Further, membrane-modified MSCs exhibited a better therapeutic effect than wide-type MSCs in a rabbit model of PCD. Thus, this unique cell membrane modification strategy provides a new cell-based therapeutic approach for the early treatment of articular cartilage defects and other joint diseases.Severe peripheral nerve injuries often result in permanent loss of function of the affected limb. Current treatments are limited by their efficacy in supporting nerve regeneration and behavioral recovery. Here we demonstrate that electrical stimulation through conductive nerve guides (CNGs) enhances the efficacy of human neural progenitor cells (hNPCs) in treating a sciatic nerve transection in rats. Electrical stimulation strengthened the therapeutic potential of NPCs by upregulating gene expression of neurotrophic factors which are critical in augmenting synaptic remodeling, nerve regeneration, and myelination. Electrically-stimulated hNPC-containing CNGs are significantly more effective in improving sensory and motor functions starting at 1-2 weeks after treatment than either treatment alone. Electrophysiology and muscle assessment demonstrated successful re-innervation of the affected target muscles in this group. Furthermore, histological analysis highlighted an increased number of regenerated nerve fibers with thicker myelination in electrically-stimulated hNPC-containing CNGs. The elevated expression of tyrosine kinase receptors (Trk) receptors, known to bind to neurotrophic factors, indicated the long-lasting effect from electrical stimulation on nerve regeneration and distal nerve re-innervation. These data suggest that electrically-enhanced stem cell-based therapy provides a regenerative rehabilitative approach to promote peripheral nerve regeneration and functional recovery.
To evaluate formulations of 1 % silver (Ag) nanoparticles for treating traumatic lesions induced in the oral mucosa of rats, because these lesions are commonly observed in the dental clinic, and their therapeutic forms are scarce.
Wistar rats were punch-injured (two circular fragments, 4.0 mm in diameter) in the oral mucosa (one on each side), and were treated topically (twice per week) with the treatments/groups including no injury, control, vehicle, diluted Ag, soluble Ag, and solid Ag. On the 2nd, 7th, and 14th days postinjury, biopsies were collected for immunohistochemistry and biochemical analysis.
The group diluted Ag revealed a higher level of inflammatory infiltrate on the 2nd day, whereas solid Ag presented lower levels. The Ag solid group presented higher IL-1β on the 2nd day and increased IL-10 and TGF-β1 throughout the follow-up. Moreover, all three Ag groups presented lower levels of oxidative stress markers and, on the 7th day, the diluted Ag and solid Ag groups revealed higher antioxidants. Diluted Ag and soluble Ag groups presented greater blood vessels proliferation, whereas soluble Ag and solid Ag groups revealed greater VEGF on the 2nd and 14th days. Furthermore, all three Ag groups were highlighted during fibroplasia, although collagenesis was similar to that observed in the control group.
Although diluted Ag was noticeable for its important angiogenesis and fibroplasia, solid Ag was the most suitable formulation in healing oral lesions as it efficiently controlled inflammation and oxidative stress, thus favoring angiogenesis and tissue formation.

Chondrocytes, isolated from articular cartilage from the knee, hip, and shoulder joints of Wistar rats, were subjected to 20 % tensile stress under hypoxic (5% O2) conditions for 24 h. HIF-1α and aggrecan expression was significantly enhanced with mechanical stimulation under hypoxia but not significantly altered with mechanical stimulation under normoxia. The nuclear translocation of HIF-1α was enhanced by mechanical stress under hypoxia. Under both normoxia and hypoxia, a disintegrin and metalloproteinase with thrombospondin motifs (ADAM-TS) 5 expression was significantly reduced with mechanical stimulation compared to that in the group without mechanical stimulation. However, HIF-1α knockdown mitigated changes in aggrecan and ADAM-TS5 expression mediated by mechanical stimulation under hypoxia. EPZ005687 purchase The effects of treadmill running on HIF-1α production in the articular cartilage of rat knee joints were also analyzed. HIF-1α production increased in the moderate running group and decreased to the same levels as those in the control group in the excessive running group. This suggests that HIF-1α regulates aggrecan and ADAM-TS5 expression in response to mechanical stimulation under hypoxia and general mechanical stimulation in articular cartilage under hypoxia, while controlling cartilage homeostasis.Rapidly and accurately monitoring the physiological and biochemical parameters of grape leaves is the key to controlling the quality of wine grapes. In this study, a Pika L hyperspectral imaging system (400-1000 nm) was used to acquire hyperspectral image information from grape leaves. New vegetation indices were developed on the basis of the screened sensitive wavebands to quantitatively predict changes in these parameters (the leaf chlorophyll level (SPAD), leaf nitrogen content (LNC) and chlorophyll fluorescence parameters (ChlF parameters)). The results showed that SPAD reached its maximum at the grape turning stage and declined thereafter. The vegetation index (D735-D573)/(D735+D573) was able to predict SPAD fairly well (validation dataset R2 = 0.50). LNC reached its maximum at the grape maturity stage. D682/R525 was highly correlated with LNC. Except for NPQ, all ChlF parameters showed a decreasing trend from the fruiting to harvesting stages. Among the dark-adapted ChlF parameters, FV/Fm had the strongest correlation to the new vegetation index (D735-D544)/(D735+D544) (modelling dataset R2 = 0.
