Flow cytometrists must extrapolate information from relatively few markers. We evaluated the expression pattern of the panleukocyte markers CD18 and CD44 on leukocyte (white blood cell, WBC) subclasses in the peripheral blood (PB) of 14 healthy cats. The degree of expression of CD18 and CD44 was calculated as the ratio between the median fluorescence intensity (MFI) value of antibody-stained cells and autofluorescence. All samples were acquired with the same cytometer with constant photomultiplier setting and compensation matrices. Both molecules were expressed at higher levels on monocytes, intermediate levels on polymorphonuclear cells (PMNs), and lower levels on lymphocytes. CD18-MFI discriminated well among the 3 populations, whereas CD44-MFI mostly overlapped between monocytes and PMNs. However, CD44-MFI had a lower intra-population variability. Evaluation of CD18 and CD44, together with morphologic parameters, was useful for discriminating among WBC subclasses in healthy cats. This information may be helpful for future studies given that an increase in CD18-MFI may indicate reactive changes, whereas fluctuations in CD44-MFI may suggest neoplasia.This study examined the effect of curcumin on T-helper (Th17) and T-regulatory (Treg) cells regarding the mRNA of cytokines/mediators in the gingiva. Thirty-five male albino Wistar rats were divided into four groups Group 1 periodontitis (n = 9); Group 2 periodontitis with curcumin treatment (n = 8); Group 3 periodontally healthy with curcumin treatment (n = 10); and Group 4 periodontally healthy (n = 8). Curcumin was administered via oral gavage (30 mg/kg/day) for a total of 15 days. The gingival tissues were investigated regarding mRNA expressions of Th17/Treg cytokines with qRT-PCR. The distributional properties of the data were evaluated using the Anderson-Darling normality test. Kruskal-Wallis and Mann-Whitney U tests were employed for multiple group comparisons. Partial least squares regression discriminant analysis (PLS-DA) was used to evaluate the degree of contribution of each mRNA to the separation of treatment groups. When the periodontitis groups were compared, curcumin treatment resulted in lower IL-1β (Group 2 median 0.002, Group 1 median 0.12) and IL-6 (Group 2 median 0.031, Group 1 median 0.078) and higher IL-17 (Group 2 median 1.07, Group 1 median 0.583) relative mRNA expression in Group 2 than in Group 1 (p less then 0.001). Group 3 also had higher IL-10 relative expression (median 0.067) than Groups 1 and 4 (median 0.028, 0.007, respectively. p less then 0.001). These results indicate that curcumin might be a promising agent for the prevention and/or treatment of periodontal diseases due to its decreasing effect on IL-1β and IL-6 mRNA expression.
To describe clinical, anterior segment optical coherence tomography (AS-OCT), in vivo confocal microscopy (IVCM), histopathologic, and immunohistochemical findings in a patient with multiple endocrine neoplasia type 2b (MEN 2b) syndrome.

Retrospective case report of a patient with MEN 2b.

A 31-year-old male diagnosed with MEN 2b presented with eye redness, burning, and visible conjunctival mass in both eyes. The patient's past medical history revealed that he underwent bilateral adrenalectomy and total thyroidectomy. Genetic testing revealed M918T heterozygous mutation in the RET proto-oncogene. Corrected visual acuity was 20/20 in both eyes. Anterior segment examination revealed bilateral thickened lid margins, ectropion, blepharitis, conjunctival injection, temporal and inferonasal subconjunctival lesions with corneal invasion, corneal neovascularization, and marked corneal nerves. AS-OCT showed a subepithelial mixed reflective lobular mass in both eyes. Hyperreflective and noticeable thickened stromal corneal nerves were observed on IVCM in the left eye. After incisional biopsy of the right perilimbal lesions, histopathological examination revealed that lesions consisted of spindle cells in hypocellular bundles with no atypia and mitosis. Immunohistochemical examination revealed diffuse staining with S100, focal staining with synaptophysin, and no staining with neurofilament protein. These findings were considered compatible with a benign nerve sheath tumor, probably schwannoma.

We present clinical, AS-OCT, IVCM, histopathological, and immunohistochemical findings in a patient with MEN 2b. To our knowledge, this is the first case of a conjunctival schwannoma diagnosed histopathologically in MEN 2b.
