In the 12,758 women included in the analysis (mean age 70 years, current smokers 4%, BMI ≥ 25 kg/m2 32%, low education 11%, current asthma 23%), 47% reported weekly use of at least one irritant cleaning product at home. Weekly use of irritant products was associated with a higher risk of current asthma (adjusted Odds-Ratio 1.17, 1.07-1.27). A statistically significant dose-response association was reported (p trend less then 0.0001), with both the number of irritant products used weekly (1 irritant 1.12, 1.02-1.23; 2 irritants 1.21, 1.05-1.39; 3 irritants or more 2.08, 1.57-2.75) and the frequency of use (1-3 days/week 1.12, 1.02-1.23; 4-7 days/week 1.41,1.22-1.64). A dose-response association was observed with the frequency of products used (p trend less then 0.05), for both non-allergic (4-7 days/week 1.27, 1.02-1.57) and allergic asthma (1.52, 1.27-1.82). In conclusion, weekly use of common cleaning irritants was associated with an increased risk of current asthma, whatever the allergic status.mRNAs play key roles in regulating diverse cellular functions. In many cases, mRNAs exhibit distinct intracellular localizations that are necessary for the spatiotemporal control of protein expression in cells. Therefore, imaging the localization and dynamics of these mRNAs is crucial for understanding diverse aspects of cellular function. In this review, we summarize how mRNA imaging can be achieved using tethered fluorescent proteins and fluorogenic aptamers. We discuss 'fluorogenic proteins' and describe how these recently developed RNA-regulated fluorescent proteins simplify mRNA imaging experiments.Age-related macular degeneration (AMD) has been linked to memory deficits, with no established neural mechanisms. We collected resting-state brain functional magnetic resonance imaging and standardized verbal recall tests from 42 older adults with AMD and 41 age-matched controls. We used seed-based whole brain analysis to quantify the strength of functional connectivity between hubs of the default mode network and a network of medial temporal regions relevant for memory. Our results indicated neither memory performance nor network connectivity differed by AMD status. However, the AMD participants exhibited stronger relationships than the controls between memory performance and connectivity from the memory network hub (left parahippocampal) to 2 other regions the left temporal pole and the right superior/middle frontal gyri. Also, the connectivity between the medial prefrontal cortex and posterior cingulate cortex of default mode network correlated more strongly with memory performance in AMD compared to control. We concluded that stronger brain-behavior correlation in AMD may suggest a role for region-specific connectivity in supporting memory in the context of AMD.Estrogen receptor alfa (ERα) is expressed in approximate 70% of breast cancer (BC) which is the most common malignancy in women worldwide. To date, the foremost intervention in the treatment of ER positive (ER+) BC is still the endocrine therapy. However, resistance to endocrine therapies remains a major hurdle in the long-term management of ER + BC. Although the mechanisms underlying endocrine resistance are complex, cumulative evidence revealed that ERα still plays a critical role in driving BC tumor cells to grow in resistance state. Fulvestrant, a selective estrogen receptor degrader (SERD), has moved to first line therapy for metastatic ER + BC, suggesting that removing ERα would be a useful strategy to overcome endocrine resistance. Proteolysis-Targeting Chimera (PROTAC) technology, an emerging paradigm for protein degradation, has the potential to eliminate both wild type and mutant ERα in breast cancer cells. Excitingly, ARV-471, an ERα-targeted PROTAC developed by Arvinas, has been in phase 1 clinical trials. In this review, we will summarize recent progress of ER-targeting PROTACs from publications and patents along with their therapeutic opportunities for the treatment of endocrine-resistant BC.Dysfunction of bone marrow mesenchymal stem cells (BMSCs) is involved in the pathogenesis of steroid-induced osteonecrosis of the femoral head (SONFH). Long noncoding RNAs (lncRNAs) contribute to biological effects exerted on BMSCs by regulating the expression of genes. RGT-018 purchase However, most recent studies have focused on the role of lncRNAs in modulating the imbalance between osteogenic and adipogenic differentiation but not apoptosis, proliferation, cell cycle and migration of BMSCs, especially in Dex-treated BMSCs. In this study, we conducted a microarray analysis to investigate the differential expression profiles of lncRNAs between human BMSCs (hBMSCs) obtained from patients with SONFH and control patients with femoral neck fracture. The microarray analysis showed that a total of 48 differentially expressed lncRNAs were identified in hBMSCs between the two groups, including 24 upregulated lncRNAs and 24 downregulated lncRNAs. Among of them, LINC00473 was found to be significantly downregulated. In the following study, we found that 10-6 mol/L Dex significantly inhibited proliferation, arrested cell cycle at the G1 phase, increased caspase-3 activity, induced apoptosis and impeded the migration of hBMSCs, while downregulation of the expression of LINC00473 produced results that were in line with the results of the microarray analysis in a time-dependent manner. Interestingly, upregulation of LINC00473 attenuated the negative effects caused by 10-6 mol/L Dex on hBMSCs, except for cell cycle arrest. Furthermore, it should be noted that LINC00473 had no effect on the proliferation and cell cycle of hBMSCs in the absence of Dex. Collectively, our data revealed that LINC00473 attenuated apoptosis, promoted the proliferation and migration of Dex-induced hBMSCs, which are not involved in interference with the cell cycle of hBMSCs.The correlation between potato components and Maillard reaction derivative harmful products (MRDHPs) formation during heat-processing was assessed in nine commercial potato varieties in China. Principal component analysis (PCA) combined with canonical correlation analysis (CCA) approach was performed to explore their relationships. The variables contributing most to the PCA results were extracted for CCA, and the results indicated that several amino acids, including lysine, tryptophan, alanine, phenylalanine, aspartate, and glutamate, have significant impacts on acrylamide and β-carboline heterocyclic amine formation. Moreover, 5-O-caffeoylquinic acid, 3-O-caffeoylquinic acid, α-solanine, and α-chaconine were also important factors associated with acrylamide and β-carboline heterocyclic amine formation. Optimally using raw potato varieties based on the impacts of these factors can help control MRDHP formation during thermal processing. For the first time, such approach was applied, which may be a useful tool for discovering the correlation of food components and MRDHPs.
