We evaluated the effects of applying a combination inoculant to 4 corn hybrids harvested at high moisture on their nutritive value and microbial populations. The treatment design was the factorial combination of corn hybrids ensiled with (INO) and without (CON) inoculant. The hybrids were TMF2R737 (MCN), F2F817 (MBR), P2089YHR (PCN), and PI144XR (PBR), ensiled at dry matter (DM) concentrations of 30.5, 26.3, 31.1, and 31.5%, respectively; MBR and PBR were brown midrib mutants (BMR). The inoculant contained Lactobacillus buchneri and Pediococcus pentosaceus (4 × 10 5 and 1 × 10 5 cfu/g of fresh corn). The experiment had a complete randomized design with treatments replicated 6 times. Corn was treated or not with inoculant, packed into 7.6L bucket silos, and stored for 100 d. At d 0, the relative abundance (RA, %) of Enterobacteriaceae was lower in PBR vs. the other hybrids [51.3 vs x= (average of) 58.4] and in the case of fungi, incertae sedis (i.s.) Tremellales and Mucoraceae were more and less abundant, resp to a taxonomically uneven one dominated by Lactobacillaceae. In the case of fungi, INO application increased the fungal diversity at d 100 mainly by reducing the dominance of Monascaceae vs. CON. In conclusion, the INO treatment overwhelmed the disparate microbial populations found across BMR and conventional hybrids ensiled at low DM concentrations and ensured a significant concentration of acetic acid that modified fungal populations and in turn extended the aerobic stability of all hybrids.The wool of the invasive, non-native hemlock woolly adelgid, Adelges tsugae Annand (Hemiptera Adelgidae), occurs mostly on hemlock (Tsuga sp.) twigs, but can be dislodged from the canopy and end up on the forest floor and tree stem underneath the canopy. Herein, we developed and tested the efficacy of two novel sampling techniques, which are based on a visual examination of the forest floor and the hemlock stem for A. tsugae wool. Subsequently, we compared these two techniques to a visual examination of foliage and ball sampling which are two methods currently used operationally. We sampled 11 hemlock stands, with low to moderate incidence A. tsugae populations, near Ithaca, New York in 2016 and assessed the probability of detecting wool on a tree and in a stand, as well as the relative variation and relative net precision for each of the four techniques. We found that sampling the tree stem outperformed foliage and ground sampling, likely because of its higher detection rate and lower relative variation, but not ball sampling. Our findings suggest that combining stem, ball and ground sampling was the most effective combination of techniques and gave a high probability of detecting an infested tree or an infested stand. All techniques were an improvement over foliage sampling, even after increasing the foliage sampling effort fivefold.The use of radioactive isotopes to measure de novo lipogenesis in pigs has been well established. Different from radioactive isotopes, stable isotopes present little or no risk to human and animal subjects. Therefore, the objective of this study was to adapt the method of bolus injection of radioactive glucose (14C) to use 13C-labeled glucose to estimate de novo lipogenesis in finishing pigs. Five vein-catheterized gilts received 3.0 kg/d of a commercial diet for 2 wk. On the last day, the pigs received a bolus injection of [U-13C]glucose (12 mg/kg body weight). A serial of blood samples was taken for 4 h to determine the glucose rate of disappearance (Rd) from plasma glucose isotopic enrichment (IE). The 13C IE of lipids was determined from adipose tissue biopsies collected at 1, 2, and 3 h after the bolus injection and from adipose tissue collected after pig euthanasia 4 h after the bolus. Lipogenesis was estimated from the incorporation of 13C from glucose into adipose tissue lipids. Glucose Rd, estimated using a double-exponential function, averaged 5.4 ± 1.4 mmol/min. The IE of lipids increased linearly during the 4 h following the bolus injection (P less then 0.05). The rate of incorporation of glucose into lipids, estimating lipogenesis, averaged 9.0 µg glucose/(min × g of lipids) 4 h after the bolus injection. In conclusion, the in vivo method using a bolus injection of [U-13C]glucose allows a successful estimation of de novo lipogenesis in finishing pigs.Multiple resources currently exist that predict orthologous relationships between genes. An indication of the reliability of a prediction is provided by the number of resources that support it. Both diets had an STTD Ca to STTD P ratio of 1.901. Growth performance and carcass weights were determined. Metacarpals, metatarsals, femur, tibia, fibula, 3rd and 4th ribs, and 10th and 11th ribs, and all other bones from the left half of the carcass were collected separately. Each bone was defatted and ashed. Pigs fed the diet containing 100% of required Ca and P had greater (P 0.95. In conclusion, metacarpals, metatarsals, and tibia were more representative of total body bone ash compared with other bones.
