Efficient prediction of the progression of mild cognitive impairment (MCI) to Alzheimer's disease (AD) is important for the early intervention and management of AD. The aim of our study was to develop a longitudinal structural magnetic resonance imaging-based prediction system for MCI progression.
A total of 164 MCI patients with longitudinal data were collected from the Alzheimer's Disease Neuroimaging Initiative (ADNI). After preprocessing, a discriminative dictionary learning framework was applied to differentiate MCI patches, avoiding the segmentation of regions of interest. Then, the proportion of patches classified as more severe atrophy patches in a patient was calculated as his or her feature to be input into a simple support vector machine. Finally, a new subject was predicted with fourfold cross-validation (CV), and the area under the receiver operating characteristic curve (AUC) was determined.
The average accuracy and AUC values after fourfold CV were 0.973 and 0.984, respectively. The effects of the data from one or two time points were also investigated.
The proposed prediction system achieves desirable and reliable performance in predicting progression for MCI patients. Additionally, the prediction of MCI progression with longitudinal data was more effective and accurate.

The developed scheme is expected to advance the clinical research and treatment of MCI patients.Sex estimation is an important part of creating a biological profile, and ultimately assisting in creating a presumptive identification of unidentified skeletal remains. However, manual methods of anthropometric are time-consuming and prone to observer variability. The present study is an attempt to estimation of sex from automatic measurement of patella by multidetector computed tomography (MDCT) in a contemporary Chinese population. Four measurements for every patella, including maximum height (MAXH), maximum breadth (MAXB), maximum thickness (MAXT) and patellar volume (PV), were automatically provided by the software from CT image of 300 Chinese. The sample is composed of 156 males and 144 females with an average age of 41.44 and 45.68 years, respectively. The statistical analyses showed that all variables were sexually dimorphic. Receiver operating characteristic (ROC) analysis was performed to estimate sex from patella. The univariate analysis of each patellar parameter yielded a sex classification accuracy rate of 73.1% to 85.7%. The classification accuracy rates of sex estimation using the combination of the patellar parameters are 81.9% to 91.6%. This paper provides indications that the patella is important bone for sex estimation and it may be used as an alternative in forensic cases when the skull and pelvis are unavailable.Etiological models of the development of eating disorders among midlife and aging men are lacking. The present study provides a review and integration of existing theoretical frameworks and empirical data relevant to the examination of eating pathology in this population and introduces an integrated model of risk and protective factors for the development of eating disorder symptoms. Several etiological models of eating pathology that have gained some empirical support, or present relevance to this population, are briefly reviewed including sociocultural theory, objectification theory, social identity theory, positive body image and the developmental theory of embodiment, and existential and terror management theory, and finally biologically based theories. #link# In addition, these frameworks were extended to account for developmental and gender-specific factors that may influence the development of eating disorders. An integrated model of risk and protective factors that brings together critical elements of these extended theoretical frameworks is presented, with the goal of guiding and stimulating research in this neglected group.Identification of safe and effective compounds to increase or activate UCP1 expression in brown or white adipocytes remains a potent therapeutic strategy to combat obesity. Here we reported that, glyburide, one of the FDA-approved drugs currently used to treat type 2 diabetes, can significantly enhance UCP1 expression in both brown and white adipocytes. Glyburide-fed mice exhibited a clear resistance to high-fat diet-induced obesity, reduced blood triglyceride level, and increased UCP1 expression in brown adipose tissue. Moreover, in situ injection of glyburide to inguinal white adipose tissue remarkably enhanced UCP1 expression and increased thermogenesis. Further mechanistic studies indicated that the glyburide effect in UCP1 expression in adipocytes was KATP channel independent but may involve the regulation of the Ca2+-Calcineurin-NFAT signal pathway. Overall, our findings revealed the significant effects of glyburide in regulating UCP1 expression and thermogenesis in adipocytes, which can be potentially repurposed to treat obesity.In bacteria, the dissociable σ subunit of the RNA polymerase (RNAP) is responsible for initiating RNA synthesis from specific DNA sites. As nascent RNA grows, downstream DNA unwinds and is pulled into the RNAP, causing stress accumulation and initiation complex destabilization. Processive transcription elongation requires at least partial separation of the σ factor from the RNAP core enzyme. Here, we present a series of transcription complexes captured between the early initiation and elongation phases via in-crystal RNA synthesis and cleavage. Crystal structures of these complexes indicate that stress accumulation during transcription initiation is not due to clashing of the growing nascent RNA with the σ3.2 loop, but results from scrunching of the template strand DNA that is contained inside the RNAP by the σ3 domain. AP20187 shed light on how scrunching of template-strand DNA drives both abortive initiation and σ-RNAP core separation to transition transcription from initiation to elongation.Transient receptor potential vanilloid 6 (TRPV6), a calcium-selective channel possessing six transmembrane domains (S1-S6) and intracellular N and C termini, plays crucial roles in calcium absorption in epithelia and bone and is involved in human diseases including vitamin-D deficiency, osteoporosis, and cancer. The TRPV6 function and regulation remain poorly understood. Here we show that the TRPV6 intramolecular S4-S5 linker to C-terminal TRP helix (L/C) and N-terminal pre-S1 helix to TRP helix (N/C) interactions, mediated by Arg470Trp593 and Trp321Ile597 bonding, respectively, are autoinhibitory and are required for maintaining TRPV6 at basal states. Disruption of either interaction by mutations or blocking peptides activates TRPV6. The N/C interaction depends on the L/C interaction but not reversely. Three cationic residues in S5 or C terminus are involved in binding PIP2 to suppress both interactions thereby activating TRPV6. This study reveals "PIP2 - intramolecular interactions" regulatory mechanism of TRPV6 activation-autoinhibition, which will help elucidating the corresponding mechanisms in other TRP channels.
