This research proposes a nondestructive single-sensor acoustic emission (AE) scheme for the detection and localization of cracks in steel rail under loads. In the operation, AE signals were captured by the AE sensor and converted into digital signal data by AE data acquisition module. The digital data were denoised to remove ambient and wheel/rail contact noises, and the denoised data were processed and classified to localize cracks in the steel rail using a deep learning algorithmic model. The AE signals of pencil lead break at the head, web, and foot of steel rail were used to train and test the algorithmic model. In training and testing the algorithm, the AE signals were divided into two groupings (150 and 300 AE signals) and the classification accuracy compared. The deep learning-based AE scheme was also implemented onsite to detect cracks in the steel rail. The total accuracy (average F1 score) under the first and second groupings were 86.6% and 96.6%, and that of the onsite experiment was 77.33%. The novelty of this research lies in the use of a single AE sensor and AE signal-based deep learning algorithm to efficiently detect and localize cracks in the steel rail, unlike existing AE crack-localization technology that relies on two or more sensors and human interpretation.Prior studies have found that exercise has a positive effect on depressive symptoms in the general population. We examined whether regular exercise is related to depressive symptoms in 5379 Korean adults aged ≥ 55 years using data from a 2016 survey administered in the Korean Longitudinal Study of Aging. We found that a lack of regular exercise was significantly related to an increased frequency of depressive symptoms (OR = 1.18, 95% CI = 1.03-1.35). Moreover, hand-grip strength may increase the effect of regular exercise on depressive symptoms in individuals 65 years and older (OR = 1.01 vs. 1.70, 95% CI = 1.05-1.96). Moreover, because sperm suitable for oocyte fertilization is released from oviduct epithelial cells (OECs), in response to the capacitation process, we established an in vitro OEC culture model to select a sperm population with potential fertilizing capacity in this species. We demonstrated E-cadherin and cytokeratin expression in cultures of OECs obtained. When sperm-OEC cocultures were performed, the attached spermatozoa were motile and presented an intact acrosome, suggesting a selection by the oviductal model. When co-cultures were incubated in capacitating conditions a greater number of alive (p less then 0.05), capacitated (p less then 0.05), with progressive motility (p less then 0.05) and with the intact acrosome sperm population was observed (p less then 0.05) suggesting that the sperm population released from OECs in vitro presents potential fertilizing capacity. Two related types of guanine nucleotide exchange factors (GEFs) activate Arf1 at different Golgi sites. In yeast, Gea1 in the cis-Golgi and Gea2 in the medial-Golgi activate Arf1 to form COPI-coated vesicles for retrograde cargo sorting, whereas Sec7 generates clathrin/adaptor-coated vesicles at the trans-Golgi network (TGN) for forward cargo transport. A central question is how the same activated Arf1 protein manages to assemble different coats depending on the donor Golgi compartment. A previous study has postulated that the interaction between Gea1 and COPI would channel Arf1 activation for COPI vesicle budding. Here, we found that the p24 complex, a major COPI vesicle cargo, promotes the binding of Gea1 with COPI by increasing the COPI association to the membrane independently of Arf1 activation. Furthermore, the p24 complex also facilitates the interaction of Arf1 with its COPI effector. Therefore, our study supports a mechanism by which the p24 complex contributes to program Arf1 activation by Gea1 for selective COPI coat assembly at the cis-Golgi compartment.Neuromuscular electrical stimulation (NMES) is useful for muscle strengthening and for motor restoration of stroke patients. Using a portable ultrasound instrument, we developed an M-mode imaging protocol to visualize contractions elicited by NMES in the quadriceps muscle group. To quantify muscle activation, we performed digital image processing based on the Teager-Kaiser energy operator. The proposed method was applied for 35 voluntary patients (18 women and 17 men), of 63.8 ± 14.1 years and body mass index (BMI) 30.2 ± 6.70 kg/m2 (mean ± standard deviation). Biphasic, rectangular electric pulses of 350 µs duration were applied at two frequencies (60 Hz and 120 Hz), and ultrasound was used to assess the sensory threshold (ST) and motor threshold (MT) amplitude of the NMES signal. The MT was 23.4 ± 4.94 mA, whereas the MT to ST ratio was 2.69 ± 0.57.
