We demonstrate the usefulness of our framework on single tube assembly of replicating plant expression constructs based on the geminivirus Bean yellow dwarf virus (BeYDV).Silicon promotes nodule formation in legume roots which is crucial for nitrogen fixation. However, it is very time-consuming and laborious to count the number of nodules and to measure nodule size manually, which led nodule characterization not to be study as much as other agronomical characters. Thus, the current study incorporated various techniques including machine learning to determine the number and size of root nodules and identify various root phenotypes from root images that may be associated with nodule formation with and without silicon treatment. Among those techniques, the machine learning for characterizing nodule is the first attempt, which enabled us to find high correlations among root phenotypes including root length, number of forks, and average link angles, and nodule characters such as number of nodules and nodule size with silicon treatments. The methods here could greatly accelerate further investigation such as delineating the optimal concentration of silicon for nodule formation.Stomatal pores control leaf gas exchange and are one route for infection of internal plant tissues by many foliar pathogens, setting up the potential for tradeoffs between photosynthesis and pathogen colonization. Anatomical shifts to lower stomatal density and/or size may also limit pathogen colonization, but such developmental changes could permanently reduce the gas exchange capacity for the life of the leaf. I developed and analyzed a spatially explicit model of pathogen colonization on the leaf as a function of stomatal size and density, anatomical traits which partially determine maximum rates of gas exchange. The model predicts greater stomatal size or density increases the probability of colonization, but the effect is most pronounced when the fraction of leaf surface covered by stomata is low. I also derived scaling relationships between stomatal size and density that preserves a given probability of colonization. These scaling relationships set up a potential anatomical conflict between limiting pathogen colonization and minimizing the fraction of leaf surface covered by stomata. Although a connection between gas exchange and pathogen defense has been suggested empirically, this is the first mathematical model connecting gas exchange and pathogen defense via stomatal anatomy. A limitation of the model is that it does not include variation in innate immunity and stomatal closure in response to pathogens. Nevertheless, the model makes predictions that can be tested with experiments and may explain variation in stomatal size and density among plants. The model is generalizable to many types of pathogens, but lacks significant biological realism that may be needed for precise predictions.Willow (Salix), a dioecious plant, is an important ornamental tree species in the world. Salix linearistipularis, a perennial woody plant species naturally distributed on the Songnen Plain saline-alkali land in northeast China, has a high saline condition. To study the sexual differences of S. linearistipularis in salinity tolerance, the physiological and transcriptional responses to salinity were compared between female and male cuttings. Under salinity stress, the female leaves exhibited higher superoxide dismutase and peroxidase activities and photosynthetic capacity, and lower H2O2 contents than those of male leaves. Under salinity stress, sodium (Na+) accumulation in female leaves was lower than that in the male leaves. The non-invasive micro-test showed that the net Na+ efflux in the salt-treated female roots was higher than that in male roots. Physiological responses revealed that female cuttings were more tolerant than males, which may be mainly due to females having lower leaf Na+ accumulation and higher root Na+ efflux capacity than males. Transcriptional analyses showed that 108 differentially expressed salt-responsive genes were identified in both female and male roots; most of these showed sexual differences in expression patterns under salinity stress. RNA-seq combined with qPCR analysis showed that the salt-induced expression of four Na+/H+ antiporter (NHX) genes (SlNHX3, 5, 6, 7) in female roots was higher than that in male roots. Transcriptional analyses revealed that the higher Na+ efflux capacity in female roots than in male roots may be closely related to the differential expression of salt-responsive genes, especially NHX genes.Rainfall regimes are expected to shift on a regional scale as the water cycle intensifies in a warmer climate, resulting in greater extremes in dry versus wet conditions. Such changes are having a strong impact on the agro-physiological functioning of plants that scale up to influence interactions between plants and microorganisms and hence ecosystems.
