The development of new instrumentation, such as droplet microfluidic platforms, may help to constrain our understanding of the kinetics of homogeneous freezing via the analysis of monodisperse, size-selected water droplets in temporally and spatially controlled environments. Here, we evaluate droplet freezing data obtained using the Lab-on-a-Chip Nucleation by Immersed Particle Instrument (LOC-NIPI), in which droplets are generated and frozen in continuous flow. This high-throughput method was used to analyse over 16,000 water droplets (86 μm diameter) across three experimental runs, generating data with high precision and reproducibility that has largely been unrepresented in the microfluidic literature. Using this data, a new LOC-NIPI parameterisation of the volume nucleation rate coefficient (JV(T)) was ets.The ATP-competitive inhibitors of Hsp90 have been tested predominantly in kinase addicted cancers; however, they have had limited success. A mechanistic connection between Hsp90 and oncogenic K-Ras is not known. Here, we show that K-Ras selectivity is enabled by the loss of the K-Ras membrane nanocluster modulator galectin-3 downstream of the Hsp90 client HIF-1α. Gemcitabine mouse This mechanism suggests a higher drug sensitivity in the context of KRAS mutant, HIF-1α-high and/or Gal3-high cancer cells, such as those found, in particular, in pancreatic adenocarcinoma. The low toxicity of conglobatin further indicates a beneficial on-target toxicity profile for Hsp90/Cdc37 interface inhibitors. We therefore computationally screened >7 M compounds, and identified four novel small molecules with activities of 4 μM-44 μM in vitro. All of the compounds were K-Ras selective, and potently decreased the Hsp90 client protein levels without inducing the heat shock response. Moreover, they all inhibited the 2D proliferation of breast, pancreatic, and lung cancer cell lines. The most active compounds from each scaffold, furthermore, significantly blocked 3D spheroids and the growth of K-Ras-dependent microtumors. We foresee new opportunities for improved Hsp90/Cdc37 interface inhibitors in cancer and other aging-associated diseases.Pedagogical tools are needed that link multidisciplinary nanoscience and technology (NST) to multiple state-of-the-art applications, including those requiring new fabrication routes relying on green synthesis. These can both educate and motivate the next generation of entrepreneurial NST scientists to create innovative products whilst protecting the environment and resources. Nanoporous silicon shows promise as such a tool as it can be fabricated from plants and waste materials, but also embodies many key educational concepts and key industrial uses identified for NST. Specific mechanical, thermal, and optical properties become highly tunable through nanoporosity. We also describe exceptional properties for nanostructured silicon like medical biodegradability and efficient light emission that open up new functionality for this semiconductor. Examples of prior lecture courses and potential laboratory projects are provided, based on the author's experiences in academic chemistry and physics departments in the USA and UK, together with industrial R&D in the medical, food, and consumer-care sectors. Nanoporous silicon-based lessons that engage students in the basics of entrepreneurship can also readily be identified, including idea generation, intellectual property, and clinical translation of nanomaterial products.Background The objectives of this study were to describe the oral health status in the institutionalized geriatric population in an area of southern Italy and to identify the impact of oral health on the Oral Health Related Quality of Life (OHRQoL). Methods Data were collected from individuals aged ≥60 years in randomly selected Calabrian long-term care facilities. The dental health status was assessed recording the decayed, missing, or filled dental elements due to the carious lesions (DMFT) index, the presence of visible dental plaque, and the gingival condition. The influence of the dental health status on the self-perceived value of life was assessed using the Geriatric Oral Health Assessment Index (GOHAI). Results Among the 344 elderly individuals included, 18.4% reported frequent tooth-brushing, and only 39.9% reported the need of dental care. The DMFT index was 26.4. Less than a third of the participants had a GOHAI score of ≤50 which is suggestive of highly compromised OHRQoL. The GOHAI score was significantly better for elderly individuals with no self-perceived need of dental care and with a lower DMFT index. Conclusions The burden of oral conditions among residents in long-term care facilities was considerable, with a high prevalence of missing teeth and dentures. Strategies targeting care providers are needed.Continuous orthogonal moments, for which continuous functions are used as kernel functions, are invariant to rotation and scaling, and they have been greatly developed over the recent years. Among continuous orthogonal moments, polar harmonic Fourier moments (PHFMs) have superior performance and strong image description ability. In order to improve the performance of PHFMs in noise resistance and image reconstruction, PHFMs, which can only take integer numbers, are extended to fractional-order polar harmonic Fourier moments (FrPHFMs) in this paper. Firstly, the radial polynomials of integer-order PHFMs are modified to obtain fractional-order radial polynomials, and FrPHFMs are constructed based on the fractional-order radial polynomials; subsequently, the strong reconstruction ability, orthogonality, and geometric invariance of the proposed FrPHFMs are proven; and, finally, the performance of the proposed FrPHFMs is compared with that of integer-order PHFMs, fractional-order radial harmonic Fourier moments (FrRHFMs), fractional-order polar harmonic transforms (FrPHTs), and fractional-order Zernike moments (FrZMs).
