Plasma free VPA C
is a better predictor than peripheral blood mononuclear cell histone acetylation for brain H3 and H4 acetylation (r
=0.7189 for H3K27ac, r
=0.7189 for pan-H3ac, and r
=0.7554 for pan-H4ac; p<0.0001).

Up to 150mg/kg VPA can be safely tolerated as 90-min intravenous infusion in a swine CA model. High-dose VPA induced dose-dependent increases in brain histone H3 and H4 acetylation, which can be predicted by plasma free VPA C
as the pharmacodynamics biomarker for VPA target engagement after CA.

Prognosis in out-of-hospital cardiac arrest (OHCA) depends on cardiopulmonary resuscitation (CPR) duration. Therefore, the optimal biphasic defibrillation waveform shows high conversion rates besides low energy. learn more Matthew Fishler theoretically predicted it to be truncated ascending exponential. We realised a prototypic defibrillator and compared ascending with conventional rectangular waveforms in modelled OHCA and CPR.

Approved by the authorities, 57 healthy swine (Landrace×Piétrain) were randomised to ASCDefib (
26) or CONVDefib (
26). Five swine served as sham control. We induced ventricular fibrillation (VF) electrically in anaesthetised swine randomised to ASCDefib or CONVDefib and discontinued mechanical ventilation. After 5min of untreated cardiac arrest, we started CPR with mechanical chest compressions and ventilation. We performed transthoracic biphasic defibrillations after 2, 4, 6 and 8min CPR targeting 4J/kg in either group. Depending on the randomised group, the defibrillation protocol was either three ascending followed by one rectangular waveform (ASCDefib) or three rectangular followed by one ascending waveform (CONVDefib).

Under our model-specific conditions, VF was initially terminated by 13/80 ascending waveforms and 13/79 rectangular waveforms and persistent return of spontaneous circulation was achieved in 8/26 (ASCDefib) vs. 10/26 (CONVDefib) animals. Mean current rather than waveform design was predictive for defibrillation success in a generalised linear model.

Contrary to theoretical assumptions, transthoracic biphasic defibrillation with ascending waveforms is not superior to rectangular waveforms in modelled OHCA. We advocate defibrillation dosage to be guided by current, that has proven its predictive value again.

84-02.04.2017.A176.Few semiconducting polymers are known that possess more than one semi-crystalline structure. Guidelines for rationalizing or creating polymorphism in these materials do not exist. Two different semi-crystalline polymorphs, β 1 and β 2, and an amorphous α phase have recently been identified for alternating diketopyrrolopyrrole-quaterthiophene copolymers (PDPP4T). The polymorphs differ structurally by the π-π stacking distance, and electronically by the optical bandgap and charge carrier mobility. Here we investigate the corresponding terthiophene (PDPP3T) derivatives, to study the effect of the relative orientation of adjacent DPP units on the polymorphism. In PDPP3T, the relative orientation of DPP units alternates along the chain, while in PDPP4T it is constant. We show that the two polymorphs, β 1 and β 2, can also be generated for a PDPP3T polymer in solution and thin film. Interestingly, compared to PDPP4T, more solvents can induce the two distinct semi-crystalline polymorphs for PDPP3T via a β 1 → α → β 2 polymorphic transition.A comprehensive understanding of the structure, self-assembly mechanism, and dynamics of one-dimensional supramolecular polymers in water is essential for their application as biomaterials. Although a plethora of techniques are available to study the first two properties, there is a paucity in possibilities to study dynamic exchange of monomers between supramolecular polymers in solution. We recently introduced hydrogen/deuterium exchange mass spectrometry (HDX-MS) to characterize the dynamic nature of synthetic supramolecular polymers with only a minimal perturbation of the chemical structure. To further expand the application of this powerful technique some essential experimental aspects have been reaffirmed and the technique has been applied to a diverse library of assemblies. HDX-MS is widely applicable if there are exchangeable hydrogen atoms protected from direct contact with the solvent and if the monomer concentration is sufficiently high to ensure the presence of supramolecular polymers during dilution. In addition, we demonstrate that the kinetic behavior as probed by HDX-MS is influenced by the internal order within the supramolecular polymers and by the self-assembly mechanism.Reliable, wide range, and highly sensitive joint movement monitoring is essential for training activities, human behavior analysis, and human-machine interfaces. Yet, most current motion sensors work on the nano/microcracks induced by the tensile deformation on the convex surface of joints during joint movements, which cannot satisfy requirements of ultrawide detectable angle range, high angle sensitivity, conformability, and consistence under cyclic movements. In nature, scorpions sense small vibrations by allowing for compression strain conversion from external mechanical vibrations through crack-shaped slit sensilla. Here, we demonstrated that ultraconformal sensors based on controlled slit structures, inspired by the geometry of a scorpion's slit sensilla, exhibit high sensitivity (0.45%deg-1), ultralow angle detection threshold (~15°), fast response/relaxation times (115/72 ms), wide range (15° ~120°), and durability (over 1000 cycles). Also, a user-friendly, hybrid sign language system has been developed to realize Chinese and American sign language recognition and feedback through video and speech broadcasts, making these conformal motion sensors promising candidates for joint movement monitoring in wearable electronics and robotics technology.
