In zebrafish lacking Foxc1 paralogs, we find a global decrease in chromatin accessibility in chondrocytes, consistent with a later loss of dorsal facial cartilages. Zebrafish transgenesis assays confirm that many of these Foxc1-dependent elements function as enhancers with region- and stage-specific activity in facial cartilages. These results show that Foxc1 promotes chondrogenesis in the face by establishing chromatin accessibility at a number of cartilage-associated gene enhancers.Learning and decision-making are interactive processes, yet cognitive modeling of error-driven learning and decision-making have largely evolved separately. Recently, evidence accumulation models (EAMs) of decision-making and reinforcement learning (RL) models of error-driven learning have been combined into joint RL-EAMs that can in principle address these interactions. However, we show that the most commonly used combination, based on the diffusion decision model (DDM) for binary choice, consistently fails to capture crucial aspects of response times observed during reinforcement learning. We propose a new RL-EAM based on an advantage racing diffusion (ARD) framework for choices among two or more options that not only addresses this problem but captures stimulus difficulty, speed-accuracy trade-off, and stimulus-response-mapping reversal effects. The RL-ARD avoids fundamental limitations imposed by the DDM on addressing effects of absolute values of choices, as well as extensions beyond binary choice, and provides a computationally tractable basis for wider applications.Cooperation, paying a cost to benefit others, is widespread. Cooperation can be promoted by pleiotropic 'win-win' mutations which directly benefit self (self-serving) and partner (partner-serving). Previously, we showed that partner-serving should be defined as increased benefit supply rate per intake benefit. Here, we report that win-win mutations can rapidly evolve even under conditions unfavorable for cooperation. Specifically, in a well-mixed environment we evolved engineered yeast cooperative communities where two strains exchanged costly metabolites, lysine and hypoxanthine. Among cells that consumed lysine and released hypoxanthine, ecm21 mutations repeatedly arose. ecm21 is self-serving, improving self's growth rate in limiting lysine. ecm21 is also partner-serving, increasing hypoxanthine release rate per lysine consumption and the steady state growth rate of partner and of community. ecm21 also arose in monocultures evolving in lysine-limited chemostats. Thus, even without any history of cooperation or pressure to maintain cooperation, pleiotropic win-win mutations may readily evolve to promote cooperation.Lymphoblastoid cell lines (LCLs) are generated by transforming primary B cells with Epstein-Barr virus (EBV) and are used extensively as model systems in viral oncology, immunology, and human genetics research. In this study, we characterized single-cell transcriptomic profiles of five LCLs and present a simple discrete-time simulation to explore the influence of stochasticity on LCL clonal evolution. Single-cell RNA sequencing (scRNA-seq) revealed substantial phenotypic heterogeneity within and across LCLs with respect to immunoglobulin isotype; virus-modulated host pathways involved in survival, activation, and differentiation; viral replication state; and oxidative stress. A review of current research focused on these analgesic adjuncts and the drawbacks of opioid use reveals increased understanding and complexity of the physiology of pain, its treatment, and our understanding of pharmacologic mechanisms of action of analgesics. This course will examine the current limitations of opioids, provide a brief overview of the physiology of pain, and discuss the pharmacologic mechanism of action of dexmedetomidine and patient populations in which its use might be advantageous.The ongoing coronavirus disease 2019 (COVID-19) pandemic has created many changes and difficulties in healthcare, and the anesthesia specialty is no exception. Policies and protocols have been adapted worldwide to help neutralize infection risk and exposure. Transmission of the virus to healthcare workers has been a major concern, and the risk of infection is exceptionally high for Certified Registered Nurse Anesthetists (CRNAs) because of their close contact with infected patients. CRNAs are in a unique position to help decrease exposure for themselves and other members of the healthcare team by taking extra precautions during airway manipulation. A great deal of focus has been placed on reducing risks during intubation, but reports describing methods of reducing contamination and exposure to respiratory droplets during emergence and extubation are scarce.
