Breast milk samples (100 μL) were submitted to a simple protein precipitation for the extraction of the analytes after the addition of deuterated internal standards (10 μL). A Kinetex C8 column was used for the separation of analytes with mobile phases composed of acetonitrile with 0.1 % formic acid and water with 0.1 % formic acid in gradient elution mode. Analytes were detected using an AB/SCIEX 4000 QTRAP instrument with positive electrospray ionization and operating in scheduled multiple reaction monitoring mode. Validation covered a large range of concentrations (0.5-500 ng/mL) for most of the analytes except bisoprolol, lacosamide, vilazodone (1-500 ng/mL), acid mycophenolic, letrozole, clomiphene (2-500 ng/mL) and hydroxy-melatonin (10-500 ng/mL). Within-run and between-run accuracy and precision for 4 levels of quality controls (QC) spiked at the lower limit of quantification (LLOQ), at 3 times the LLOQ, 50 % of the upper limit of quantification (ULOQ) and 80 % of the ULOQ were in agreement with the criteria from international guidelines. Matrix effect and extraction recovery ranged from 40.7 to 106.5 % and 87.3 to 110.8 %, respectively with relative standard deviations less than 15 %. Furthermore, all analytes were stable in breast milk at room temperature for 24 h, at -20 °C for two weeks, at -80 °C for 1 month, and after 3 freeze-thaw cycles. Using diffusion tensor imaging, this study explored premorbid, sex-specific white matter microstructural features that related to future emergence of major depressive disorder (MDD) during adolescence and young adulthood. Adolescents from the National Consortium on Alcohol and Neurodevelopment in Adolescence study, who were 12-21 years old at study entry and had not experienced major depression as of the baseline assessment, were selected for inclusion (N = 462, n = 223 female adolescents). Over five years of annual follow-up, 63 participants developed a diagnosis of MDD, as determined by the Computerized Semi-Structured Assessment for the Genetics of Alcoholism (n = 39 female adolescents). A whole-brain multivariate modeling approach was used to examine the relationship between fractional anisotropy (FA) at baseline and emergence into MDD, as a function of sex, controlling for age at baseline. Among female adolescents, those who developed MDD had significantly lower baseline FA in a portion of left precentral gyrus white matter, while male adolescents exhibited the opposite pattern. These results may serve as indirect microstructural markers of risk and targets for the prevention of depression during adolescence.Realizing the early diagnosis of acute liver injury has become a hotspot in recent years. Since the current indexes are not specific, developing novel probes with new recognition group remains a necessity. In this work, we developed a novel fluorescent probe, NNP, for the detection of ONOO- in acute liver injury (ALI). The in vitro evaluation of NNP indicated the advantages including high sensitivity (LOD = 0.13 μM), rapid response (48 h) conditions. Accordingly, NNP achieved the dose-dependent detection of ONOO- in human hepatic stellate LX-2 cells. Further, in the ALI model mice, NNP could monitor the ONOO- level in the occurrence of ALI in situ (in the liver region) with a steady performance. Subsequent immunohistochemical and imaging studies confirmed that NNP could achieve the detection of endogenous ONOO- in ALI liver tissues. This work introduced a practical implement for the ONOO- detection in ALI as well as a referable example for the establishment of molecular indexes in pre-clinical diagnosis.
Congenital or hypoplasia vaginal agenesis is a very rare condition caused by the failure of developmental Mullerian ducts. The prevalence is 0.001%-0.025% populations. This condition often misdiagnosed because the symptom does not appear. Acute symptoms such as abdominal pain may occur due to the obstruction of retrograde menstrual flow. In this case, we presented a case complex management of vaginal atresia with pyosalpinx, hematometra and bilateral hematosalpinx.

A 12years old teenager, non-sexually active, complained cyclic abdominal pain that worsening in seven months before admission. Patient never had menstrual blood flow during her life. Patient was diagnosed with hematometra, hematocolpos, bilateral hematosalpinx and distal vaginal agenesis. Amnion graft neovagina was performed. Five days after surgery, patient started to have fever. On the seventh days after surgery, amnion graft was removed. The next two days patient still had fever. Because of continuous fever, patient was test of COVID 19. The reconstructive surgery for vaginal agenesis with obstruction complications such as hematometra and hematosalpinx to prevent the worst condition like ascending infection or misdiagnosed other severe conditions.
The surgical intervention in vaginal agenesis must be considered as a treatment and not only focus on the reconstruction. Laparoscopy or laparotomy may offered as options for combination treatment with vaginal approach reconstructive surgery for vaginal agenesis with obstruction complications such as hematometra and hematosalpinx to prevent the worst condition like ascending infection or misdiagnosed other severe conditions.
