Healthcare-associated Staphylococcus aureus bloodstream infection (HA-SAB) causes preventable harm in hospitalized patients. Currently, there is no standardized method available to review HA-SAB events in order to identify and target preventable risks requiring action at an organizational level.

To develop a tool to classify SAB events, and the necessary response actions, according to the degree of preventability.

Following a literature review, a tool was developed. Consensus feedback and development of the tool was sought from experts (N= 11) in healthcare-associated infection surveillance using a Delphi technique. The completed tool was retrospectively applied to HA-SAB events (N= 43) that occurred at a large healthcare organization.

Survey completion rates were high (91-100%). Clinicians' poor adherence to infection prevention practices and lack of engagement with feedback processes was established as the key modifiable element. The present study elucidates an array of molecular events underlying apoptosis in the gills of polystyrene microplastics (PS-MPs) exposed zebrafish (Danio rerio). 20s Proteasome activity PS-MPs at different concentrations (10 and 100 μg L-1) induced the reactive oxygen species (ROS) generation, in turn affecting the oxidative and immune defense mechanism. The expression profile of antioxidant genes cat, sod1, gpx1a and gstp1 were altered significantly. PS-MPs also significantly inhibited the neurotransmission in zebrafish. In addition, the PS-MPs exposure upregulated the expression of p53, gadd45ba, and casp3b resulting in apoptosis. We demonstrate that PS-MPs significantly upregulate the transcriptional pattern of tnfa and ptgs2a which are essential gene markers in inflammatory mechanism. Further, the oxidative damage induced by PS-MPs exposure could lead to cytological damage resulting in altered lamellar structures, capillary dilation, and necrosis in gill histomaps. In conclusion, the findings of this work strongly suggest that PS-MPs induce dose-and time-dependent ROS mediated apoptotic responses in zebrafish. Furthermore, the physiological responses observed in the gills correlate with the above observations and helps in unravelling the potential molecular mechanism underpinning the PS-MPs toxicity in zebrafish.Haemonchus contortus, a blood-feeding nematode, inhibits blood coagulation at the site of infection to facilitate blood-sucking and digesting for successful parasitism. However, the mechanism underlying anti-coagulation at the host-parasite interface is largely unknown. In the current study, Hc-spi-i8, which has two greatly different transcripts named Hc-spi-i8a and Hc-spi-i8b, respectively, was described. Hc-SPI-I8A was a serine protease inhibitor containing a trypsin inhibitor-like cysteine rich (TIL) domain, while Hc-SPI-I8B was not. Hc-SPI-I8A/B were primarily expressed in the hypodermis, intestines and gonads in the parasitic stages of H. contortus. Hc-SPI-I8A interacted with Ovis aries TSP1-containing protein (OaTSP1CP), which was determined by yeast two-hybrid, co-immunoprecipitation (Co-IP), pull down and co-localization experiments. The blood clotting time contributed by the TIL domain was prolonged by Hc-SPI-I8A. Hc-SPI-I8A is most likely interfering in the extrinsic coagulation cascade by interacting with OaTSP1CP through its TIL domain and intrinsic coagulation cascade by an unknown mechanism. These findings depict a crucial point in the host-parasite interaction during H. contortus colonization, which should contribute to drug discovery and vaccine development in fighting against this important parasite worldwide.Forkhead box O (FoxO) protein, a major downstream transcription factor of insulin/insulin-like growth factor signaling/target of rapamycin pathway (IIS/TOR), is involved in the regulation of larval growth and the determination of organ size. FoxO also interacts with 20-hydroxyecdysone (20E) and juvenile hormone (JH) signal transduction pathways, and hence is critical for larval development in holometabolans. However, whether FoxO plays a critical role during larval metamorphosis needs to be further determined in Leptinotarsa decemlineata. We found that 20E stimulated the expression of LdFoxO. RNA interference (RNAi)-aided knockdown of LdFoxO at the third-instar stage repressed 20E signaling and reduced larval weight. Although the resultant larvae survived through the third-fourth instar ecdysis, around 70% of the LdFoxO depleted moribund beetles developmentally arrested at prepupae stage. These LdFoxO depleted beetles were completely wrapped in the larval exuviae, gradually darkened and finally died. Moreover, approximately 12% of the LdFoxO RNAi beetles died as pharate adults.
