Project 2: with the Xi lab
Jiang provides the heart muscle cells
Mainly between PIs
Not a very specific proposal
Main ideas:
RE-differentiation to regenerate heart tissue
Heart regeneration
Epigenetic engineering/modification to de-differentiate (make it back to the progenitor)
DLL4 expression is necessary for heart muscle defects to not occur especially in outflow tract
SHF is responsible for a lot of this--second heart field
First many slides: pictures showing that DLL4 is present in the heart at many age stages
E9.5 and E10.5: lots of DLL4 shown by F2 LacZ which turns the cells blue
Wherever LacZ is present DLL4 is also present
Also shown by immunofluorescence staining
As time goes on to E14.5 then the amount of Dll4 is diminished
Second section: DLL4 knockout leads to heart defects and lethality
Sample defects: Double Outlet Right Ventricle, aortic arch, hypoplastimia or something
The mouse was a Mef2cCree Eph2? DLL4 F/F and when F/F then DLL4 expression was a lot less
In pictures it showed that the area of the right ventricle for F/F was a lot less than the same for WT
