We discuss limitations and identify approaches that researchers may use to further evaluate the validity of the K-factor and other applications of LH to human variation.Hsp110s are unique and essential molecular chaperones in the eukaryotic cytosol. They play important roles in maintaining cellular protein homeostasis. Candida albicans is the most prevalent yeast opportunistic pathogen that causes fungal infections in humans. As the only Hsp110 in Candida albicans, Msi3 is essential for the growth and infection of Candida albicans. In this study, we have expressed and purified Msi3 in nucleotide-free state and carried out biochemical analyses. Sse1 is the major Hsp110 in budding yeast S. cerevisiae and the best characterized Hsp110. Msi3 can substitute Sse1 in complementing the temperature-sensitive phenotype of S. cerevisiae carrying a deletion of SSE1 gene although Msi3 shares only 63.4% sequence identity with Sse1. Consistent with this functional similarity, the purified Msi3 protein shares many similar biochemical activities with Sse1 including binding ATP with high affinity, changing conformation upon ATP binding, stimulating the nucleotide-exchange for Hsp70, preventing protein aggregation, and assisting Hsp70 in refolding denatured luciferase. These biochemical characterizations suggested that Msi3 can be used as a model for studying the molecular mechanisms of Hsp110s.This study aimed to investigate the effects of cofD gene knock-out on the synthesis of coenzyme F420 and production of methane in Methanobrevibacter ruminantium (M. ruminantium). The experiment successfully constructed a cofD gene knock-out M. ruminantium via homologous recombination technology. The results showed that the logarithmic phase of mutant M. ruminantium (12 h) was lower than the wild-type (24 h). The maximum biomass and specific growth rate of mutant M. ruminantium were significantly lower (P less then 0.05) than those of wild-type, and the maximum biomass of mutant M. ruminantium was approximately half of the wild-type; meanwhile, the proliferation was reduced. The synthesis amount of coenzyme F420 of M. ruminantium was significantly decreased (P less then 0.05) after the cofD gene knock-out. Moreover, the maximum amount of H2 consumed and CH4 produced by mutant were 14 and 2% of wild-type M. ruminantium respectively. In conclusion, cofD gene knock-out induced the decreased growth rate and reproductive ability of M. ruminantium. Subsequently, the synthesis of coenzyme F420 was decreased. Ultimately, the production capacity of CH4 in M. ruminantium was reduced. Our research provides evidence that cofD gene plays an indispensable role in the regulation of coenzyme F420 synthesis and CH4 production in M. ruminantium.Interspecies intimacy defines an inescapable reality of lab animal research. Such principles determine quotidian practices of welfare and care that, in peculiar ways, privilege animal health (as key to reliable data) while obscuring, erasing, or denying human forms of self care. As such, they presuppose a regulatory ability to formulate, shape, and (re)direct human action. Yet attentiveness to the "messiness of the moral" of lab work exposes other realities indeed, lab personnel regularly engage in a host of subversive responses that test or cross the boundaries of mandated behavior that (re)invigorate the meaning of moral acts of care as interspecies responsibility. The ethnographer's ability to witness, record, and write about these actions within the lab rests comfortably on the relativist principle of suspended judgment. Once one moves outside the lab, however, I ask, wherein lies ethnographic responsibility, when one's accounts of the moral messiness of quotidian lab practices become unbounded and go public? I argue that a dialectical inter- and intraspecies framework-inspired by the existential anthropologist Michael Jackson-offers the ethnographer (and still other scholars) possibilities for forging a productively "unbounded" methodological analytic in and beyond domains of animal science.The mineralogical and geochemical characterization of sediments of the Reconquista River allows analyzing the geochemical partition of trace elements in one of the most polluted water courses of Argentina. The low dissolved oxygen and high ammonia contents, together with the high chemical oxygen demand, attest to the poor water quality. Ammonia, Cd and Cu content in surficial water exceeds the maximum guidelines for freshwater in Argentina. The recent sediments of the uppermost bed are enriched in organic matter (OM), sulfur, Zn, Cu and Pb. The enrichment factor is moderate, and the geoaccumulation index (Igeo) for Cu and Pb indicates uncontaminated to moderately contaminated sediments. The positive and significant correlation between As, Cr, Pb and Zn with the iron content suggests that their retention is controlled by the amount of iron oxy (hydr)oxides in the sediments, probably combined with the silt + clay abundance. In comparison with its tributary, the Las Catonas Stream, the Reconquista River, has less OM and trace elements in the sediments and more dissolved trace elements in the interstitial water. We interpret that OM is the main sorbent of the trace element. In the absence of OM, the iron oxy (hydr)oxides and the silt + clay fraction are a less efficient substitute. Consequently, the interstitial waters of the Reconquista River are enriched in these elements. Therefore, minor changes in the environmental conditions may generate significant release of hazardous trace elements from the sediments to the interstitial water and, in turn, to the surficial water of the river. As most of the big cities and the agricultural activities of Argentina are developed on the loessic substrate, the understanding of its interaction with polluted waters is crucial.
