Here, we describe a new rat EVLP model that addresses this need, while enabling the study of lung injury due to cold ischemic time (CIT). The technique involves perfusing and ventilating male Lewis rat donor lungs for 4h before transplanting the left lung into a recipient rat, then evaluating lung function 2h after reperfusion. To test injury within this model,lungswere divided into groups and exposed to different CITs (ie, 20 min, 6h, 12h, 18h and 24h).Experiments involving the 24h CIT group were prematurely terminated due to the development of severe edema. For the other groups, no differences in the ratio of arterial oxygen partial pressure to fractional inspired oxygen (PaO2/FiO2) were observed during EVLP; however, lung compliance decreased over time in the 18h group (P=0.012) and the PaO2/FiO2of the blood from the left pulmonary vein 2h after transplantation was lower compared to 20 min CIT group (P=0.0062).This new model maintained stable lung function during 4h EVLP and after transplantation when exposed to up to 12h of CIT.Femur component fracture is very rare with Oxford® unicompartmental knee replacement. Then, participants rated their level of comfort disclosing diagnostic terms to parents/caregivers. Participants provided rationale for their ratings and answered various questions regarding academic and clinical experiences to identify factors that may have influenced ratings. Results SLPs were significantly less likely to feel comfortable using the term "stuttering" compared to other communication disorders. Thematic responses revealed increased experience with a specific speech-language population was related to higher comfort levels with using its diagnostic term. Additionally, knowing a person who stutters predicted greater comfort levels as compared to other clinical and academic experiences. Thus, our objective was to explore the first pressure injury and characteristics of individuals who develop subsequent pressure injuries during acute care and inpatient rehabilitation following spinal cord injury.Design Secondary analysis of longitudinal data from a cohort of adults following initial traumatic spinal cord injury.Setting Urban acute care hospital and inpatient rehabilitation facilities.Participants A convenience sample of adults (n = 38) who developed at least one pressure injury during acute care and inpatient rehabilitation.Interventions Not applicable.Outcome Measures The primary outcomes were number of additional pressure injuries and stage of care during which they occurred, prior to community discharge.Results A covariate-adjusted model revealed that participants with ASIA D injury had a 67% decrease in the rate of additional pressure injury incidence compared to participants with ASIA A injury (Rate Ratio = .33, 95% CI [0.13, 0.88]). The severity of the first pressure injury had no significant association with subsequent pressure injury incidence (P = .10).Conclusion These findings indicated that individuals with greater sensory and motor loss had an increased risk of developing additional pressure injuries compared to individuals with less impairment. These results are meaningful for stakeholders interested in understanding factors associated with developing subsequent pressure injuries during the index rehabilitation stay and provide a foundation for future research in this area.Rationale Peripheral artery disease, common in metabolic syndrome and diabetes, responds poorly to medical interventions, and is characterized by chronic vessel immaturity leading to lower extremity amputations. Objective To define the role of reversible palmitoylation at the endothelium in the maintenance of vascular maturity. Methods and Results Endothelial knockout of the depalmitoylation enzyme acyl-protein thioesterase 1 (APT1) in mice impaired recovery from chronic hindlimb ischemia, a model of peripheral artery disease. Endothelial APT1 deficiency decreased fibronectin processing, disrupted adherens junctions, and inhibited in vitro lumen formation. In an unbiased palmitoylation proteomic screen of endothelial cells from genetically modified mice, R-Ras, known to promote vessel maturation, was preferentially affected by APT1 deficiency. R-Ras was validated as an APT1 substrate, and click chemistry analyses demonstrated increased R-Ras palmitoylation in cells with APT1 deficiency. APT1 enzyme activity was decreased in endothelial cells from db/db mice.
