Dual luciferase reporter assays demonstrated that SVCV could not activate the NF-κB pathway. In addition, the PI3K signaling pathway was involved in the expression of the chemokine IL-8 in the SVCV-induced inflammatory response. We also showed that inhibition of the MAPK or PI3K signaling pathway facilitated the expression of SVCV-G as well as increased the SVCV viral titer. Altogether these results reveal the mechanism of the SVCV-mediated inflammatory response. Thus, targeting these signaling pathways may provide novel treatment strategies for SVCV-mediated diseases. Desiccation is a commonly stressful situation experienced by sea cucumber during transportation without/less water. The present study was conducted to investigate the effects of aerial exposure on the survival, oxidative damage, antioxidant capacity, immune-related response and gene expression of Apostichopus japonicus at different low temperatures. After acclimation, sea cucumbers were randomly divided into 3 groups, which were exposed to 5 °C, 10 °C and 15 °C in the closed laboratory condition, respectively. Each group has three parallel replicates. During the experiment, coelomic fluid and respiratory tree of A. japonicus were sampled at the time points of 0, 3, 6, 12, 24 and 48 h post-desiccation for further analysis. The results showed that the survival rates of sea cucumber significantly decreased as time prolonged, and those of 5 °C at 6-48 h of desiccation were significantly higher than 15 °C. Most oxidative damage parameters (e.g., O2- production, MDA, LPO and PC contents) significant increased after stress, but also provide valuable information for sea cucumber culture and transportation. Interferons play an important role in the fish innate immune system against viral infection by inducing the interferon stimulated genes, such as Mx gene. We cloned the MxII gene promoter from orange-spotted grouper and found three MxII gene promoters. All of them contained two interferon stimulated response elements (ISREs), and three dinucleotide repeat sequences located at 5' end of ISREs. Interestingly, there is a polymorphic GT repeat element located upstream of these ISREs. The three MxII gene promoters respectively contained 27, 29, and 31 GT repeats, namely EcMx_27, EcMx_29, and EcMx_31. To determine whether GT repeat element influence the MxII gene expression, the MxII gene promoters were subcloned into promoterless reporter plasmid and transfected into grouper kidney (GK) cells. The results showed that a significant induction by poly(IC) was detected in GK cells transfected with pEcMx_31 (2.65 folds) whereas there was no induction in GK cells transfected with pEcMx_27 and pEcMx_29. However, the significant induction by nervous necrosis virus (NNV) infection was found in GK cells separately transfected with three reporter plasmids. These results suggest that the GT repeat element plays an important role in modulation of MxII gene expression and the induction by poly(IC) and NNV may be mediated through different signal transduction pathways. Chronic spontaneous urticaria (CSU) is considered to be primarily a mast cell-driven disease. However, recent evidence suggests that eosinophils may also have an axial role in symptomology. Histological studies have demonstrated the presence of both eosinophils and eosinophil granules, indicative of activation, in CSU lesions. While many allergic and inflammatory conditions are associated with a peripheral blood eosinophilia, the converse appears to be the case in CSU, a peripheral blood eosinopenia being observed in many patients. Possible mechanisms include the depletion of blood eosinophils by recruitment into the skin during active disease and immunological destruction in the blood. We also address in some detail the interactions between eosinophils and mast cells, particularly the cytokine cross talk of these cells and mediator release possibly leading to clinical symptoms. Finally, treatments aimed at reducing eosinophil accumulation and activation, such as the anti-IL-5 antibodies mepolizumab, reslizumab and benralizumab, have been reported to reduce CSU symptoms. Clearly, a new picture of an important role of eosinophils in the pathogenesis of CSU is emerging. The giant ciliate Stentor coeruleus (S. coeruleus) is a suitable model organism for studying morphogenesis and regeneration at the single-cell level. It contains a prominent structure on the anterior end of the cell, known as the oral apparatus (OA). OA can be induced to shed by urea treatment and then new OA regenerates via a series of defined morphological events and the cell resumes normal feeding activity. We identified OA constituents in S. coeruleus by mass spectrometry. A total of 882 OA-associated proteins were identified; the homologs of 181 of these are known OA constituents in other organisms. The expression pattern of OA-associated genes during regeneration was investigated using single-cell transcriptome sequencing. The expression of most OA-associated genes was high during regeneration, indicating their stable expression after OA shedding. We also identified OA-associated differentially expressed genes that may be involved in regulating OA reconstruction. In summary, this study gives preliminary insight into the molecular basis of OA in S. coeruleus. Circular RNAs (circRNAs) are reported to participate in many diseases including tumorigenesis, which can modulate gene expression post-transcriptionally. Up to now, the detailed function of circRNAs in endometrial carcinoma (EC) progression are to be well established. hsa_circ_0061140 has been revealed to act as crucial regulators in several cancers. This current study was carried out to explore the function of hsa_circ_0061140 in EC development. Here, hsa_circ_0061140 expression was modulated using functional assays and we found loss of hsa_circ_0061140 significantly restrained EC cells progression. Moreover, we revealed hsa_circ_0061140 could act as a molecular sponge for miR-149-5p. STAT3 was predicted as a downstream target gene of miR-149-5p. As well known, miR-149-5p is reported to be involved in tumor growth and metastasis. Then, dual luciferase reporter experiment and RIP assay were carried out to evidence the direct binding association between hsa_circ_0061140 and miR-149-5p. L-glutamate purchase miR-149-5p was able to negatively modulate STAT3 expression.
