Pep19-4LF is a synthetic antimicrobial peptide (GKKYRRFRWKFKGKLFLFG) that was previously designed with the main focus on high antimicrobial activity. The hydrophobic motif, LFLFG, was found to be essential for antimicrobial activity. However, this motif shows several limitations such as aggregation in biological media, low solubility, and small yields in peptide synthesis. In order to obtain more appropriate peptide characteristics, the hydrophobic motif was replaced with fatty acids. For this purpose, a shortened variant of Pep19-4LF (Pep19-short; GKKYRRFRWKFKGK) was synthesized and covalently linked to saturated fatty acids of different chain lengths. The peptide conjugates were tested with respect to their antibacterial activity by microdilution experiments on different bacterial strains. The length of the fatty acid was found to be directly correlated to the antimicrobial activity up to an ideal chain length (undecanoic acid, C110). This conjugate showed high antimicrobial activity in absence of toxicity. Time-kill studies revealed a fast and bactericidal mode of action. Furthermore, the first in vivo experiments of the conjugate in rodents demonstrated pharmacokinetics appropriate for application as a drug. These results clearly indicate that the hydrophobic motif of the peptide can be replaced by a single fatty acid of medium length, simplifying the design of this antimicrobial peptide while retaining high antimicrobial activity in the absence of toxicity.Chitosans represent a group of multifunctional drug excipients. Here, we aimed to estimate the impact of high-molecular weight chitosan on the physicochemical properties of clotrimazole-chitosan solid mixtures (CL-CH), prepared by grinding and kneading methods. We characterised these formulas by infrared spectroscopy, differential scanning calorimetry, and powder X-ray diffractometry, and performed in vitro clotrimazole dissolution tests. Additionally, we examined the antifungal activity of clotrimazole-chitosan mixtures against clinical Candida isolates under neutral and acid conditions. The synergistic effect of clotrimazole and chitosan S combinations was observed in tests carried out at pH 4 on Candida glabrata strains. The inhibition of C. glabrata growth reached at least 90%, regardless of the drug/excipient weight ratio, and even at half of the minimal inhibitory concentrations of clotrimazole. Our results demonstrate that clotrimazole and high-molecular weight chitosan could be an effective combination in a topical antifungal formulation, as chitosan acts synergistically with clotrimazole against non-albicans candida strains.Fall armyworm is one of the main pests of conventional and Bacillus thuringiensis (Bt) corn in many countries in the Americas, Africa, Asia and in Australia. We conducted diet-overlay bioassays to determine the status of susceptibility to four Bt proteins (Cry1A.105, Cry2Ab2, Cry1F and Cry1Ac) in three different populations of fall armyworm from Mexico, and one population from Puerto Rico. Bioassays showed that fall armyworms from Puerto Rico were resistant to Cry1F with a resistance ratio 50 (RR50) higher than 10,000 ng/cm2 and to Cry1Ac with a RR50 = 12.2 ng/cm2, displaying the highest median lethal concentration (LC50) values to all Bt proteins tested. The effective concentration 50 (EC50) values further confirmed the loss of susceptibility to Cry1F and Cry1Ac in this population. However, LC50 and EC50 results with Cry1A.105 and Cry2Ab2 revealed that fall armyworm from Puerto Rico remained largely susceptible to these two proteins. The Mexican populations were highly susceptible to all the Bt proteins tested and displayed the lowest LC50 and EC50 values to all Bt proteins. Our results suggest that Cry1F and Cry1Ac resistance is stable in fall armyworm from Puerto Rico. AZ 628 in vivo However, this population remains susceptible to Cry1A.105 and Cry2Ab2. Process parameters for FDM are divided into controllable or uncontrollable parameters. The most critical ones are built orientation, layer thickness, infill pattern, infill density, and nozzle diameter. This study investigates the effects of combined infill patterns in 3D printed products. Five patterns (solid, honeycomb, wiggle, grid, and rectilinear) were combined in samples to analyze their effects on mechanical properties for tensile strength analysis. Polylactic acid (PLA) samples were printed in different build orientations through two directions flat and on-edge. The limitation was that the software and machine could not combine the infill patterns. Thus, the patterns were designed and assembled in computer aided design (CAD) software. Finite element analysis (FEA) was used to determine the patterns' features and results showed honeycomb and grid have the highest strength while their weights were lighter compared to solid. Moreover, 0° samples in both flat and on-edge direction had the strongest layer adhesion and the best quality. In contrast, perpendicular samples like 60° and 75° showed poor adhesion and were the weakest specimens in both flat and on-edge, respectively. In brief, by increasing the build orientation, the strength decreases in this study.Mice lacking the GABAB(1) subunit of gamma-aminobutyric acid (GABA) type B receptors exhibit spontaneous seizures, hyperalgesia, hyperlocomotor activity, and memory impairment. Although mice lacking the GABAB(1) subunit are viable, they are sterile, and to generate knockout (KO) mice, it is necessary to cross heterozygous (HZ) mice. The aim of our study was to detect the metabolic differences between the three genotypes of GABAB(1) KO mice in order to further characterize this experimental animal model. Plasma samples were collected from wild-type (WT), HZ, and KO mice. Samples were analyzed by means of a gas chromatography-mass spectrometry (GC-MS) platform. Univariate t-test, and partial least square discriminant analysis (PLS-DA) were performed to compare the metabolic pattern of different genotypes. The metabolomic analysis highlighted differences between the three genotypes and identified some metabolites less abundant in KO mice, namely elaidic acid and other fatty acids, and chiro-inositol.
