Cancer therapy with tyrosine kinase inhibitors (TKIs) is a rapidly developing field, and several TKIs have been reported to have an impact on the activities of UDP-glucosyltransferases (UGTs), implying a potential risk for drug-drug interaction (DDI). Herein, we investigated the inhibitory effects of two commonly used TKIs, midostaurin and ruxolitinib, on human UGTs and quantitatively evaluated their DDI potential via UGT inhibition. It was found that midostaurin was a potent inhibitor of the majority of human UGTs, including UGT1A3, 1A4, 1A7, 1A8, 1A9, 1A10, 2B7, 2B15, and 2B17, with IC50 values lower than 4 μM (IC50 0.0128-3.85 μM), while ruxolitinib exhibited weak inhibition towards the activity of almost all the tested UGT isoforms. Furthermore, based on reversible inhibition, the co-administration of midostaurin at the clinical available dose was predicted to increase the plasma exposure to sensitive UGT1A3, 1A7, and 1A8 substrates by at least 61.4%, 25.6%, and 651%, respectively. In summary, our data identify that midostaurin is a potent inhibitor of the majority of human UGTs and may bring a potential risk of DDI via inhibition against UGT1A3, 1A7, and 1A8, while ruxolitinib cannot trigger UGT-mediated DDI due to its weak inhibition towards UGTs.Issues Non-medical cannabis policies are changing, including towards legalisation-with-regulation frameworks. New Zealand will hold a public referendum on cannabis legalisation in 2020. We reviewed data on cannabis use and health/social harms; policy reform options; experiences with and outcomes of reforms elsewhere; and other relevant considerations towards informing policy choices in the upcoming referendum. Approach Relevant epidemiological, health, social, criminal justice and policy studies and data were identified and comprehensively reviewed. Key findings Cannabis use is common (including in New Zealand) and associated with risks for health and social harms, mainly concentrated in young users; key harms are attributable to criminalisation. 'Decriminalisation' reforms have produced ambivalent results. Existing cannabis legalisation frameworks vary considerably in main parameters. Legalisation offers some distinct advantages, for example regulated use, products and user education, yet outcomes depend on essential regulation parameters, including commercialisation, and policy ecologies. While major changes in use are not observed, legalisation experiences are inconclusive to date, including mixed health and social outcomes, with select harms increasing and resilient illegal markets. It is unclear whether legalisation reduces cannabis exposure or social harms (e.g. from enforcement) for youth. XST-14 mouse Implications/conclusions No conclusive overall evidence on the outcomes of legalisation elsewhere exists, nor is evidence easily transferable to other settings. Legalisation offers direct social justice benefits for adults, yet overall public health impacts are uncertain. Legalisation may not categorically improve health or social outcomes for youth. Legalisation remains a well-intended, while experimental policy option towards more measured and sensible cannabis control and overall greater policy coherence, requiring close monitoring and possible adjustments depending on setting-specific outcomes.Aims Over the past two decades, animal intracerebral hemorrhage (ICH) model studies have indicated that iron, released after hemoglobin degradation, is neurotoxic. Iron phantom and animal experiments have shown that magnetic resonance imaging (MRI) relaxivity maps correlate with iron concentration. This study expands this into patients. Methods Eighteen human subjects with ICH underwent MRI at 3, 14, and 30 days. R2* relaxivity maps were used to calculate perihematomal iron concentrations and T2 imaging to determine hematoma and edema volumes. Results Perihematomal iron concentrations were increased at all three time points and decreased with distance from the hematoma. While perihematomal iron concentrations did not vary with hematoma size, the total iron overload (increased iron concentration x volume of affected tissue) did. Total iron overload correlated with edema volume. Conclusions These results demonstrate the feasibility of measuring perihematomal iron in ICH patients which may be important for monitoring treatment strategies and assessing efficacy noninvasively.Conceptual advances in faculty development are broadening to emphasize the importance of relationships and social networks to faculty learning.1 In‐person events traditionally provide a venue for these relationships to develop and sustain. In this study, extracts of M. oleifera leaves from South Korea and Cambodia were evaluated for their antioxidant and antiobesity activities and for food and natural medicine use. The extracts were made using water and ethanol with leaves from South Korea and Cambodia, and then, the ethanol extracts were further fractionated with ethyl acetate. The antioxidant and antiobesity activities of fractionated ethanol extracts were higher than those of water extracts. Although the expression of C/EBPα in 3T3-L1 cell differentiation did not have a concentration-dependent inhibitory effect on the M. oleifera leaf extracts, the expression of PPARγ, FAS, and ACC was inhibited in a concentration-dependent manner with the M. oleifera leaf extracts. This study shows that M. oleifera leaves from South Korea and Cambodia may be an effective candidate for antiobesity prevention. PRACTICAL APPLICATIONS Antioxidants and antiobesity factors are important for metabolic syndrome including obesity. Recently, natural antiobesity medication containing polyphenol ingredients has been developed to replace synthetic antiobesity medication, which has various side effects. This study evaluates the antioxidant and antiobesity activities of Moringa oleifera leaves from different cultivation regions. The leaves grow rapidly and leaf extracts contain a large amount of nutrients and phytochemicals, which enables commercial production of the leaves as natural antiobesity medications including functional foods and nutraceuticals.
