But anti-inflammatory IL-10 significantly decreased. Th1 cytokines IL-2 and interferon-γ (IFN-γ) did not change, and Th2 cytokines IL-5 increased by 1.65 times and IL-10 decreased by 0.82 times. Conclusion Acute severe air pollution exposure can lead to inflammatory response in lung tissues of rats. The secretion of chemokines eotaxin, MCP-1, MIP-2, RANTES and proinflammatory cytokines IL-1β, IL-17, IL-18, TNF-α are promoted in this process. The infiltrated T cells in lung tissues are dominated by Th2 cells. Objective To perform in vitro characterization and comparison of stromal cell antigen 1 (STRO-1) positive (STRO-1+) and negative (STRO-1-) cell populations from human periodontal ligament cells sorted by magnetic-activated cell sorting (MACS). Glycochenodeoxycholic acid Methods Periodontal ligament cells were isolated from the periodontal ligament by tissue digesting method. The cells were magnetically sorted using anti-STRO-1 antibodies. Subsequently, cells were characterized by flow cytometry for the expression of CD90, CD105, CD29, STRO-1, CD34 and CD45. Colony-forming assay was used to detect the ability of colony-forming. When the cells are induced to differentiate by osteogenesis and lipogenesis, alizarin red staining, and oil red O staining were used to identify the pluripotent differentiation of STRO-1+ and STRO-1- cells. Results STRO-1+ cells highly expressed CD105 and CD90 and lowly expressed the haematopoietic markers CD45 and CD34. In contrast to STRO-1- cells, STRO-1+ cells showed a significantly higher number of colony-forming units (CFUs). STRO-1+ cells were demonstrated to have osteogenic and adipogenic differentiation potential. Conclusion STRO-1+ cells isolated by MACS are human periodontal ligament stem cells with stem cell characteristics. Objective To investigate the effect of Brucella abortus mutant strain S2308δrfbE on the maturation and immune response of mouse bone marrow-derived dendritic cells (BMDCs). Methods The expression of major histocompatibility complex I (MHC I), MHC II, CD40, CD86 were detected by flow cytometry; the secretion of cytokines tumor necrosis factor α (TNF-α), interleukin 6 (IL-6), IL-10 was detected by ELISA in S2308δrfbE-infected mouse BMDCs. Results The number of cells expressing MHC I, MHC II, CD40, CD86 and the secretion of cytokines TNF-α, IL-6 and IL-10 significantly increased in BMDCs infected with S2308δrfbE as compared with control groups. Conclusion S2308δrfbE promotes the maturation and enhances the inflammatory cytokines secretion in BMDCs.Objective To analyze the differentially expressed genes (DEGs) in alveolar macrophages (AMs) of patients with chronic obstructive pulmonary disease (COPD) and their potential roles in the pathogeneses of COPD using bioinformatics. Methods Gene chip and RNA sequencing data sets of AMs in patients with COPD were downloaded from GEO. Limma and Degseq2 packages in R software were applied to obtain DEGs, and the GO enrichment analysis, the Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis, protein-protein interaction analysis (PPI), and the hub gene analysis were performed to predict the molecular mechanisms of DEGs. Results Through the integration of three data sets, a total of 43 DEGs of AMs were obtained, and the function predictive analysis found that the 43 DEGs were primarily related to chemokines, cytokines, complement, cytochrome P450, etc., which mainly included the significantly low expression of C-X-C motif chemokine ligand 9 (CXCL9), CXCL11, etc. and the significantly high expression of cytochrome P450 family 1 subfamily B member 1 (CYP1B1). Conclusion The DEGs of AMs in patients with COPD are related to the molecular mechanisms of immunity and inflammation and might be involved in the pathogenesis of chronic inflammation of COPD.Plasmids are autonomously replicating sequences that help cells adapt to diverse stresses. Theta plasmids are the most frequent plasmid class in enterobacteria. They co-opt two host replication mechanisms replication at oriC, a DnaA-dependent pathway leading to replisome assembly (theta class A), and replication fork restart, a PriA-dependent pathway leading to primosome assembly through primer extension and D-loop formation (theta classes B, C, and D). To ensure autonomy from the host's replication and to facilitate copy number regulation, theta plasmids have unique mechanisms of replication initiation at the plasmid origin of replication (ori). Tight plasmid copy number regulation is essential because of the major and direct impact plasmid gene dosage has on gene expression. The timing of plasmid replication and segregation are also critical for optimizing plasmid gene expression. Therefore, we propose that plasmid replication needs to be understood in its biological context, where complex origins of replication (redundant origins, mosaic and cointegrated replicons), plasmid segregation, and toxin-antitoxin systems are often present. Highlighting their tight functional integration with ori function, we show that both partition and toxin-antitoxin systems tend to be encoded in close physical proximity to the ori in a large collection of Escherichia coli plasmids. We also propose that adaptation of plasmids to their host optimizes their contribution to the host's fitness while restricting access to broad genetic diversity, and we argue that this trade-off between adaptation to host and access to genetic diversity is likely a determinant factor shaping the distribution of replicons in populations of enterobacteria.
To investigate factors associated with anaemia in preschool children.
A home survey was conducted in 2018. Anaemia in children (capillary blood Hb level < 110 g/l) was the outcome, and socio-economic, demographic and health factors of the mother and child were the independent variables. The measure of association was the prevalence ratio, and its 95 % CI was calculated using Poisson's regression with robust variance and hierarchical selection of independent variables.
Afro-descendants communities living in the state of Alagoas, northeast Brazil.
Children aged 6-59 months and their mothers (n 428 pairs).
The prevalence of child anaemia was 38·1 % (95 % CI 33·5, 42·7). The associated factors with child anaemia were male sex, age < 24 months, larger number of residents at home (> 4), relatively taller mothers (highest tertile) and higher z-score of BMI for age, after further adjustment for wealth index, vitamin A supplementation in the past 6 months and clinical visit in the last 30 d.
The high prevalence of anaemia observed reveals a relevant public health problem amongst children under five from the quilombola communities of Alagoas.
