Methods The current prospective observational study was conducted in three hospitals in Istanbul, Turkey from March 10 to June 22, 2020. The participants were categorized as high-risk or low-risk OSA according to the Berlin questionnaire that was administered in the out-patient clinic, in hospital, or shortly after discharge from hospital blinded to the clinical outcomes. A modified high-risk (mHR)-OSA score based on the snoring patterns (intensity and/or frequency), breathing pauses, and morning/daytime sleepiness, without taking obesity and hypertension into account, were used in the regression models. Docusate is often utilized for OIC prophylaxis and treatment because of its low cost, easy accessibility and tolerability. However, the costs of docusate use may not outweigh the benefits given limited reported efficacy. The authors seek to provide the first critical review of clinical evidence for docusate use in OIC management. PubMed, Google Scholar, OVID Medline, and EMBASE were queried for primary literature, guidelines, and consensus recommendations (CR) evaluating docusate use for OIC management. Thirteen guidelines or CR and four primary literature studies were evaluated. The primary literature studies had significant design limitations and collectively revealed non-compelling evidence for use. The cytokine storm in SARS-CoV2 infection has been linked with both diabetes and Vitamin D deficiency. This study examined the hypothesis that supplementation with vitamin D, in combination with l-cysteine (LC), is better at reducing oxidative stress and thereby, more effective, at inhibiting the secretion of the pro-inflammatory cytokines, Interleukin-8 (IL-8) and monocyte chemoattractant protein-1 (MCP-1) in U937 monocytes exposed to high glucose concentrations. Methods U937 monocytes were pretreated with 1,25 (OH)2 vitamin D (VD, 10 nM) or LC (250 µM) or VD + LC for 24 h and then exposed to control or high glucose (HG, 25 mM) for another 24 h. Results There were significantly greater reactive oxygen species (ROS) levels in monocytes treated with HG than those in controls. Combined supplementation with VD and LC showed a more significant reduction in ROS (46%) in comparison with treatment with LC (19%) or VD (26%) alone in monocytes exposed to HG. Similarly, VD supplementation, together with LC, caused a more significant inhibition in the secretion of IL-8 (36% versus 16%) and MCP-1 (46% versus 26%) in comparison with that of VD (10 nM) alone in high-glucose treated monocytes. Conclusions These results suggest that combined supplementation with vitamin D and LC has the potential to be more effective than either VD or LC alone in lowering the risk of oxidative stress and inflammation associated with type 2 diabetes or COVID-19 infection. Further, this combined vitamin D with LC/N-acetylcysteine may be a potent alternative therapy for SARS-CoV2 infected subjects. This approach can prevent cellular damage due to cytokine storm in comorbid systemic inflammatory conditions, such as diabetes, obesity, and hypertension.Brombuterol, a new β-adrenergic agonist to enhance animal growth and increase feeding efficiency, is forbidden as an additive in animal feed for livestock production due to its adverse effects on consumers. In this study, a highly specific and sensitive monoclonal antibody was obtained and an indirect competitive monoclonal ELISA was developed to detect brombuterol, with an IC50 value of 0.1 μg/kg (μg/L) and no cross-reactivity to other structurally related β-adrenergic agonists. The average recovery of brombuterol using the icELISA method ranged from 72.9% to 106.4% with the coefficient of variation lower than 18.9%, which was determined by analysing spiked animal feed, swine urine, pork and liver samples (n = 5). Finally, the icELISA gave results having a good correlation with those obtained by liquid chromatography-tandem mass spectrometry. These results demonstrated that the developed icELISA for the detection of brombuterol is highly specific, sensitive, and reliable, indicating good potential for use in the area of food safety to improve consumer protection.
