Werewolf secara fundamental berbeda dari Animagus. Lycanthropy disebabkan oleh infeksi darah yang ditularkan melalui gigitan werewolf, sedangkan keterampilan animagus itu didapat melalui proses belajar. Manusia serigala tidak bisa menjaga pikiran mereka setelah transformasi (kecuali mereka meminum ramuan wolfbane ), juga tidak dapat mereka kontrol ketika mereka ingin berubah, sementara Animagi dapat melakukan keduanya. Manusia serigala diketahui dapat membahayakan manusia dan di paksa untuk melakukan transformasi ketika bulan purnama. Sedangkan animagus bertransformasi atas kehendaknya sendiri.
Kesimpulannya, Werewolf merupakan manusia serigala hanya bisa menjadi serigala dan hanya pada saat bulan purnama, yang disebabkan oleh infeksi darah dari gigitan werewolf. Sedangkan Animagus merupakan manusia serigala yang bisa berubah kapanpun dia mau.
