Modulation of tyrosinase activity is expected to alter the body pigmentation in fish and other vertebrate species. In the present study, physicochemical, functional, and structural properties of tyrosinase of three fish species viz., goldfish, Japanese medaka, and common carp were determined. The homology model was developed using the Chimera1.1.2, Swiss model, and Phyre2, and the best model was selected upon evaluation. Further, a virtual screening method was applied to identify the putative modulators using the PyRx- Virtual screening tool. The estimated physicochemical and functional properties of tyrosinase from the three species suggested that they all are hydrophobic, acidic, thermostable, with a high extinction coefficient (Cys, Trp, and Tyr) and have transmembrane-segment. Based on virtual screening against 13,000 compounds from the zinc database, five compounds were determined as potent modulators of fish tyrosinase with a binding energy of -7.0 to -8.8 Kcal/mol. Of these, Pilosine (ZINC13469966) was found to be the best putative modulator with low binding energy and properties of standardized drugs. This study showed that the tyrosinase function could be modulated to alter the pigment formation in fish species by using small compound.We investigated whether an 11-day heat acclimation programme (HA) enhanced endurance performance in a temperate environment, and the mechanisms underpinning any ergogenic effect. Twenty-four males (V̇O2max 56.7 ± 7.5 mL·kg-1·min-1) completed either (i) HA consisting of 11 consecutive daily exercise sessions (60-90 min·day-1; n = 16) in a hot environment (40°C, 50% RH) or; (ii) duration and exertion matched exercise in cool conditions (CON; n = 8 [11°C, 60% RH]). Before and after each programme power at lactate threshold, mechanical efficiency, VO2max, peak power output (PPO) and work done during a 30-minute cycle trial (T30) were determined under temperate conditions (22°C, 50% RH). HA reduced resting (-0.34 ± 0.30°C) and exercising (-0.43 ± 0.30°C) rectal temperature, and increased whole-body sweating (+0.37 ± 0.31 L·hr-1) (all P≤0.001), with no change in CON. Plasma volume increased in HA (10.1 ± 7.2%, P  less then  0.001) and CON (7.2 ± 6.3%, P = 0.015) with no between-groups difference, whereas exercise heart rate reduced in both groups, but to a greater extent in HA (-20 ± 11 b·min-1) than CON (-6 ± 4 b·min-1). VO2max, lactate threshold and mechanical efficiency were unaffected by HA. PPO increased in both groups (+14 ± 18W), but this was not related to alterations in any of the performance or thermal variables, and T30 performance was unchanged in either group (HA Pre = 417 ± 90 vs. Post = 427 ± 83 kJ; CON Pre = 418 ± 63 vs. Z-LEHD-FMK ic50 Post = 423 ± 56 kJ). In conclusion, 11-days HA induces thermophysiological adaptations, but does not alter the key determinants of endurance performance. We tested two training modalities, ascending or descending stair walking, representing concentric (CON) and eccentric (ECC) exercise, respectively. We also tested the effects of additional weight during eccentric exercise (ECC+). Thirty-two healthy men and women (70±3 [mean ± SE] yrs.) were randomly assigned to CON, ECC, or ECC+ (carrying +15 % of body weight in a vest) in a 3 (n=32) or 6 (n=21) week intervention (3 sessions/week). Data was analysed by mixed models approach. Rate of perceived exertion (RPE; Borg scale 6-20; mean values from 3 and 6weeks) during training did not differ between CON (12.3±0.4), ECC (11.5±0.3), and ECC+ (11.7±0.4). After 6weeks, leg muscle mass increased more in ECC+ (+0.29±0.09 kg) vs CON (+0.08±0.05 kg) (P less then 0.05) but not different from ECC (+0.16±0.06 kg). 6-minute walk test (6MWT) increased after 6 weeks more (P less then 0.05) in ECC+ (+85±23 m) compared with ECC (+37±13 m) and CON (+27±12 m). Intramyocellular glycogen content increased from 359±19 nmol/mg d.w. in CON (to 511±65 and 471±44 after 3 and 6 wks, respectfully (P less then 0.05)), but not in ECC (to 344±28 after 6 weeks) or in ECC+ (to 389±20 after 6 weeks). Conclusion carrying extra weight while descending stair walking do not increase RPE, but the ECC+ training resulted in greater muscle responses compared with CON, but glycogen synthesis was stimulated only in CON. Descending stairs is a simple model for prevention and treatment of sarcopenia and the stimulus is enhanced by carrying extra weights.Aim Evaluation of the biocompatibility and radiosensitizer potential of citrate-coated cobalt (cit-CF) and nickel (cit-NF) ferrite nanoparticles (NPs). Materials & methods Normal fibroblast and breast cancer cells were treated with different concentrations of citrate-coated ferrite NPs (cit-NPs) and irradiated with a cobalt-60 source at doses of 1 and 3 Gy. After 24 h, cell metabolism, morphology alterations and nanoparticle uptake were evaluated. Results Cit-CF and cit-NF NPs showed no toxicity to normal cells up to 250 and 100 μg.ml-1, respectively. Combination of cit-NP and ionizing radiation resulted in up to fivefold increase in the radiation therapeutic efficacy against breast cancer cells. Conclusion Cit-CF and cit-NF NPs are suitable candidates for application as breast cancer cell radiosensitizers.
