Many wheat varieties have the potential to develop unacceptably high levels of α-amylase in the grains if exposed to a cool temperature shock or simply cool temperature during the early to middle stages of grain filling. This phenomenon is referred to as late maturity α-amylase (LMA). The enzyme persists in the grain until harvest and may result in wheat with a low Falling Number that does not meet receival and export specifications. Resistance to LMA is therefore a valuable target for wheat breeders and wheat industries in general. Genetic evidence implicating a locus on the long arm of chromosome 7B in variation in LMA phenotype was confirmed in this investigation. Through intensive fine-mapping an ent-copalyl diphosphate synthase (CPS), hitherto named LMA-1, was identified as the likely candidate gene associated with variation in LMA phenotype. Single Nucleotide Polymorphisms (SNPs) within the LMA-1 coding sequence of Chinese Spring, Maringa and Halberd result in either prematurely terminated or functionalstanding of the role and potential impact of other genetic loci. Diagnostic markers for sources of resistance and SNP variation reported in this study should assist breeders to deploy resistance associated with LMA-1 variants in breeding programs.The post-transcriptional regulator RsmA globally controls gene expression in bacteria. Previous studies showed that RsmA2 and RsmA3 played critical roles in regulating type III secretion system (T3SS), motility, syringafactin, and alginate productions in Pseudomonas syringae pv. tomato strain DC3000 (PstDC3000). U0126 In this study, we investigated global gene expression profiles of the wild-type PstDC3000, the rsmA3 mutant, and the rsmA2/A3 double mutant in the hrp-inducing minimum medium (HMM) and King's B (KB) medium. By comparing the rsmA2/A3 and rsmA3 mutants to PstDC3000, a total of 1358 and 1074 differentially expressed genes (DEGs) in HMM, and 870 and 1463 DEGs in KB were uncovered, respectively. When comparing the rsmA2/A3 mutant with the rsmA3 mutant, 277 and 741 DEGs in HMM and KB, respectively, were revealed. Transcriptomic analysis revealed that the rsmY, rsmZ, and rsmX1-5 non-coding small RNAs (ncsRNAs) were positively affected by RsmA2 and RsmA3, while RsmA3 positively regulates the expression of thetential interplay among the RsmA proteins, which exhibit distinct and overlapping roles in modulating virulence and survival in P. syringae under different nutritional conditions.Wheat powdery mildew (Blumeria graminis f. sp. tritici) and stripe rust (Puccinia striiformis Westend f. sp. tritici) restrict wheat production in southwest China. Nitrogen fertilizers may influence outbreaks of these wheat diseases where wheat/faba beans are intercropped. To clarify how intercropping and varying nitrogen levels influence wheat powdery mildew and stripe rust and their relationship with crop yield, two consecutive field experiments were conducted from 2015 to 2017. Three cropping regimens (monocropped wheat, monocropped faba beans, and intercropped wheat/faba beans) and four nitrogen levels [N0 (0 kg⋅ha-1), N1 (90 kg⋅ha-1), N2 (180 kg⋅ha-1), and N3 (270 kg⋅ha-1)] were evaluated. In two consecutive planting seasons, the incidence and disease index of powdery mildew and stripe rust increased, while the disease index was more affected by nitrogen levels than their incidence. Both diseases were most prevalent at the N3 level. Compared with monocropping, intercropping (N0-N3 levels) reduced the incidence of powdery mildew by 2.8-37.0% and disease index by 15.5-47.4%, increased the relative control effect by 10.7-56.2 and 16.3-47.2%, reduced the incidence of stripe rust by 2.9-42.7% and disease index by 8.3-42.2%, and increased the relative control effect by 5.9-43.7 and 8.8-42.1%. The relative control efficacy of intercropping was most affected by N2 level. Intercropping yield increased with increasing nitrogen by 25.0-46.8%, and overall land equivalent ratio (LER) was 1.30-1.39. The correlation coefficient between disease index and wheat yield for both diseases was -0.7429 to -0.9942, a significant negative correlation, most significant at N1. Nitrogen regulation in intercropped wheat/faba beans can control powdery mildew and stripe rust, and optimize wheat yield. Intercropping at 180 kg ha-1 N2 resulted in the highest yield.ULTRAPETALA1 (ULT1) is a versatile plant-exclusive protein, initially described as a trithorax group (TrxG) factor that regulates transcriptional activation and counteracts polycomb group (PcG) repressor function. As part of TrxG, ULT1 interacts with ARABIDOPSIS TRITHORAX1 (ATX1) to regulate H3K4me3 activation mark deposition. However, our recent studies indicate that ULT1 can also act independently of ATX1. Moreover, the ULT1 ability to interact with transcription factors (TFs) and PcG proteins indicates that it is a versatile protein with other roles. Therefore, in this work we revised recent information about the function of Arabidopsis ULT1 to understand the roles of ULT1 in plant development. Furthermore, we discuss the molecular mechanisms of ULT1, highlighting its epigenetic role, in which ULT1 seems to have characteristics of an epigenetic molecular switch that regulates repression and activation processes via TrxG and PcG complexes.Panicle degradation causes severe yield reduction in rice. There are two main types of panicle degradation apical spikelet abortion and basal degeneration. In this study, we isolated and characterized the apical panicle abortion mutant apical spikelet abortion (asa), which exhibits degeneration and defects in the apical spikelets. This mutant had a pleiotropic phenotype, characterized by reduced plant height, increased tiller number, and decreased pollen fertility. Map-based cloning revealed that OsASA encodes a boric acid channel protein that showed the highest expression in the inflorescence, peduncle, and anther. RNA-seq analysis of the asa mutant vs wild-type (WT) plants revealed that biological processes related to reactive oxygen species (ROS) homeostasis and salicylic acid (SA) metabolism were significantly affected. Furthermore, the asa mutants had an increased SA level and H2O2 accumulation in the young panicles compared to the WT plants. Moreover, the SA level and the expression of OsPAL3, OsPAL4, and OsPAL6 genes (related to SA biosynthesis) were significantly increased under boron-deficient conditions in the asa mutant and in OsASA-knockout plants.
