Our results show that the presence of a single gene can affect the dispersal of a bacterial pathogen and significantly alter its population dynamics.A virtual screening approach based upon a combination of docking and pharmacophore methods was utilized on a library of 1.4 million molecules to identify novel antimicrobial agents, which may potentially act via inhibition of the caseinolytic protease. Using this method, compound 6 was found to be bactericidal against three staphylococcal species (minimum inhibitory concentration (MIC)=4-16 μg/mL). Further, subsequent structural optimization of 6 led to the identification of compound 24, which was shown to be the most potent analog within the series (MIC=4 μg/mL) and outperforming the antibiotic controls for two of the staphylococcal species. The newly discovered antimicrobial agent (24) demonstrated excellent in silico ADME properties and was non-toxic when tested on two human skin cell lines. As such, compound 24 has the potential for use as a lead molecule in the development of a novel class of antimicrobial agents.The histone deacetylases (HDACs) family of enzymes possess deacylase activity for histone and nonhistone proteins; HDAC11 is the latest discovered HDAC and the only member of class IV. Here, we summarize the importance of HDAC11 in a vast array of cellular pathways, which has been recently highlighted by discoveries about its subcellular localization, biochemical features, and its regulation by microRNAs and long noncoding RNAs, as well as its new targets and interactors. Additionally, we discuss the recent work showing the consequences of HDAC11 dysregulation in brain, skeletal muscle, and adipose tissue, and during regeneration in response to kidney, skeletal muscle, and vascular injuries, underscoring HDAC11 as an emerging hub protein with physiological functions that are much more extensive than previously thought, and with important implications in human diseases.
To describe the effect of alcohol policy on the incidence of intensive care unit (ICU) admissions associated with hazardous and harmful alcohol use in the Northern Territory (NT) of Australia DESIGN, SETTING AND PARTICIPANTS Before and after analysis of admissions to NT ICUs between April 2018 and September 2019, extending on both a descriptive study describing hazardous and harmful alcohol use and single-centre analyses of harm minimization policies. This systematic review reports the prevalence of Mycobacterium tuberculosis complex (MTBC) organisms in slaughtered cattle showing tuberculosis-like lesion (TBL) with available literature worldwide. Appropriate keywords were used to search various databases to collect articles pertaining to slaughterhouse studies. Bovine TB prevalence, based on the prevalence of MTBC organisms in slaughtered cattle showing TBL by culture, microscopy, PCR and spoligotyping, was assessed in each study using a random-effects model and standardized mean with 95% confidence interval (CI). Heterogeneity was assessed by the I2 statistic. Publication bias was evaluated using funnel plots. Out of 72 hits, 37 studies were selected based on title and abstract. Ten articles were excluded due to lack of desired data, and 27 studies were included in the final analysis. From the selected articles, it was found that 426 [95% CI 302-560] per 1,000 slaughtered cattle with TBL were positive for the presence of MTBC organisms. The sensitivity analysis showed that no individual study alone influenced the estimation of pooled prevalence. The prevalence of MTBC organisms in slaughtered cattle showing TBL by culture, microscopy, PCR and spoligotyping was 474[95% CI 342-610], 385 [95% CI 269-515], 218 [95% CI 132-338], 326 [95% CI 229-442], respectively, per 1,000 slaughtered cattle. Most of the slaughtered cattle were from the same locality as the slaughterhouse. The results obtained in this study suggest that abattoir monitoring can give an estimate of the prevalence of bTB in that locality. This study also emphasizes the need to test cattle and animal handlers who were in contact with bTB-positive cattle.
Focal cortical dysplasia IIIa (FCD IIIa) is a common histopathological finding in temporal lobe epilepsy. However, subtle alterations in the temporal neocortex of FCD IIIa renders presurgical diagnosis and definition of the resective range challenging.
