We investigated the synergistic effect between vancomycin and ß-lactams against vancomycin-susceptible (VSSA) and non-susceptible MRSA isolates (heterogeneous vancomycin-intermediate S. aureus (hVISA) and VISA).

To confirm synergistic activity, the representative strains of VISA, hVISA and VSSA were then selected for the time-kill curve method.

The combination of vancomycin with imipenem, meropenem and cefotaxime exhibited synergistic effects against 17 (2 VISA, 9 hVISA and 6 VSSA), 14 (3 VISA, 9 hVISA and 2 VSSA), and 5 (3 VISA and 2 hVISA) isolates respectively. Additive and indifferent effects were found in the remaining isolates but no antagonistic effect was observed. Using time-kill assay, the vancomycin combined with either imipenem or cefotaxime demonstrated synergism against both VISA and hVISA isolates, while the synergistic effect with meropenem was obtained in the VISA isolates only.

This study demonstrated in vitro enhanced antibacterial activity of vancomycin plus ß-lactams against clinical hVISA or VISA isolates. These combinations may be an alternative treatment for MRSA infections in clinical practice.

Targeting the new and unique proteins is an important medical strategy for treating breast cancer. It is quite important to find out proteins that have a role in the development of cancer. Sirtuins (SIRT) are well related in different physiological activities and connected with cancer. We aimed to determine the effect of radiotherapy on SIRT1 and SIRT2, which have not been yet clarified as a tumor suppressor or promoter.

22 women with non-metastatic breast cancer enrolled in the study. Blood samples were taken before and after radiotherapy, soluble SIRT1 and SIRT2 levels were determined with ELISA kits.

There was no difference in SIRT1 levels before and after radiotherapy (p=0.548). SIRT2 levels were significantly found to be decreased after radiotherapy (p=0.042). There was a strong and positive correlation before radiotherapy (p<0.001), and a moderate and positive correlation after radiotherapy (p=0.007) between SIRT1 and SIRT2.

These results suggest that SIRT2 may provide a new strategy for follow-up of breast cancer treatment. Additionally, by emphasizing the importance of SIRT2 in breast cancer, it opens ways to provide grounds for the development of the next generation of SIRT2-specific radiotracers. Finally, the most important thing in fact, the positive correlation between SIRT1 and SIRT2 both before and after radiotherapy, appears to be clear evidence suggesting more oncogenic roles of sirtuins.

Hyperbaric oxygen therapy (HBOT) causes insulin sensitivity, but the reason for this is not yet known. The aim of the present study was to investigate the effect of HBOT on insulin sensitivity via resistin, plasminogen activator inhibitor-I (PAI-I), and adiponectin.

The study was designed using HBOT and control groups, with eight rats in each group. After 20 days of HBOT under 2.5 atmospheres for 90 minutes, the fasting insulin (FI), resistin, PAI-I, homeostatic model assessment of insulin resistance scores (HOMA-IR), quantitative insulin sensitivity check index (QUICKI), fasting plasma glucose (FPG), triglyceride, and high-density lipoprotein cholesterol (HDL-C) in the plasma were measured. The resistin, PAI-I, and adiponectin mRNA expression levels were also measured in the adipose tissue.

Compared to the control group, the FI, FPG, and HOMA-IR scores were significantly lower in the HBOT group, whereas the HDL-C and QUICKI scores were found to be higher. In addition, the resistin, adiponectin, and PAI-I mRNA expression levels were also higher in the HBOT group.

The present study demonstrated that the HBOT had regulated the FI, FPG, and HDL-C associated with metabolic syndrome and diabetes mellitus. Moreover, the study showed that HBOT causes insulin sensitivity by raising adiponectin.
