Endoplasmic reticulum (ER) stress plays a critical role in the development of diabetic nephropathy (DN). We previously demonstrated that pyruvate-enriched oral rehydration solution improved glucometabolic disorders and ameliorated DN outcome in db/db mice. Here, we investigated the effects of pyruvate on high glucose (HG)-induced ER stress and apoptosis in HK-2 cells. Our results suggest that HG can induce reactive oxygen species (ROS) production, apoptosis, and ER stress in HK-2 cells, and that pyruvate treatment can ameliorate these effects and restore the expression of key proteins involved in ER stress. Thus, pyruvate may have potential for the development of novel strategies for the prevention and treatment of clinical DN. This article is protected by copyright. In many cases, the exact route of introduction into domestic pig herds cannot be determined, but most introductions are attributed to indirect virus transmission. In this review, we describe knowledge gained about different matrices that may allow introduction of the virus into pig herds. These matrices include uncooked pig meat, processed pig-derived products, feed, matrices contaminated with the virus and blood-feeding invertebrates. Knowledge gaps still exist, and both field studies and laboratory research are needed to enhance understanding of the risks for ASFV introductions, especially via virus-contaminated materials, including bedding and feed, and via blood-feeding, flying insects. Knowledge obtained from such studies can be applied to epidemiological risk assessments for the different transmission routes. Such assessments can be utilized to help predict the most effective biosecurity and control strategies. © 2020 Blackwell Verlag GmbH.We evaluated 345 wild animals from southern and south-eastern Brazil to understand their role in vaccinia virus (VACV) transmission cycle. VACV DNA was detected in rodents, marsupials, chiroptera and cingulate, expanding the knowledge of VACV host range in wildlife that could potentially act as source of infection in rural and urban areas. © 2020 Blackwell Verlag GmbH.INTRODUCTION Aflatoxin B1 (AFB1) is a toxic low-molecular-weight secondary metabolite of Aspergillus flavus and A. parasiticus. AFB1 was classified as a Group I carcinogen by the World Health Organisation for Research on Cancer in 1993. AFB1 is an unavoidable natural contaminant of some herbal medicine, able to cause serious health issues for humans consuming the related medicine. OBJECTIVE Therefore, this study aimed to develop an efficient fluorescence polarisation immunoassay (FPIA) and a rapid, low-cost, and easy-to-use membrane-based flow-through immunoassay (MBA) for determination of AFB1 in herbal medicine Origanum vulgare L., Rubus idaeus L., Urtica dioica L. and Sorbus aucuparia L. RESULTS A cut-off level of the developed MBA was 0.8 ppb. Validation of the developed test was performed with blank and spiked samples. selleck chemical Using three naturally contaminated or three artificially spiked samples. The FPIA showed a linear working range of 8.6 to 64 ppb, and a half maximal inhibitory concentration (IC50 ) of 24 ppb. CONCLUSION The results were in good correlation with the enzymelinked immunosorbent assay (ELISA) results (the IC50 0.1 ppb). Both the sample preparation and analysis are simple, cost-effective and easy to perform on-site in non-laboratory environments. Liquid chromatography with tandem mass spectrometry (LC-MS/MS) was used as a confirmatory technique. © 2020 John Wiley & Sons, Ltd.Having a comprehensive understanding of population structure, genetic differentiation and demographic history is important for the conservation and management of threatened species. High-throughput sequencing (HTS) provides exciting opportunities to address a wide range of factors for conservation genetics. Here, we generated HTS data and identified 266,884 high-quality single nucleotide polymorphisms from 82 individuals of Cupressus chengiana, to assess population genomics across the species' full range, comprising the Daduhe River (DDH), Minjiang River (MJR) and Bailongjiang River (BLJ) catchments in western China. admixture, principal components analysis and phylogenetic analyses indicated that each region contains a distinct lineage, with high levels of differentiation between them (DDH, MJR and BLJ lineages). MJR was newly distinguished compared to previous surveys, and evidence including coalescent simulations supported a hybrid origin of MJR during the Quaternary. Each of these three lineages should be recognized as an evolutionarily significant unit (ESU), due to isolation, differing genetic adaptations and different demographic history. Currently, each ESU faces distinct threats, and will require different conservation strategies. Our work shows that population genomic approaches using HTS can reconstruct the complex evolutionary history of threatened species in mountainous regions, and hence inform conservation efforts, and contribute to the understanding of high biodiversity in mountains. © 2020 John Wiley & Sons Ltd.Reactive dyes are pernicious pollutants in textile effluent, which are to be treated passably before discharging into the environment. In the present study, a potential dye degrading bacterial strain Pseudomonas guariconensis was isolated from paddy rhizosphere and was characterized by 16S rRNA gene sequencing. The biodegradation ability of the strain was evaluated by time-based study with immobilized bacterial cells in calcium alginate biocarrier matrix and also with free cells. The results indicated that the strain exhibited maximum degradation of 91% when immobilized in the biocarrier matrix. The enzymatic study revealed the production of oxidoreductase enzymes. The degraded products were identified as 2-amino-3-phenylpropanoic acid and benzoquinone by Gas Chromatography-Mass Spectroscopy (GC-MS) analysis, and a degradative pathway was derived based on the enzymatic profile. A packed bed column was designed using P. guariconensis VITSAJ5 immobilized in calcium alginate beads as a bio sorbent for the removal of Reactive Red 120.
