The chemical constituents of Coptidis Rhizoma and pharmacological effects of berberine were reviewed in this study, which was expected to provide references for the further research, development of and clinical application of Coptidis Rhizoma and berberine.Since the first separation, protoplasts have played an important role in basic and applied research of plant biology and molecular biology. There are a lot of reports on protoplast preparation and transformation, especially in the selection of explants, enzymatic environment and transformation mode. Protoplasts have also been widely used in gene expression and regulation, especially in subcellular localization, protein interaction and promoter activity. This paper attempts to provide a new way of thinking and technical method for the analysis of molecular mechanism of traditional Chinese medicine quality formation in the future.In the theories of empirical identification of "arguments on quality" and "five elephants and seven originals", color is regarded as one of the important indicators in the quality evaluation of Chinese medicine. The color of medicinal material including the surface color, the internal color, and the color after processing is caused by the pigment in the cells, which is a characteristic of the "optimal shape". Most pigments have a wide range of pharmacological activities such as anti-oxidation, anti-tumor, anti-atherosclerosis, prevention of coronary heart disease, protection of cardiovascular function, enhancement of immunity, etc. Therefore, the "optimal color" of medicinal materials is unified with the "high quality". This article systematically reviews the research status of "quality discrimination by color", the correlation between L~*, a~*, b~* color space and active ingredients such as alkaloids, flavonoids, terpenes, etc. to explain the "quality discrimination by color" in quantitative characterization. We also summary the research progress on the biosynthesis and regulation of the main pigment components of traditional Chinese medicine, and analyze the biological causes that affect the accumulation of the main pigments. We aim to provide a reference for the theory "quality discrimination by color" in biological knowledge to establish a modern quality control system for Chinese herbal medicines.With the application and popularization of PCR(polymerase chain reaction) technology in drug quality control, the ge-neral rule of PCR has been approved to be listed in the Chinese Pharmacopoeia(2020 edition). This rule has set general quality stan-dards for species identification in Chinese herbal medicine and biochemical medicine, as well as quality control of other medicines. The general rule is composed of general requirements for instruments, general requirements for PCR system, operational suitability test, measurement procedure(sample pretreatment, templates preparation, primers, nucleic acid amplification, reaction products detection, result judgment) and notes, etc. In this article, we introduced the general rule of PCR and its drafting instructions. Furthermore, its application perspective in Chinese herbal medicine and biochemical medicine was discussed.To introduce current literature reporting situations of the off-label drug use(OLDU) by analyzing relevant literatures published in China, this study comprehensively focused on literatures about OLDU in China in seven Chinese and English databases, then extracted and analyzed the data by different literature types. A total of 667 papers were analyzed. The number of literatures about OLDU data analyzed in hospitals was 325, and the number of clinical studies relating to OLDU was 329, in which case series and case reports were the majority(69.91%). In addition, there were 13 expert consensuses of OLDU and another 56 studies about drug use based on the real-world data characteristics and influencing factors. The number of OLDU data studies has increased year by year. Based on the existing studies, there were more western medicine reports than traditional Chinese medicines, and OLDU types were mainly for over-dosage use. The literatures from OLDU data in hospital were mostly limited to one or several tertiary hospitals in a certain area, and the OLDU types were not uniform.
