CBSI programs have been introduced to improve the physical and psychological well-being of older people. CBSI programs encourage older people to (1) take care of themselves and their peers, (2) sustain their well-being, and (3) create a cohesive and inclusive community. Although the emergence of CBSI programs is a global phenomenon, the variations, effectiveness, and sustainability of these programs are unknown. To uncover information about the Japanese version of CBSI programs, we conducted observations and face-to-face interviews with related personnel at two CBSI programs in rural areas of Japan in 2018. We found both positive and negative aspects in the current form of CBSI programs. As for the positives, these programs promote older people's physical and psychological well-being and enhance community cohesiveness. However, CBSI programs face challenges, including that groups tend to be gender and age specific men and the younger-old are less likely to show interest. A group-specific approach to CBSI programs could cause future community division, which would be contrary to the goal. Given the continued advancement of the aging population, a new approach to participant recruitment is needed.Characteristics of early childhood education and care (ECEC) centers might be relevant for children's health. This scoping review aims to provide an overview of the association between meso-level characteristics (MLCs) of ECEC centers with children's health, health behavior, and wellbeing. Five databases were searched for quantitative and qualitative research articles published in English or German since 1 January 2000 on health, health behavior, and wellbeing of children aged 0 to 6 years considering MLCs of ECEC centers. Two authors screened 10,396 potentially eligible manuscripts and identified 117 papers, including 3077 examinations of the association between MLCs and children's health indicators (Kappas > 0.91). Five categories of MLCs were identified (1) structural characteristics, (2) equipment/furnishings, (3) location, (4) facilities/environment, (5) culture/activities/policies/practices, and 6) staff. Only very few studies found an association of MLCs with body weight/obesity, and general health and wellbeing. Especially physical activity and mental health were related to MLCs. In general, the location (rural vs. urban, neighborhood status) seemed to be a relevant health aspect. MLCs of ECEC centers appeared relevant for child health indicators to different degrees. Future research should focus on these associations, in detail, to identify concrete ECEC indicators that can support health promotion in early childhood. The patterning of electrospun fibers is a key technology applicable to various fields. This study reports a novel focused patterning method for electrospun nanofibers that uses a cylindrical dielectric guide. The finite elements method (FEM) was used to analyze the electric field focusing phenomenon and ground its explanation in established theory. The horizontal and vertical electric field strengths in the simulation are shown to be key factors in determining the spatial distribution of nanofibers. The experimental results demonstrate a relationship between the size of the cylindrical dielectric guide and that of the electrospun area accumulated in the collector. By concentrating the electric field, we were able to fabricate a pattern of less than 6 mm. The demonstration of continuous line and square patterning shows that the electrospun area can be well controlled. This novel patterning method can be used in a variety of applications, such as sensors, biomedical devices, batteries, and composites.Enzymes, once considered static molecular machines acting in defined spatial patterns and sites of action, move to different intra- and extracellular locations, changing their function. This topological regulation revealed a close cross-talk between proteases and signaling events involving post-translational modifications, membrane tyrosine kinase receptors and G-protein coupled receptors, motor proteins shuttling cargos in intracellular vesicles, and small-molecule messengers. Here, we highlight recent advances in our knowledge of regulation and function of ADisintegrin And Metalloproteinase (ADAM) endopeptidases at specific subcellular sites, or in multimolecular complexes, with a special focus on ADAM10, and tumor necrosis factor-α convertase (TACE/ADAM17), since these two enzymes belong to the same family, share selected substrates and bioactivity. We will discuss some examples of ADAM10 activity modulated by changing partners and subcellular compartmentalization, with the underlying hypothesis that restraining protease activity by spatial segregation is a complex and powerful regulatory tool.Pancreatic fibrosis is caused by the excessive deposits of extracellular matrix (ECM) and collagen fibers during repeated necrosis to repair damaged pancreatic tissue. Tanespimycin cost Pancreatic fibrosis is frequently present in chronic pancreatitis (CP) and pancreatic cancer (PC). Clinically, pancreatic fibrosis is a pathological feature of pancreatitis and pancreatic cancer. However, many new studies have found that pancreatic fibrosis is involved in the transformation from pancreatitis to pancreatic cancer. Thus, the role of fibrosis in the crosstalk between pancreatitis and pancreatic cancer is critical and still elusive; therefore, it deserves more attention. Here, we review the development of pancreatic fibrosis in inflammation and cancer, and we discuss the therapeutic strategies for alleviating pancreatic fibrosis. We further propose that cellular stress response might be a key driver that links fibrosis to cancer initiation and progression. Therefore, targeting stress proteins, such as nuclear protein 1 (NUPR1), could be an interesting strategy for pancreatic fibrosis and PC treatment.
