The compound 2,4-diacetylphloroglucinol (DAPG) is a well-known secondary metabolite produced by Pseudomonas spp. that are used as biocontrol agents. DAPG displays a remarkably broad spectrum of toxic activity against pathogens of plants. Yet high concentrations of DAPG may also have negative effect on plants, but the phytotoxicity of DAPG is not clearly understood. Here we used genome-wide activation tagging Arabidopsis plants as the model plant to investigate the plant response to DAPG. A total of 15 lines were selected as DAPG-tolerant plants from among 62000 lines investigated. The DAPG-responsible genes were then identified via TAIL-PCR and qRT-PCR, and the gene ontology analysis showed that the distribution of these genes having different biological processes, cellular regulations, and molecular functional properties. Collectively, these findings suggest that plants may rely on several pathways to prevent DAPG phytotoxicity.Powdery mildews are obligate biotrophic fungal pathogens causing important diseases of plants worldwide. Very little is known about the requirements for their pathogenicity at the molecular level. This is largely due to the inability to culture these organisms in vitro or to modify them genetically. Here we describe a mutagenesis procedure based on ultraviolet (UV) irradiation to accumulate mutations in the haploid genome of the barley powdery mildew pathogen, Blumeria graminis f.sp. hordei (Bgh). Exposure of Bgh conidia to different durations of UV-C radiation (10 s-12 min) resulted in a reduced number of macroscopically visible fungal colonies. Bgh colony number was negatively correlated with exposure time and the total number of consecutive cycles of UV irradiation. Dark incubation following UV exposure further reduced fungal viability, implying that photoreactivation is an important component of DNA repair in Bgh. After several rounds of UV mutagenesis, we selected two mutant isolates in addition to the parental Bgh K1 isolate for whole-genome resequencing. By combined automated prediction of sequence variants and their manual validation, we identified unique UV-induced mutations in the genomes of the two isolates. Most of these mutations were in the up- or downstream regions of genes or in the intergenic space. Some of the variants detected in genes lead to predicted missense mutations. As an additional insight, our bioinformatic analyses revealed a complex population structure within supposedly clonal Bgh isolates.This study investigates the role of sexual sadism in the crime-commission process of sexual homicide (SH) involving child victims. A comparison between sadistic and nonsadistic cases involving child victims is conducted by examining the crime context, crime characteristics, methods of killing, body recovery characteristics, and forensic awareness strategies used by offenders. The sample comes from the Sexual Homicide International Database (SHIelD) including 135 cases of solved SHs involving child victims-35 cases with sexual sadism and 101 cases without sexual sadism. The Sexual Homicide Crime Scene Rating Scale for Sexual Sadism (SADSEX-SH) scale is used to identify sexual sadism from crime scene actions. Bivariate and multivariate analysis are performed to examine the differences between the two groups. Findings indicate that sadistic SH of children are characterized by an important level of structured premeditation, the commission of more diversified sexual acts, the use of specific method of killing, and the partial use of forensic awareness strategies. Practical implications in terms of criminal investigations are discussed.Purpose/aim of the study Subacute sclerosing panencephalitis (SSPE) is a degenerative disease of the brain caused by a persistent measles virus infection occurring mostly in childhood or early adolescence. The spectrum of epileptic phenomena associated with SSPE is wide, varying from partial or generalized tonic-clonic seizures and atypical absences to myoclonic-atonic attacks. Tonic seizures are very rare in SSPE.Materials and methods Herein, we discuss a case of 25 years old male that presented with adult-onset SSPE with tonic seizures accompanying myoclonic seizures.Results Patient was treated with clonazepam 5 mg/day and an isoprinosine regimen with a dose of 70 mg/kg/day. This is the fourth case of SSPE presenting with myoclonic and tonic seizures and the first case of SSPE with myoclonic and tonic seizures reported in an adult-onset case in the English literature.Conclusions Adult-onset SSPE with tonic seizures is rare and may be confusing, thus, it is important to recognize the presence of this type of tonic motor seizures in SSPE patients.In this paper, a propensity score-integrated composite likelihood (PSCL) approach is developed for cases in which the control arm of a two-arm randomized controlled trial (RCT) (treated vs control) is augmented with patients from real-world data (RWD) containing both clinical outcomes and covariates at the patient-level. RWD patients who were treated with the same therapy as the control arm of the RCT are considered for the augmentation. The PSCL approach first estimates the propensity score for every patient as the probability of the patient being in the RCT rather than the RWD, and then stratifies all patients into strata based on the estimated propensity scores. selleck chemical Within each propensity score stratum, a composite likelihood function is specified and utilized to down-weight the information contributed by the RWD source. A total of 63 subjects (age 60.8 ± 13.5 years, 75% male, 80% Type 2 diabetes, HbA1C 8.9%±2.2%) were included. The proportion of Gram-positive (DWS 49% v SBS 52%) and Gram-negative (DWS 60% v SBS 60%) isolates was similar between the techniques.
