This study examined the obligate aerobe, Pseudomonas putida, using acetate as the sole carbon and energy source, and respiration via an anode as the terminal electron acceptor under anoxic conditions. P. putida showed significantly different acetate assimilation in a closed-circuit microbial fuel cell (CC-MFC) compared to an open circuit MFC (OC-MFC). More than 72% (2.6 mmol) of acetate was consumed during 84 hrs in the CC-MFC in contrast to the no acetate consumption observed in the OC-MFC. The CC-MFC produced 150 μA (87 C) from acetate metabolization. Electrode-based respiration reduced the NADH/NAD+ ratio anaerobically, which is similar to the aerobic condition. The CC-MFC showed significantly higher acetyl-CoA synthetase activity than the OC-MFC (0.028 vs. 0.001 μmol/min/mg), which was comparable to the aerobic condition (circa 60%). Overall, electrode-based respiration enables P. putida to metabolize acetate under anoxic conditions and provide a platform to regulate the bacterial redox balance without oxygen.The integral valorization of potential sugars (cellulosic and hemicellulosic) from spent coffee grounds (SCG), a lignocellulosic residue, is proposed in this work. The current study recorded eye movements of British and Japanese infants (aged 10 and 16 months) and adults, who were presented with static and dynamic faces on screen. Cultural differences were observed across all age groups, with British participants exhibiting more mouth scanning, and Japanese individuals showing increased central face (nose) scanning for dynamic stimuli. Age-related influences independent of culture were also revealed, with a shift from eye to mouth scanning between 10 and 16 months, while adults distributed their gaze more flexibly. Against our prediction, no age-related increases in cultural differences were observed, suggesting the possibility that cultural differences are largely manifest by 10 months of age. Overall, the findings suggest that individuals adopt visual strategies in line with their cultural background from early in infancy, pointing to the development of a highly adaptive face processing system that is shaped by early sociocultural experience.Survival in high-risk neuroblastoma (HR-NB) patients remains poor despite multimodal treatment. We aimed to identify HR-NB patients with worse outcomes by analyzing the genomic instability derived from segmental chromosomal aberrations. We calculated 3 genomic instability indexes for primary tumor SNP array profiles from 127 HR-NB patients (1) Copy number aberration burden (%gainslength+%losseslength), (2) copy number load (CNL) (%gainslength-%losseslength) and (3) net genomic load (NGL) (%gainsamount-%lossesamount). Tumors were classified according to positive or negative CNL and NGL genomic subtypes. The impact of the genomic instability indexes on overall survival (OS) was assessed with Cox regression. We identified 38% of HR-NB patients with poor 5-year OS. A negative CNL genomic background was related to poor prognosis in patients ≥18 months showing tumors with homogeneous MYCN amplification (9.5% survival probability, P 2.4% CNL, P less then 0.01). A positive CNL genomic background was associated with worse outcome in patients with heterogeneous MYCN amplification (22.5% survival probability, P less then 0.05). We conclude that characterizing a tumor genomic background according to predominance of genome gained or lost contributes toward improved outcome prediction and brings greater insight into the tumor biology of HR-NB patients.
