In summary, for the first time, our research identified COPT family genes at the whole-genome level to provide guidance for effectively improving the problem of Cu deficiency in the grass-livestock chain and provide theoretical support for the subsequent development of grass and animal husbandry.In this study, an active component UP1-1 was isolated from Chinese Huangshan Umbilicaria esculenta via hot water extraction and purified by anion-exchange and gel-filtration chromatography. UP1-1 mainly composed of galactose, mannose and glucose in a molar ratio of 0.81.04.6 with an average molecular weight of 281 kDa. Methylation analysis of UP1-1 revealed the major glycosidic bonds comprised 1,6-linked Glcp, 1,4-linked Glcp, t-linked Glcp, 1,3,6-linked Manp, 1,3-linked Galp, t-linked Galp at the ratio of 2.280.380.320.630.250.29. Structural analysis results revealed that the backbone of UP1-1 consisted of →6)-β-D-Glcp-(1→, →6)-β-D-Manp-(1→, →4)-β-D-Glcp-(1 → residues with side chains of →3)-β-D-Galp-(1→, β-D-Galp-(1 → and β-D-Glcp-(1 → branches located at O-3 position of →6)-β-D-Manp-(1→. Immunostimulatory activity tests showed that UP1-1 could promote the phagocytic activity and NO production of RAW 264.7 cells in a dose-dependent manner. UP1-1 could significantly improve the proliferation effect of RAW 264.7 cells at the concentration of 50 μg/mL. Thus, UP1-1 exerted good immunostimulatory activity, suggesting that UP1-1 has a great potential application in pharmacological industry.Starch-based isomalto/malto-polysaccharides (IMMPs) are soluble dietary fibres produced by the incubation of α-(1 → 4) linked glucans with the 4,6-α-glucanotransferase (GTFB) enzyme. In this study, we investigated the reaction dynamics of the GTFB enzyme by using isoamylase debranched starches as simplified linear substrates. Modification of α-glucans by GTFB was investigated over time and analysed with 1H NMR, HPSEC, HPAEC combined with glucose release measurements. We demonstrate that GTFB modification of linear substrates followed a substrate/acceptor model, in which α-(1 → 4) linked glucans DP ≥ 6 functioned as donor substrate, and α-(1 → 4) linked malto-oligomers DP less then 6 functioned as acceptor. The presence of α-(1 → 4) linked malto-oligomers DP less then 6 resulted in higher GTFB transferase activity, while their absence resulted in higher GTFB hydrolytic activity. The information obtained in this study provides a better insight into GTFB reaction dynamics and will be useful for α-glucan selection for the targeted synthesis of IMMPs in the future.The separation of lignin from woody biomass and subsequent conversion into useful products requires a solution to the problem of its solubility. The results indicate that the dissolution behavior of lignins follows the solubility parameters theory. We have developed a lignin dissolution model according to the lignin structure. This model obeys the solubility parameter theory, overcomes the limitations of the "like dissolves like" principle in organic acid-water systems, and provides a concise method for the selection of lignin solvent systems and the quantitative determination of their solvent composition.Most of the spray products in the market for wound healing applications are loaded with antibiotics that exert their antibacterial effect within the inflammatory stage of wound healing without demonstrating any effect in the subsequent proliferation stage. This study introduces a new aerosolized nanopowder (ANP) formula that not only exhibits antibacterial effect but also antioxidant and enhanced cell proliferation effects. Within the introduced ANP formula, Avicenna marina (Am) extract and neomycin (NM) antibiotic have been loaded within silk-fibroin nanoparticles (FB NPs). The Am has been extracted via different solvent systems, and investigated for its antioxidant and antibacterial activity as well as its ability to enhance cell proliferation. The physicochemical properties, size, zeta-potential and morphology of the prepared Am/FB NPs, NM/FB NPs and ANP formula were investigated. Besides, the ANP formula exhibited good antibacterial activities against Staphylococcus aureus, Methicillin resistant S. aureus, Pseudomonas aeruginosa and Resistant P. aeruginosa. Scratch wound healing assay on human fibroblast monolayers demonstrated 100% wound closure after 24 h upon using the ANP formula as compared to 70% wound closure for positive control (NM). The wound healing ability of the ANP formula has been further confirmed by histopathological evaluation of the wound site and depicted a marked increase in fibroblast proliferation and reduction of inflammatory cells after 15 days with a complete wound closure as compared to controls. The obtained results prove the beneficial effects of the Am extract on wound healing and introduce the developed multitask nanopowder formula as a potential wound healing spray.Alginate hydrogels are extremely versatile and flexible biomaterials, with an enormous potential for bio-applications use. Their similarity with extracellular matrix is a key factor in their performance for cell and tissue regeneration. In this study superabsorbent high porous hydrogels based on sodium alginate physical crosslinked with a natural crosslinker compound namely phytic acid were prepared and evaluated from the viewpoint of their specific properties. The resulting hydrogels obtained with different ratios between alginate and phytic acid were characterized by Fourier transform infrared spectroscopy technique, scanning electron microscopy, XRD measurements, swelling tests in physiological environment, and thermal analysis by using a simultaneous TG/FT-IR/MS system. There are put into evidence the differences in physico-chemical properties of the hydrogels in relation with their composition, which endows them tunable properties and versatility.
