Interestingly, SPL9-RNAi plants accumulated more stress-alleviating anthocyanin compared to WT under both drought and well-watered control conditions, suggesting that MsSPL9 may contribute to drought tolerance in alfalfa, at least in part, by regulating anthocyanin biosynthesis. The results suggest that targeting MsSPL9 is a suitable means for improving alfalfa resilience towards drought conditions.Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is known to behave as an attractive anti-cancer agent in various cancers. Despite its promise TRAIL has limitations such as short half-life and rapid development of resistance. In this regard, approaches to sensitizers of TRAIL that can overcome the limitations of TRAIL are necessary. However, the molecular targets and mechanisms underlying sensitization to TRAIL-induced apoptosis are not fully understood. Here, we propose that reactive oxygen species modulator-1 (Romo1) as an attractive sensitizer of TRAIL. Romo1 is a mitochondrial inner membrane channel protein that controls reactive oxygen species (ROS) production, and its expression is highly upregulated in various cancers, including colorectal cancer. In the present study, we demonstrated that Romo1 inhibition significantly increased TRAIL-induced apoptosis of colorectal cancer cells, but not of normal colon cells. The combined effect of TRAIL and Romo1 inhibition was correlated with the activation of mitochondrial apoptosis pathways. Romo1 silencing elevated the protein levels of BCL-2-associated X protein (Bax) by downregulating the ubiquitin proteasome system (UPS). Romo1 inhibition downregulated the interaction between Bax and Parkin. Furthermore, Romo1 knockdown triggered the mitochondrial dysfunction and ROS generation. We validated the effect of combination in tumor xenograft model in vivo. In conclusion, our study demonstrates that Romo1 inhibition induces TRAIL-mediated apoptosis by identifying the novel mechanism associated with the Bax/Parkin interaction. We suggest that targeting of Romo1 is essential for the treatment of colorectal cancer and may be a new therapeutic approach in the future and contribute to the drug discovery.This study investigated the effect of diets negative in dietary cation-anion difference (DCAD) or restricted in Ca fed prepartum to dairy cows for three weeks on colostrum yield and composition, and the health and growth performance of their calves. Thirty-six pregnant non-lactating Holstein-Friesian cows were randomly assigned to three isoenergetic diets (1) low Ca 0.24% Ca, DCAD +86 mEq/kg; (2) high Ca 1.23% Ca, DCAD +95 mEq/kg; and (3) low DCAD 1.28% Ca, DCAD -115 mEq/kg (all dry matter (DM) basis). While colostrum quality was not affected, low Ca supply prepartum tended to increase the colostrum yield compared to high Ca (low Ca = 8.81 vs. high Ca = 5.39 kg). However, calves from cows fed low DCAD showed higher serum concentrations of K, lower body weight (BW), starter feed intake and average daily weight gain before weaning compared to low Ca and high Ca calves (53.12 vs. 57.68 and 57.32 kg) but BW was similar postweaning (d 70). In addition, calves from dams fed low DCAD were more likely to develop diarrhea and had increased number of days with abnormal fecal scores. Consequently, calves from low DCAD dams had to be treated more frequently.This study analysed the effect of three alkalizing soil amendments (limestone, dolomite chalcedonite) on aided phytostabilization with Festuca rubra L. depending on the hexavalent chromium (Cr(VI)) level in contaminated soil. Four different levels of Cr(VI) were added to the soil (0, 50, 100 and 150 mg/kg). The Cr contents in the plant roots and above-ground parts and the soil (total and extracted Cr by 0.01 M CaCl2) were determined with flame atomic absorption spectrometry. The phytotoxicity of the soil was also determined. Soil amended with chalcedonite significantly increased F. rubra biomass. Chalcedonite and limestone favored a considerable accumulation of Cr in the roots. The application of dolomite and limestone to soil contaminated with Cr(VI) contributed to a significant increase in pH values and was found to be the most effective in reducing total Cr and CaCl2-extracted Cr contents from the soil. F. rubra in combination with a chalcedonite amendment appears to be a promising solution for phytostabilization of Cr(VI)-contaminated areas. The use of this model can contribute to reducing human exposure to Cr(VI) and its associated health risks.The rotating backward extrusion (RBE) process, as a new severe plastic deformation (SPD) process, is based on conventional backward extrusion and rotation, which meets the requirement of modern industrial development with its high performance and production efficiency. However, there is little research on the microstructure evolution and texture modification of the RBE process. Thus, in this study, the effect of different rotating revolutions, e.g., n = 5, n = 10, and n = 50, on the microstructure and texture development for the RBE process based on the AZ80 magnesium (Mg) alloy were investigated at 653 K. The results disclose that the rotating revolution is an influencing processing parameter on the deformation of the RBE process. The grain refining ability is enhanced with the increase of the rotating revolutions, and the minimum grain size of the cup bottom, shearing zone, and cup wall can reach to 16.7 μm, 15.6 μm, and 13.0 μm, respectively, under the condition of n = 50. PF-3644022 Furthermore, with the increase in the rotating revolutions, the microstructure of the alloy becomes more uniform and the proportion of dynamic recrystallization (DRX) is also increased. The maximum DRX fractions of the sample for the cup bottom and cup wall are 95.4% and 86.8%, respectively, at n = 50. The DRX mechanism of the RBE process is determined by the continuous DRX and discontinuous DRX. In addition, the texture can be significant weakened during the RBE process, especially at the cup bottom, where the maximum pole intensity can be reduced from 17.6 at n = 10 to 6.5 at n = 50, which can be attributed to the higher proportion of new DRXed grains whose orientations are more random compared with the deformed grains.
