Recently, it was reported that leucine-rich repeat-containing G-protein-coupled receptor 4 (LGR4, also called GPR48) is another receptor for RANKL and was shown to compete with RANK to bind RANKL and suppress canonical RANK signaling during osteoclast differentiation. The critical role of the protein triad RANK-RANKL in osteoclastogenesis has made their binding an important target for the development of drugs against osteoporosis. In this study, point-mutations were introduced in the RANKL protein based on the crystal structure of the RANKL complex and its counterpart receptor RANK, and we investigated whether LGR4 signaling in the absence of the RANK signal could lead to the inhibition of osteoclastogenesis.; Methods The effects of point-mutated RANKL (mRANKL-MT) on osteoclastogenesis were assessed by tartrate-resistant acid phosphatase (TRAP), resorption pit formation, quantitative real-time polymerase chain reaction (qPCR), western blot, NFATc1 nuclear translocation, micro-CT and histomorphological assayt for osteoporosis by inhibiting RANKL-induced osteoclastogenesis via comparative inhibition of RANKL. Moreover, the mutant RANKL was found to lack the toxic side effects of most osteoporosis treatments.Aglaomorpha quercifolia (L.) Hovenkamp & S. Linds is an extensively used species in traditional medicinal systems in several areas of the world due to some important medicinal properties such as antioxidant, antibacterial, analgesic, and anti-inflammatory activities. In East Timor, different parts of this fern are used either as remedies or as food. The ingestion of a broth made from its rhizome improves lactation, and young fronds of this fern are boiled and eaten with rice by the locals. Nevertheless, its chemical profile is far from being established. The present work aims to establish the chemical profile of both rhizomes and leaves n-hexane extracts by Gas Chromatography- Mass Spectrometry (GC-MS). The results showed the leaves richness in fatty acids with interesting nutritional values (ω-6/ω-3 = 0.68, AI = 0.59, TI = 0.30), being linolenic acid (253.71 ± 0.93 mg/g dry leaves) and palmitic acid (237.27 ± 0.59 mg/g dry leaves) the significant compounds in the extract. Whereas the rhizome extract is mostly rich in terpenoids, such as steroid, cycloartane, and hopanoid derivatives, being hop-16-ene (166.45 ± 0.53 mg/g dry rhizome) and β-sitosterol (50.76 ± 0.11 mg/g dry rhizome) the major compounds. Several compounds are reported for the first time in the species, and the data herein reported contributes to confirming the species nutritional value.Autophagy has been suggested to have an important role in the pathogenesis of Sjögren syndrome (SS). We previously identified that autophagy related 5 (ATG5) was elevated in the tear and conjunctival epithelial cells of SS dry eyes (DE) compared to non-SS DE. The purpose of this study was to investigate the role of tear ATG5 as a potential biomarker in the diagnosis of SS. To confirm this hypothesis, we evaluated the tear ATG5 concentration, and other ocular tests (Schirmer I, tear breakup time (TBUT), ocular surface staining (OSS) score, ocular surface disease index (OSDI)) in SS and non-DE, and compared their diagnostic performance to discriminate SS from non-SS DE. Tear ATG5 showed the greatest area under the curve (AUC = 0.984; 95% CI, 0.930 to 0.999) among the tests, and a 94.6% sensitivity and 93.6% specificity at a cutoff value of >4.0 ng/mL/μg. Our data demonstrated that tear ATG5 may be helpful as an ocular biomarker to diagnose and assess SS. In the future, the diagnostic power of tear ATG for SS should be validated.Different Ag@TiO2 and Ag@ZnO catalysts, with nanowire (NW) structure, were synthesized containing different amounts of silver loading (1, 3, 5, and 10 wt.%) and characterized by FE-SEM, HRTEM, BET, XRD, Raman, XPS, and UV-vis. The photocatalytic activity of the composites was studied by the production of hydrogen via water splitting under UV-vis light and the degradation of the antibiotic ciprofloxacin. The maximum hydrogen production of all the silver-based catalysts was obtained with a silver loading of 10 wt.% under irradiation at 500 nm. Moreover, 10%Ag@TiO2 NWs was the catalyst with the highest activity in the hydrogen production reaction (1119 µmol/hg), being 18 times greater than the amount obtained with the pristine TiO2 NW catalyst. The most dramatic difference in hydrogen production was obtained with 10%Ag@TiO2-P25, 635 µmol/hg, being 36 times greater than the amount reported for the unmodified TiO2-P25 (18 µmol/hg). The enhancement of the catalytic activity is attributed to a synergism between the silver nanoparticles incorporated and the high surface area of the composites. In the case of the degradation of ciprofloxacin, all the silver-based catalysts degraded more than 70% of the antibiotic in 60 min. The catalyst that exhibited the best result was 3%Ag@ZnO commercial, with 99.72% of degradation. click here The control experiments and stability tests showed that photocatalysis was the route of degradation and the selected silver-based catalysts were stable after seven cycles, with less than 1% loss of efficiency per cycle. These results suggest that the catalysts could be employed in additional cycles without the need to be resynthesized, thus reducing remediation costs.The effect of compositing multiwalled carbon nanotubes (MWCNTs) with polystyrene (PS) to fabricate nanofibrous membrane by electrospinning technique and comparing the direct contact membrane distillation (DCMD) performance of the blank and composite membranes is evaluated numerically. Surface morphology of both the pristine and the composite membrane was studied by SEM imaging while the average fiber diameter and average pore size were measured using ImageJ software. Static water contact angle and porosities were also determined for both membranes. Results showed significant enhancement in both the hydrophobicity and porosity of the composite membrane by increasing the static water contact angle from 145.4° for the pristine PS membrane to 155° for the PS/MWCNTs composite membrane while the porosity was increased by 28%. Simulation results showed that at any given feed inlet temperature, the PS/MWCNTs membrane have higher permeate flux and better overall system performance.
