The biological significance of axon myelination is now elucidated as to provide protonic insulation and prevent any ions from interfering with action potential signal.To investigate the effect of bone marrow mesenchymal stem cells (MSCs) on ovarian and testicular function of aging Sprague-Dawley (SD) rats induced by D-galactose (D-gal) and try to clarify the underlying functional mechanism. Adherent culture was used to isolate and purify rat MSCs. The status, proliferation and differentiation of MSCs were detected by hematoxylin-eosin staining, MTT, colony formation, flow cytometry and directional differentiation. The aging rat model was established by subcutaneous injection of D-gal, and the homing of MSCs was detected by fluorescence microscope after infusion of GFP-labeled MSCs through caudal vein. ELISA was used to detect the content of sex hormone in serum, and HE staining was used to observe the structure and morphology of testis and ovary. The isolated and purified MSCs were in good condition, and most of the cells were in G1 phase, which had strong abilities of cell proliferation, colony formation and differentiation. After GFP-labeled MSCs were infused, MSCs could be homed into the testis and ovary of rats. MSCs infusion could significantly improve the morphology of testis and ovary, increase the contents of P and E2 while decrease the contents of LH and FSH in female rats, and increase the content of testosterone in male rats (P less then 0.01). It also increased the activity of superoxide dismutase (SOD) in serum of ovary and testis and significantly decreased the content of malondialdehyde (MDA). MSCs affected the content of MDA and the activity of SOD by reducing the expression of cyclin-dependent kinase inhibitor 2A (p16) and increasing proliferating cell nuclear antigen (PCNA), consequently improving the aging and injury of reproductive organs.The coronavirus disease 2019 (COVID-19) pandemic continues to devastate US nursing homes. Adequate personal protective equipment (PPE) and staffing levels are critical to protect nursing home residents and staff. Despite the importance of these basic measures, few national data are available concerning the state of nursing homes with respect to these resources. This article presents results from a new national database containing data from 98 percent of US nursing homes. We find that more than one in five nursing homes reports a severe shortage of PPE and any shortage of staff. Rates of both staff and PPE shortages did not meaningfully improve from May to July 2020. Facilities with COVID-19 cases among residents and staff, as well as those serving more Medicaid recipients and those with lower quality scores, were more likely to report shortages. Policies aimed at providing resources to obtain additional direct care staff and PPE for these vulnerable nursing homes, particularly in areas with rising community COVID-19 case rates, are needed to reduce the national COVID-19 death toll.TRIM71 is an important RNA-binding protein in development and disease, yet its direct targets have not been investigated globally. Here we describe a number of disease and developmentally-relevant TRIM71 RNA targets such as the MBNL family, LIN28B, MDM2, and TCF7L2. We describe a new role for TRIM71 as capable of positive or negative RNA regulation depending on the RNA target. We found that TRIM71 co-precipitated with IMP1 which could explain its multiple mechanisms of RNA regulation, as IMP1 is typically thought to stabilize RNAs. Deletion of the NHL domain of TRIM71 impacted its ability to bind to RNA and RNAs bound by congenital hydrocephalus-associated point mutations in the RNA-binding NHL domain of TRIM71 clustered closely with RNAs bound by the NHL deletion mutant. Our work expands the possible mechanisms by which TRIM71 may regulate RNAs and elucidates further potential RNA targets.
