BCR-ABL translocation is a key hallmark of chronic myeloid leukemia (CML). The chemistry involves transcription of a novel 8.5 kb mRNA with a b3a2 and/ or b2a2 junction. Though there is an improvement in survival using the TKIs, there are causes for the treatment failures. Thus, the study focuses on the causes underlying the failed response. This study comprises BCR-ABL expression in correlation with age, gender and transcript type and disease monitoring in follow-up samples. Eighty-seven chronic phase CML patients were enrolled in the study. Out of these 24 patients were followed further. Quantitative Real time PCR was performed to assess the BCR-ABL expression followed by gel electrophoresis of PCR products. The results were expressed as CN/µg RNA. The results obtained were correlated with SPSS 16.0 software. We found three different types of the expression that includes b2a2, b3a2 and co-expression (b2a2 + b3a2). Higher incidence of b2a2 with a relatively equal incidence of co-expression was observed. The BCR-ABL expression was higher in males, in young patients and in those exhibiting co-expression of the transcripts. Greater relapse was seen in females and in older patients. The patients with co-expression of transcripts exhibited the highest expression; however these patients showed the best treatment response suggest the co-expression is the favourable parameter for CML patients. The transcript type and BCR-ABL expression are well correlated and hence can be considered as a prognostic as well as the predictive indicator considering the BCR-ABL expression for CML patients. © Association of Clinical Biochemists of India 2018.Workers involved in battery manufacturing or recycling factories are occupationally exposed to high concentrations of lead. In humans, lead can cause a wide range of biological effects depending upon the level and duration of exposure. The purpose of this study was to find out the blood lead levels (BLL) in occupationally exposed workers involved in battery industry in Delhi NCR region and to study whether lead affected the vitamin D (vit D) and calcium metabolism. For this study 100 occupationally lead-exposed battery workers (LEBW) and 100 non-lead exposed controls (NLEC) were recruited. BLL were measured using inductively coupled plasma optical emission spectrometer (ICP-OES) technique while ELISA was performed to quantify the serum vit D levels in the study subjects. Routine biochemical parameters were measured by chemistry autoanalyzers. Statistical analysis was done using appropriate statistical tools. Results showed that BLL were significantly higher in LEBW as compared to NLEC (p less then 0.0001). Serum vitamin D, calcium and phosphorus levels were significantly decreased in battery workers as compared to controls (p less then 0.005). Spearman's rank correlation analysis showed significant negative correlation of BLL with serum Vitamin D and calcium levels. Significant positive correlation was observed between BLL and duration of lead exposure. Weak negative correlation was also observed between BLL and vit D even after adjusting for smoking status. In conclusion, this study demonstrated that higher BLL significantly alters the vit D and calcium metabolism. © Association of Clinical Biochemists of India 2018.Streblus asper Lour., traditionally used for anti-diarrheal effects as like dysentery and diarrhea. The present study aims to prove the anti-diarrheal activities of methanolic extract of leaves of S. asper in animal models. The anti-diarrheal activity was evaluated using castor oil-induced diarrhea and magnesium sulphate-induced diarrhea models whereas anti-motility activities were investigated using gastrointestinal transit test examined in animal models. In castor oil-induced diarrhea model, methanolic extract of S. asper (MESA) at the doses of 100, 200, and 400 mg/kg produced statistically significant (P less then 0.001) decreased the number of diarrheal feces of rats against castor oil-induced diarrhea as well as magnesium sulphate-induced diarrhea model also showed the same manner. In gastrointestinal transit test, delayed gastric emptying time decreased significantly (P less then 0.001), the propulsion of charcoal meal in the gastrointestinal tract which also showed a dose-dependent manner in rats. The recent study indicates that MESA possesses anti-diarrheal property. The findings represent a rational explanation for its use in traditional medicine for the management of diarrhea management. © Association of Clinical Biochemists of India 2018.Catechol-O-methyletransferase (COMT) enzyme is involved in the inactivation of catecholamine and catechol estrogens. Catechol estrogens have carcinogenic potential and DNA damaging ability. Several studies investigated COMT Val158Met polymorphism as risk factor for endometrial cancer but the results were inconclusive. Hence the objective of present study was to find out exact association between COMT gene Val158Met polymorphism and endometrial cancer by a meta-analysis. Pubmed, Google Scholar, Springer Link and Science Direct databases were searched for case-control articles which investigated COMT Val158Met polymorphism in endometrial cancer cases. All statistical analysis was performed using MetaAnalyst and Mix programs. The results of meta-analysis suggested that there were no association between COMT Val158Met polymorphism and endometrial cancer risk (allele contrast model-ORA vs. G = 0.97, 95% CI = 0.86-1.10, p = 0.67; co-dominant model-ORAG vs. GG = 0.91, 95% CI = 0.77-1.06, p = 0.23; homozygote model-ORAA vs. GG = 1.01, 95% CI = 0.84-1.19, p = 0.29; dominant model-ORAA+AG vs. GG PD184352 nmr , 95% CI = 0.77-1.11, p = 0.43; recessive model-ORAA vs. AG+GG = 1.02, 95% CI = 0.89-1.20, p = 0.62). #link# Publication bias was absent. Subgroup analysis based on source of controls was also performed. In conclusion, results of present meta-analysis showed no association between COMT Val158Met polymorphism and susceptibility to endometrial cancer. © Association of Clinical Biochemists of India 2018.
