The successful licensure of vaccines for biodefense is contingent upon the availability of well-established correlates of protection (CoP) in at least two animal species that can be applied to humans, without the need to assess efficacy in the clinic. In this report we describe a multivariate model that combines pre-challenge serum antibody endpoint titers (EPT) and values derived from an epitope profiling immune-competition capture (EPICC) assay as a predictor in mice of vaccine-mediated immunity against ricin toxin (RT), a Category B biothreat. EPICC is a modified competition ELISA in which serum samples from vaccinated mice were assessed for their ability to inhibit the capture of soluble, biotinylated (b)-RT by a panel of immobilized monoclonal antibodies (mAbs) directed against four immunodominant toxin-neutralizing regions on the enzymatic A chain (RTA) of RT. In a test cohort of mice (n = 40) vaccinated with suboptimal doses of the RTA subunit vaccine, RiVax®, we identified two mAbs, PB10 and SyH7, which had EPICC inhibition values in pre-challenge serum samples that correlated with survival following a challenge with 5 × LD50 of RT administered by intraperitoneal (IP) injection. Analysis of a larger cohort of mice (n = 645) revealed that a multivariate model combining endpoint titers and EPICC values for PB10 and SyH7 as predictive variables had significantly higher statistical power than any one of the independent variables alone. Establishing the correlates of vaccine-mediated protection in mice represents an important steppingstone in the development of RiVax® as a medical countermeasure under the United States Food and Drug Administration's "Animal Rule."A series of scandals involving the rabies vaccine and the diphtheria, tetanus, and pertussis combined vaccine (DTP) were reported in July 2018, in China. Our goal was to assess the effects of the vaccine scandal on parents' attitudes and decisions regarding vaccination. A cross-sectional study was conducted by contacting kindergarteners' parents through an online questionnaire from March 1 to 8, 2019. In total, 14,300 valid questionnaire responses were received, and 92.53% of respondents were aware of the recent vaccine scandal. Among parents who were aware of the vaccine scandal, 13.62% preferred that their children receive optional vaccines (optional vaccine group) or delay, stop or reject vaccinations (hesitate group) instead of receiving mandatory vaccines (mandatory vaccine group). Awareness of the vaccine scandal and self-reporting of vaccine-associated side effects in their children were more common among respondents in the optional vaccine and the hesitate groups (P less then 0.001). Compared with the mandatory vaccine group, parents in the hesitate group were more likely to report having attempted to dissuade others from vaccinating their children (P less then 0.001). Because the scandal changed parents' decisions regarding vaccination of their children, efforts should be made to restore public confidence in vaccines and to boost immunization rates.
Guizhou Province of China implements a vaccination program specifying that children's vaccination records are to be checked upon entry to kindergarten and primary school; children missing one or more recommended vaccinations are to be offered the missed vaccinations; school-level vaccination coverage levels are assessed at the time of school enrollment and six months later to monitor compliance rates.
We obtained the number of doses of each vaccine in the national immunization schedule that were administered before and six months after kindergarten and school enrollment and reported to Guizhou Province during 2004 through 2018. MCV2 coverage at kindergarten entry increased from 53% in 2004 to 98% in 2018. So, they are susceptible to infections. Studies showed that, mesenchymal stem cells (MSCs) have protective effects on the function of neutrophils and an approach that MSCs use to apply their effects, is secreting soluble factors and exosomes. So, we investigated the effects of MSC-exosomes and MSC-conditioned media (MSC-CM) on the function and apoptosis of neutrophils in CGD patients. In this study, neutrophils were isolated from healthy donors and CGD patients and then incubated with exosomes or CM that were prepared from MSCs. Then, neutrophil respiratory burst, apoptosis and phagocytosis capacity were evaluated by NBT assay, Annexin V-PI method and Giemsa staining. It was demonstrated that both MSC-exosomes and CM could improve the phagocytosis capacity and ROS production of neutrophils in CGD patients and healthy donors. In contrast to the healthy group, in CGD patients, exosomes significantly reduced the percentage of viable neutrophils. This report indicated that MSC exosomes and CM could increase the function of the neutrophils isolated from CGD patients. But decreasing the number of the living cells is one of the limitations of them. However, it is hoped that this intervention will be developed in future studies to minimize its limitations.
To investigate the relationship between the right and left foot posture in terms of body mass index (BMI), sex, and age in adolescents aged 10 to 14.
A total of 1400 adolescents were included. For assessment, the Foot Posture Index (FPI-6) was used. Each foot was assessed and ranked as supinated, normal, or pronated by the sum of the FPI-6 criteria. Each criterion was scored on a scale of -2 to +2, with negative for supinated and positive for pronated posture. A linear mixed model with repeated measures was used for statistical analysis.
Sex, BMI, and right and left foot are associated with FPI-6. The total score attributed for male sex (β = 0.29, P = .04) and the left foot (β = 0.73, P < .001) was higher (male right foot mean ± standard deviation [SD], 3.09 ± 2.84, male left foot 3.76 ± 2.80; female right foot mean ± SD, 2.28 ± 2.61, female left foot 3.45 ± 2.66; laterality for left foot mean ± SD, 3.55 ± 2.71, laterality for right foot mean ± SD, 2.82 ± 2.7).
