To better understand the highly mechanical pathophysiology of pulmonary embolism, we set out to develop an in-vitro thrombus mimic and to test this mimic under large deformation simple shear. In addition to reporting on the mechanics of our mimics under simple shear, we explore the sensitivity of their mechanics to coagulation conditions and blood storage time, and compare three hyperelastic material models for their ability to fit our data. We found that thrombus mimics made from whole blood demonstrate strain-stiffening, a negative Poynting effect, and hysteresis when tested quasi-statically to 50% strain under simple shear. Additionally, we found that the stiffness of these mimics does not significantly vary with coagulation conditions or blood storage times. Of the three hyperelastic constitutive models that we tested, the Ogden model provided the best fits to both shear stress and normal stress. In conclusion, we developed a robust protocol to generate regularly-shaped, homogeneous thrombus mimics that lend themselves to simple shear testing under large deformation. Future studies will extend our model to include the effect of maturation and explore its fracture properties toward a better understanding of embolization.Indoor biomass burning produces large amounts of small particles and hazardous contaminants leading to severe air pollution and potentially high health risks associated with inhalation exposure. Personal samplers provide more accurate estimates of inhalation exposure. In this study, inhalation exposure to size-segregated particles and particulate polycyclic aromatic hydrocarbons (PAHs) for the biomass user was studied by deploying personal samplers. The study found that daily PM2.5 inhalation exposure level was as high as 121 ± 96 μg/m3, and over 84% was finer PM1.0. For PAHs, the exposure level was 113 ± 188 ng/m3, with over 77% in PM1.0. High molecular weight PAHs with larger toxic potentials enriched in smaller particles resulting in much high risks associated with PAHs inhalation exposure. Indoor exposure contributed to ~80% of the total inhalation exposure as a result of high indoor air pollution and longer residence spent indoor. The highest exposure risk was found for the male smoker who conducted cooking activities at home.Citric acid (CA) can regulate the balance of anions and cations in plants, and improve their resistance to heavy metals. It is not clear if foliar application with CA has any effect on migration of Cd and Mn in rice plant. In this work, a low-Cd-accumulating indica rice line (P7) and a high-Cd-accumulating line (HZ) were used to investigate the influence of CA on the transport of Cd and Mn as well as amino acid metabolism in grains. Content of Cd in grains and other organs increased with the increase of Cd content (0.1-2.4 mg kg-1) in soil, while decreased with the foliar application with CA. With the increase of Cd content in rice grains, the content of most amino acids in HZ, P7, HZ+CA and P7 + CA showed an obvious decline trend. Foliar application with CA efficiently raised the MnCd ratio in grains and nodes of both HZ and P7. Meanwhile, the expression levels of OsNramp2, 3 and 5 in panicles were efficiently enhanced by CA application when plants grew in soil with Cd content of 0.6-2.4 mg kg-1. The increasing effect of CA on the content of 4 amino acids (i.e., Glu, Phe, Thr and Ala) in grains was related to varieties and Cd pollution. These results indicate that foliar application with CA can regulate the transport of Cd and Mn in the opposite directions in tissues and inhibit Cd accumulation in grains by enhancing expression of OsNRAMP 2, 3 or 5 and triggering the defense response of some amino acids in Cd-contaminated environment.Fine particulate matter (PM2.5) constitutes the most significant air pollutant that causes health risks. However, the mechanism(s) underlying PM2.5-induced male reproductive injury has not been clarified. In the present study we explored whether PM2.5 activated the inositol-requiring enzyme 1 (IRE1)/c-Jun NH 2-terminal kinase (JNK)/autophagy-signaling pathway, and whether this pathway mediated reproductive injury in male rats. Dyngo-4a We established a male Sprague-Dawley rat model of PM2.5 (1.5 mg/kg) exposure-induced reproductive injury, and observed the intervention effects of STF083010 (an IRE1 inhibitor, 1 mg/kg). After 4 weeks of exposure, reproductive injury-related indicators and IRE1-cascade protein expression were analyzed. Our results showed that sperm quality and serum testosterone level significantly decreased and apoptotic index increased after exposure to PM2.5. After STF083010 intervention, sperm quality and serum testosterone level were significantly improved, while the apoptotic index was reduced. Under light microscopy, we observed that the structure of spermatogenic cells in the PM2.5 group was loose, and that the numbers of spermatogenic cells and mature spermatozoa were reduced. After STF083010 intervention, the structural damage to spermatogenic cells was improved, and the number of cells shed was reduced. Western blotting analysis showed that the expression of IRE1, phosphorylated JNK (p-JNK), beclin-1, and microtubule-associated protein 1 light chain 3(LC3)II/LC3I proteins was significantly upregulated, and that the expression of p62 protein was significantly downregulated in the PM2.5 group. The concomitant administration of STF083010 significantly antagonized the aforementioned adverse effects. STF083010 exerted specific protective effects on reproductive injury-related effects in male rats exposed to PM2.5, with effects mediated via IRE1/JNK/autophagy signaling.
