Enzyme cascade reactions can greatly improve catalytic efficiency and achieve selective and sensitive detection of targets. This paper presents a novel strategy for colorimetric sensing of targets by loading natural enzymes into a hemin-doped G-quadruplex (G4-hemin) biohydrogel to form a flexible enzyme cascade sensing (FECS) platform. The biohydrogel has advantages of biocompatibility, printability and flexibility. The biohydrogel not only participates in the cascade reaction as the mimic enzyme but also provides a mild microenvironment for the natural enzyme. The FECS platform has a linear range from 0.4 μM to 120 μM and a detection limit of 3.6 × 10-6 M for hydrogen peroxide detection. Additionally, the FECS platform has a low detection limit and wide linear range for glucose and xanthine by loading xanthine oxidase (XOD) and glucose oxidase (GOD) into the biohydrogel, respectively. These results indicate that the novel FECS platform is an effective detection platform that can detect multiple targets and is expected to be widely used in flexible sensing.On-line solid-phase extraction capillary electrophoresis-mass spectrometry (SPE-CE-MS) is a powerful technique for high throughput sample clean-up and analyte preconcentration, separation, detection, and characterization. see more The most typical design due to its simplicity and low cost is unidirectional SPE-CE-MS. However, in this configuration, the sample volumes introduced by pressure depend on the dimensions of the separation capillary and some matrix components could be irreversibly adsorbed in its inner walls. Furthermore, in many cases, the requirements of on-line preconcentration are incompatible with the background electrolyte necessary for an efficient separation and sensitive MS detection. Here, we present SPE-CE-MS with a nanoliter valve (nvSPE-CE-MS) to overcome these drawbacks while keeping the design simple. The nvSPE-CE-MS system is operated with a single CE instrument and two capillaries for independent and orthogonal SPE preconcentration and CE separation, which are interfaced through an external and electrically isolated valve with a 20 nL sample loop. The instrumental setup is proved for the analysis of opioid and amyloid beta peptide biomarkers in standards and plasma samples. NvSPE-CE-MS allowed decreasing the limits of detection (LODs) 200 times with regard to CE-MS. Compared to unidirectional SPE-CE-MS, peak efficiencies were better and repeatabilities similar, but total analysis times longer and LODs for standards slightly higher due to the heart-cut operation and the limited volume of the valve loop. This small difference on the LODs for standards was compensated for plasma samples by the improved tolerance of nvSPE-CE-MS to complex sample matrices. In view of these results, the presented setup can be regarded as a promising versatile alternative to avoid complicated matrix samples entering the separation capillary in SPE-CE-MS.C. D. In this paper, I critically evaluate Norton's dissolution. In particular, I argue that the problem of induction is an epistemological problem, that Norton's material theory entails an externalist epistemology, and that it is a common feature of such epistemologies that they dissolve the problem of induction. The upshot is that the material theory is not unique in its ability to reap the specifically epistemic benefits of dissolving the problem of induction, and thus that the epistemic advantages of the material theory over extant alternatives in this regard are fewer than it may appear at first sight.Examining tensions between the past and present uses of scientific concepts can help clarify their contributions as tools in experimental practices. This point can be illustrated by considering the concepts of mental imagery and hallucinations despite debates over their respective referential reliabilities remaining unresolved within their interdependent histories, both are used as independently stable concepts in neuroimaging experiments. Building on an account of how these concepts function as tools structured for pursuit of diverging goals in experiments, this paper explores this tension by re-examining the continued reliance of each concept on inverse characterisations inherited from the nominally-discarded 'mediator-view' of sensory-like mental phenomena (SLMP). In doing so, I seek to demonstrate how examining unresolved tensions can help highlight that entrenched associations can remain both integral to, and obscured by, the uses of concepts as goal-directed tools within experimental practices.A pessimistic strain of thought is fomenting in the health studies literature regarding the status of medicine. Ioannidis's (2005) now famous finding that "most published research findings are false" and Stegenga's (2018) book-length argument for medical nihilism are examples of this. In this paper, we argue that these positions are incorrect insofar as they rest on an untenable account of the nature of facts. Proper attention to fallibilism and the social organization of knowledge, as well as Bayesian probabilities in medical reasoning, prompt us to ask why the cynics expect the results of quantitative studies to be incontrovertibly true in the first place. While we agree with Ioannidis and others' identified flaws in the medical research enterprise, and encourage rectification, we conclude that medical nihilism is not the natural outcome of the current state of research.The fragmentation of academic disciplines forces individuals to specialise. In doing so, they become experts over their narrow area of research. However, ambitious scientific projects, such as the search for gravitational waves, require them to come together and collaborate across disciplinary borders. How should scientists with expertise in different disciplines treat each others' expert claims? An intuitive answer is that the collaboration should defer to the opinions of experts. In this paper we show that under certain seemingly innocuous assumptions, this intuitive answer gives rise to an impossibility result when it comes to aggregating the beliefs of experts to deliver the beliefs of a collaboration as a whole. We then argue that when experts' beliefs come into conflict, they should waive their expert status.
