With mice, cellular stress response, interferon gamma-, adipocytokine- and chemokine signaling provided a rationale for the AOP of immune-mediated hepatitis. Despite all measures taken, immission of Pb onto agricultural soils still occurs, with fertilizer application, lead shot from hunting activities, and Pb from air deposition representing major sources. Little is known about the intermediate and long-term consequences of these emissions. To gain more insight, we established a mathematical model that considers input from fertilizer, ammunition, deposition from air, uptake of Pb by crops, and wash-out to simulate the resulting Pb concentrations in soil over extended periods. In a further step, human oral exposure by crop-based food was simulated and blood concentrations were derived to estimate the margin of exposure to Pb-induced toxic effects. Simulating current farming scenarios, a new equilibrium concentration of Pb in soil would be established after several centuries. The results suggested that GPX activity can be used as a more sensitive biomarker in contrast with GPX gene expression, and mRNA expressions of Hsp70, Hsp90 genes are more sensitive than Hsp40 for planarians in response to glyphosate stress. Besides, the deduced T-AOC as well as varied GPX activity and mRNA expression levels of Hsps also indicated that glyphosate exposure would inhibit antioxidation and induce oxidative stress in D. japonica, while specific antioxidant systems and stress proteins tried to protect cells by their own regulation. The results of this study will be helpful to elucidate the stress response mechanisms of freshwater planarians to herbicide glyphosate.The origin and distribution of hexavalent chromium, Cr (VI) over four seasonal cycles was investigated through a conceptual model that included three aquifer systems in Cyprus. An extensive water sampling grid covered two sea water intruded coastal aquifers, namely the Kokkinochoria (A1) and Kiti-Pervolia (A2) aquifers and the Troodos massif (A3). Analytical results give a first insight to the presence of Cr (VI) in Cyprus ground and surface waters. The highest Cr (VI) value of 26 μg L-1 is observed in the Troodos area (A3) where Cr(VI) is detected in all sampled water systems (surface and ground/spring). Nonetheless, the highest median values of 4.6 and 4.5 μg L-1 are observed in the Kokkinochoria (A1) and Kiti-Pervolia (A2) study areas respectively, where the local aquifers are under increased pressure from agriculture, urbanization and seawater intrusion. Stable isotopes were used to discern the effect of return irrigation and seawater intrusion. Stable isotope analyses from Troodos samples show two distinct groups of meteoric and near meteoric waters whereas isotopically enriched water is shown to correspond to the Kiti and Kokkinochoria area.AIMS/HYPOTHESIS We determined whether empagliflozin altered renal sympathetic nerve activity (RSNA) and baroreflexes in a diabetes model in conscious rabbits. METHODS Diabetes was induced by alloxan, and RSNA, mean arterial pressure (MAP) and heart rate were measured before and after 1 week of treatment with empagliflozin, insulin, the diuretic acetazolamide or the ACE inhibitor perindopril, or no treatment, in conscious rabbits. Lazertinib purchase RESULTS Four weeks after alloxan administration, blood glucose was threefold and MAP 9% higher than non-diabetic controls (p less then 0.05). One week of treatment with empagliflozin produced a stable fall in blood glucose (-43%) and increased water intake (+49%) but did not change RSNA, MAP or heart rate compared with untreated diabetic rabbits. The maximum RSNA to hypotension was augmented by 75% (p less then 0.01) in diabetic rabbits but the heart rate baroreflex was unaltered. Empagliflozin and acetazolamide reduced the augmentation of the RSNA baroreflex (p less then 0.05) to be similar to the non-diabetic group. Noradrenaline (norepinephrine) spillover was similar in untreated diabetic and non-diabetic rabbits but twofold greater in empagliflozin- and acetazolamide-treated rabbits (p less then 0.05). CONCLUSIONS/INTERPRETATION As empagliflozin can restore diabetes-induced augmented sympathetic reflexes, this may be beneficial in diabetic patients. A similar action of the diuretic acetazolamide suggests that the mechanism may involve increased sodium and water excretion. Graphical abstract.PURPOSE The purpose of this study is to evaluate diagnostic performance of a commercially available radiomics research prototype vs. an in-house radiomics software in the binary classification of CT images from patients with pancreatic ductal adenocarcinoma (PDAC) vs. healthy controls. MATERIALS AND METHODS In this retrospective case-control study, 190 patients with PDAC (97 men, 93 women; 66 ± 9 years) from 2012 to 2017 and 190 healthy potential renal donors (96 men, 94 women; 52 ± 8 years) without known pancreatic disease from 2005 to 2009 were identified from radiology and pathology databases. 3D volume of the pancreas was manually segmented from preoperative CT scans. Four hundred and seventy-eight radiomics features were extracted using in-house radiomics software. Eight hundred and fifty-four radiomics features were extracted using a commercially available research prototype. Random forest classifier was used for binary classification of PDAC vs. normal pancreas. Accuracy, sensitivity, and specificity of commercially available radiomics software were compared to in-house software.
