The present study was to investigate whether dihydroartemisinin (DHA), which is a highly effective and safe drug in the treatment of malaria, could be repurposed for the treatment of skin fibrosis and vascular dysfunction in systemic sclerosis (SSc).
The value of DHA was determined using a bleomycin-induced model of skin fibrosis. mRNA transcriptome analysis was performed, and the targets of DHA on fibroblasts were identified. Immunofluorescence staining was used to identify dermal vessels undergoing endothelial-to-mesenchymal transition (EndoMT). Autophagic flux was detected by western blot and mRFP-GFP-LC3 adenovirus vector transfection.
Both systemic and topical administration of DHA decreased dermal thickness and collagen deposition and alleviated EndoMT in bleomycin-induced skin fibrosis mice model. Treatment of human umbilical vein endothelial cells (HUVECs) with TGF-β1 resulted in the acquisition of the activation marker (α-SMA) and loss of endothelial markers (CD31 and VE-cadherin), a process ths from bleomycin-induced EndoMT. Our study has elucidated the value of repurposing DHA for the treatment of SSc. Key Points • Oral or topical usage of DHA alleviated dermal fibrosis and EndoMT in bleomycin-induced skin fibrosis mice models. • DHA autophagy dependently inhibited fibroblast activation and collagen deposition via PI3K-ATK pathway. • DHA inhibited EndoMT of HUVECs induced by TGF-β1 by the downregulation of α-SMA and the upregulation of CD31 and VE-cadherin.
Hypnotics are one of the most frequently prescribed drugs worldwide, especially for women, and their chronic use may lead to tolerance, dosage escalation, dependence, withdrawal syndrome, and cognitive impairments, representing a significant public health problem. Consistent evidence from previous studies shows benefits of mindfulness-based interventions (MBIs) for substance use disorders (SUD) and insomnia. However, to date, there is a lack of research about effects of MBIs on reduction/cessation of chronic hypnotic use among women with insomnia.
The present randomized trial evaluated the efficacy of the 8-week group-delivered mindfulness-based relapse prevention (MBRP) program in an intervention group (IG, n = 34) compared with weekly phone monitoring only in the control group (CG, n = 36) in reducing hypnotic use and insomnia severity over a 6-month follow-up period.
There were significant differences between groups at baseline regarding hypnotic use but not insomnia. Group effects on hypnotic use were found immediately after the intervention (bT1 = 2.01, p < 0.001) and at the 2-month follow-up (bT2 = 2.21, p < 0.001), favoring the IG. The IG also had a greater reduction from baseline levels than the control group in insomnia severity at the 4-month (bT3 = 0.21, p = 0.045) and 6-month (bT4 = 0.32, p = 0.002) follow-ups.
The findings provide preliminary evidence of benefits of MBRP for reducing insomnia severity and potentially chronic hypnotic use. However, IG effects on chronic hypnotic use may have resulted from IG and control group differences in chronic hypnotic use at baseline.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection has been spreading rapidly around the world, while "multisystem inflammatory syndrome in children" (MIS-C) is a new type of syndrome that has now been reported in many countries. Similar and different characteristics between KD and MIS-C have been reported in a variety of literature. We aimed to focus on reviewing clinical presentations, diagnosis, and treatment of KD and MIS-C.
We searched articles in the electronic databases, including the Cochrane Library database, EMBASE, and MEDLINE with the keywords "multiple inflammatory syndrome" and/or "COVID-19" and/or "Kawasaki disease" and "children".
Main presentations of MIS-C and KD include fever, rashes, mucous membrane involvement, conjunctivitis, hands and feet erythema/edema, and cervical lymphadenopathy. However, compared with the highest incidence of KD among some Asian countries, MIS-C is common among Black and Hispanic children. MIS-C is common in older children and teenagers, e a good prognosis; however, the long-term clinical outcomes of MIS-C are not clear.
The overall presentation and treatment of MIS-C appear to overlap with KD. However, there are still great differences between the syndromes, and it is controversial to say whether MIS-C is a new entity or is a "severe type" of KD.
The overall presentation and treatment of MIS-C appear to overlap with KD. However, there are still great differences between the syndromes, and it is controversial to say whether MIS-C is a new entity or is a "severe type" of KD.The aim of the current work is to examine the concentrations of nine heavy metals such as chromium, nickel, copper, cadmium, zinc, cobalt, iron, lead, and metal (aluminium) in two body parts (carapace and walking legs) of Liocarcinus depurator from different stations (İlyas bey, Cide koyu, Anadolu feneri, igneada) in the western Black Sea coast of Turkey. And this study was carried out to examine morphological properties of Liocarcinus depurator in the region. The sex ratio (female/male) of 387 samples was calculated as F/M = 10.945. The mean carapace length and width were measured as 20.41 ± 2.41 mm and 25.10 ± 2.78 mm for female and 25.49 ± 3.33 mm and 32.32 ± 4.50 mm for male individually. The mean weight of crabs was measured as 4.09 ± 1.37 g for females and 8.33 ± 3.35 g for males. The samples were investigated trace metals by inductively coupled plasma-optical emission spectrometry (ICP-OES). selleck kinase inhibitor Ranges over trace metals in all samples of L. depurator as follows Cr 0.6-1.5, Mn 21-130, Cu 8-37, Zn 6-40, Cd 0.2-1.5, Pb 0.1-3.6, Co 0.1-1.1, Fe 22-160, and Ni 0.6-1 mg/kg wet wt. The walking legs and carapace/exoskeleton of female and male crabs were analyzed and contained various concentrations of Pb, Co, Ni, Cr, Cd, Fe, Cu, Zn, and Mn elements. The order of the heavy metal in all samples of L. depurator is shown as Fe > Cu > Mn > Zn > Cr > Cr > Ni > Cd > Pb > Co. In this research, metal levels of the crab samples compared between female and male and found highest mean metal concentrations in the station number 3 (Anadolu feneri) that the Al concentration in the carapace of female was 295.0 mg/kg wet wt and in the walking legs was 306.67 mg/kg wet wt, in the carapace of male was 264.4 mg/kg wet wt, and in the walking legs was 162.61 mg/kg wet wt in station number 3 (Anadolu feneri). The purpose of this work is to analyze the metal composition of this type of crab, which is abundant in the Black Sea region and is not planned to be attached to fisherman's nets, to evaluate whether it will be useful or not.
