Although altered structural-functional connectivity couplings (SC-FC couplings) have been previously reported in schizophrenia patients, the alterations in SC-FC couplings of different illness stages of schizophrenia (SZ) remain largely unknown. In this study, we collected structural and resting-state functional MRI data from 73 normal controls (NCs), 61 first-episode (FeSZ) and 78 chronic (CSZ) schizophrenia patients. Positive and negative syndrome scale (PANSS) scores were assessed for all patients. Structural and functional brain networks were constructed using gray matter volume (GMV) and resting-state magnetic resonance imaging (rs-fMRI) time series measurements. At the connectivity level, the CSZ patients showed significantly increased SC-FC coupling strength compared with the FeSZ patients. At the node strength level, significant decreased SC-FC coupling strength was observed in the FeSZ patients compared to that of the NCs, and the coupling strength was positively correlated with negative PANSS scores. These results demonstrated divergent alterations of SC-FC couplings in FeSZ and CSZ patients. Our findings provide new insight into the neuropathological mechanisms underlying the developmental course of SZ.Visual working memory (VWM) relies on sustained neural activities that code information via various oscillatory frequencies. Previous studies, however, have emphasized time-frequency power changes, while overlooking the possibility that rhythmic amplitude variations can also code frequency-specific VWM information in a completely different dimension. Here, we employed the recently-developed Holo-Hilbert spectral analysis to characterize such nonlinear amplitude modulation(s) (AM) underlying VWM in the frontoparietal systems. We found that the strength of AM in mid-frontal beta and gamma oscillations during late VWM maintenance and VWM retrieval correlated with people's VWM performance. When behavioral performance was altered with transcranial electric stimulation, AM power changes during late VWM maintenance in beta, but not gamma, tracked participants' VWM variations. This beta AM likely codes information by varying its amplitude in theta period for long-range propagation, as our connectivity analysis revealed that interareal theta-beta couplings-bidirectional between mid-frontal and right-parietal during VWM maintenance and unidirectional from right-parietal to left-middle-occipital during late VWM maintenance and retrieval-underpins VWM performance and individual differences.
Laccase is one member of the blue multicopper oxidase family. It can catalyze the oxidation of various substrates. The Thermus thermophilus SG0.5JP17-16 laccase (lacTT) is thermostable, pH-stable, and high tolerance to halides, and can decolorize the synthetic dyes. In lacTT, the function of the loop 6 constructing the substrate-binding pocket wasn't clear.

The residues Asp394 and Asp396 located in loop 6, and were used to probe how the loop 6 influenced catalytic properties of the laccase. Site-directed mutagenesis was performed for two amino acids. Kinetic assay was utilized to characterize the catalytic efficiency of mutants. Mutants with different catalytic activities were used to decolorize the synthetic dyes to clarify the relationship between the catalytic efficiency and dye decolorization. Redox potential, structural and spectral analyses were performed to explain the differences in laccase activity between wild type and mutant enzymes.

D394M, D394E and D394R mutants with the lower laccase activity displayed a decreased decolorization efficiency, while D396A, D396M and D396E mutant enzymes with higher catalytic efficiency decolorized the synthetic dye more efficiently than the wild type enzyme.

The pocket loop 6 might experience a conformational dynamics. The D394 residue controlled this conformation change by amino acid interaction networks containing the D396 residue at the entrance of substrate channel.

These studies may provide clues to improve the activity of the laccase for the better use in industrial applications, and/or contribute to further understanding the mechanism of laccase oxidation on the substrate.
