Renal Cell Carcinoma (RCC) is the ninth leading cause of death among kidney cancer. Xp11.2 translocation harboring TFE3 fusion proteins, act as an oncogene in translocation cancers that constitute the hallmark of translocation renal cell carcinoma (tRCC). G-quadruplex (G4), an alternative nucleic acid structure is an emerging and promising factor in cancer. The presence of G4 within the genome plays a pioneering role in cancer as it contributes to genomic aberration as well as inhibition in cell proliferation.
Here we discuss the link between G4 and tRCC. We compile the available information of G-quadruplex & propose their dual role in tRCC, suggesting both stabilization and destabilization of G-quadruplex could be considered targets for tRCC.
Our in Silico analysis of TFE3 and their three fusions partner's PRCC, SFPQ, and ASPSCR1 discloses a few putative G4 forming sequences (PQS) in their corresponding fusion gene or fusion transcript. Stabilization of G4 structure within fusion gene/transcript can be of great use towards potential therapeutics targeting fusion protein derived oncogenesis, as G4 is a serious menace for DNA polymerization, transcription & translation. G-quadruplex at intron-2 of the TFE3 has been reported to mediate its translocation also. Both stabilization and destabilization of the G4 structure would be a promising approach in the suppression of cancerous cell proliferation.

Selectively targeting G4 in oncogenic fusion transcript will emerge as potential druggable structures.Microorganisms are generally involved in the nucleation, growth and phase transformation of carbonate minerals, and influence the development of their morphology and polymorphism. However, understanding of the process of microbially induced carbonate precipitation (MICP) remains limited. Herein, MICP experiments were carried out using Curvibacter sp. HJ-1 and Arthrobacter sp. MF-2 in M2 medium, and the processes of MICP were monitored. Bacterial cells induced the precipitation of carbonate by creating favorable physicochemical conditions and acting as nucleation templates for carbonate particles and thereby, markedly influenced the morphology and growth of the carbonate structure. The extracellular polymeric substance (EPS) secreted by the bacteria was readily absorbed by the precipitated carbonate, which modified its crystal growth orientation. The MgCO3 content of Mg-calcite, induced by MF-2, was dramatically higher than that induced by HJ-1; HJ-1 promoted the formation and stability of aragonite. Multiple formation mechanisms coexisted during the evolution process of the mineral morphologies in the presence of the bacteria. The spherulites observed mainly evolved from dumbbell-like precursors in the presence of MF-2, whereas aggregate growth was the main formation mechanism of radial spherulites in the presence of HJ-1.
Due to the outbreaks such as SARS, bird flu and swine flu, which we frequently encounter in our century, we need fast solutions with no side effects today more than ever. Due to having vast ethnomedical experience and the richest flora (34% endemic) of Europe and the Middle East, Turkey has a high potential for research on this topic. Plants that locals have been using for centuries for the prevention and treatment of influenza can offer effective alternatives to combat this problem. In this context, 224 herbal taxa belonging to 45 families were identified among the selected 81 studies conducted in the seven regions of Turkey. However, only 35 (15.6%) of them were found to be subjected to worldwide in vitro and in vivo research conducted on anti-influenza activity. Quercetin and chlorogenic acid, the effectiveness of which has been proven many times in this context, have been recorded as the most common (7.1%) active ingredients among the other 56 active substances identified.
