Onosma bracteata Wall. (Boraginaceae), commonly known as "gaozaban" is a highly valuable medicinal herb, useful in the treatment of body swellings, abdominal pain, eye-related problems, fever, and urinary calculi. The present study was performed to investigate the antioxidant properties of extract/fractions, viz. ethanol (Obeth) extract, hexane (Obhex) fraction, chloroform (Obcl) fraction, ethyl acetate (Obea) fraction, butanol (Obbu) fraction, and aqueous (Obaq) fraction isolated from O. bracteata. Obea fraction showed stronger free radical quenching ability in various antioxidant assays, as compared to the other fractions. Obea fraction with effective free radical-scavenging properties was further evaluated for the antiproliferative activity against human osteosarcoma MG-63, human neuroblastoma IMR-32, and human lung cancer A549 cell lines using MTT assay. Obea fraction showed strong cytotoxicity with GI50 value of 88.56, 101.61, and 112.7 μg/ml towards MG-63, IMR-32, and A549 cells respectively. Mechanistic studies revealed that Obea fraction in osteosarcoma MG-63 cells increased reactive oxygen species (ROS) level and reduced mitochondrial membrane potential. In the presence of Obea, the cells were found to be arrested in the G0/G1 phase in a dose-dependent manner which is also confirmed by the enhancement in the early apoptotic cell population in flow cytometer analysis. Western blotting demonstrated the decrease in expression of p-NFκB, COX-2, p-Akt, and Bcl-xL, whereas upregulation was observed in the expression of GSK-3β, p53, caspase-3, and caspase-9 proteins. RT-qPCR studies revealed downregulation of Bcl-2, cyclin E, CDK2, and mortalin gene expression and upregulation in the expression of p53 genes. The antioxidant and cytotoxic potential of Obea was attributed to the presence of catechin, kaempferol, onosmin A, and epicatechin, as revealed by HPLC analysis. This is the first report regarding the antiproliferative potential of O. bracteata against osteosarcoma.Silver nanoparticles (Ag-NPs) are widely used in daily life and inevitably discharged into the aquatic environment, causing increasingly serious pollution. Research on the toxicity of Ag-NPs is still in infancy, little information is available on the relationships between oxidative stress and antioxidant, as well as damaging degrees of Ag-NPs to cellular structural components of Chlamydomonas reinhardtii (C. reinhardtiii). In the present study, we revealed the toxicity mechanism of C. Sodium orthovanadate in vitro reinhardtii under Ag-NPs stress using flow cytometry (FCM), metabolic methods, and transmission electron microscopy. The results showed that the chloroplasts were damaged and the synthesis of photosynthetic pigments was inhibited under Ag-NPs stress, which inhibited the growth of C. reinhardtii. Meanwhile, Ag-NPs also caused C. reinhardtii to produce excessive reactive oxygen species (ROS), increased malondialdehyde content and changed the permeability of cell membrane, resulting in the acceleration of internalization of Ag-NPs. The decrease of cell size and intracellular chlorophyll autofluorescence was observed with FCM. To deal with the induced excessive ROS that could lead to lethal and irreversible structure damage, C. reinhardtii activated antioxidant enzymes including superoxide dismutase and peroxidase. This study provides new information for better understanding the potential toxicity risks of Ag-NPs in the aquatic environment.Countries throughout the whole world, including China, are working together to curb the greenhouse effect, but the effects seem very limited in spite of the fact that various low-carbon development strategies have been adopted, particularly in industrial enterprises. Therefore, carbon emissions caused by the public should be taken seriously, and the public should be encouraged to engage in behavior that limits carbon emissions. Therefore, this article introduces a new incentive mechanism called the Carbon Generalized System of Preferences (CGSP), which was first introduced in Guangdong Province, China. It is believed that this new mechanism matches the role of leadership in Guangdong in the urbanization and economic development of China by addressing the small sources of greenhouse gases (GHGs) and by issuing carbon coins. Compared with Chinese Certified Emission Reduction (CCER), the development scope, management level, and novel criteria of CGSP are very different but relatively easy for the public to accept. The CGSP shows that the network platform, reduced carbon emissions, and urban pilots are all compatible with the goals of the nation and city, and they promote the CGSP in different ways. Because of its consistency with ecological civilization in China, the prospect of the CGSP is bright; however, there are some challenges, such as policy and economic factors, combined with pollution control.Marine algae have made a strong contribution to global food security in the future. This study is the first report describing the concentration, pathways, and interactions of halogens in 15 species of marine algae collected from the Eastern Harbor in Alexandria, Egypt, relative to 22 key variables. The relationship between halogen content and chemical and biochemical parameters was studied through multivariate analysis. Among all the tested algae, the iodine content was the lowest (2.53-3.00 μg/g). The range of fluoride and chloride in macroalgae (1.12-1.70 and 0.10-0.46 mg/g) was smaller than that of microalgae (0.10-0.46 and 1.48-3.17 mg/g). The bromide content in macroalgae (0.36-5.45 mg/g) was higher than that in microalgae (0.40-0.76 mg/g). The halogen content in macroalgae was arranged in the order of Br > F > Cl > I. In addition, the biochemical parameters such as carbohydrates, proteins, lipids, and certain heavy metals (Fe, Zn, Cu, Mn, Pb, Ni, Co, Cd, and Cr) were determined. Calories, energy, total antioxidant activity (TAC), K/Na, and ion quotient amounts were estimated. The results showed that the green seaweed species had the highest TAC content. In most of the studied algal species, the calculated ion quotient referred to their likelihood of overcoming high blood pressure. The estimated daily intake (EDI) of algae showed no adverse effects on human health. Most of the research variables are below the acceptable WHO/FAO level. Generally, the calorie content of the selected algae is less than 2 kcal, which makes the algae considered an alternative source of healthy food to reduce obesity.Graphical abstract.
