heat-induced oxidative stress results in impairment of reproduction organ weight and serum biochemical, testes structure, and function in male cavies. Cellular pathways that include proteasomal destruction and autophagic disposal are components of normal proteostasis. Here we report a novel molecule that plays a non-redundant role in maintaining homeostasis, Fas Apoptosis Inhibitory Molecule (FAIM). FAIM is highly conserved throughout evolution and bears no homology to any other protein. We found that FAIM counteracts heat and oxidative stress-induced loss of cell viability. FAIM is recruited to ubiquitinated proteins induced by cellular stress and the levels of stress-induced protein aggregates are much greater in FAIM-deficient cell lines. Primary fibroblasts from FAIM-deficient mice showed the same proteostasis deficits as cell lines. Administration of a mediator of oxidative stress to FAIM-deficient animals induced more ubiquitinated protein aggregates and more organ damage as compared to wild type mice. These results identify a completely new actor that protects cells against stress-induced loss of viability by preventing protein aggregation. To determine the gene expression, single nucleotide polymorphisms, and co-mutations participating in the initiation and progression of lung adenocarcinoma, we comprehensively analyzed the data of 491 patients from The Cancer Genome Atlas. Using log-rank and Kruskal-Wallis analysis, Oncoprint, Kaplan-Meier survival plots, and a nomogram, we found that EGFRL858R with co-mutation TP53 was significant prognostic determinant versus that with co-wild TP53 (hazard ratio, 2.77, P = 0.012). Further gene co-expression network and functional enrichment analysis indicated that co-mutation of EGFRL858R/TP53 increases the expression of COMP and ITGB8, which are involved in extracellular matrix organization and cell surface receptor signaling pathways, thus contributing to poor prognosis in lung adenocarcinoma. Validation was performed using three GEO profiles along with colony formation and CCK-8 assays for proliferation, transwell and wound-healing for migration in transfected H1299 and A549 cell lines. To the best of our knowledge, these results are the first to indicate that patients harboring the co-mutation of EGFRL858R/TP53 show increased expression of COMP and ITGB8, which participate in extracellular matrix dysfunction and can be used as prognostic biomarkers in patients with lung adenocarcinoma.
