Although significant achievements have led to advanced therapeutic success with targeted therapy options, more efforts are required for different subtypes of tumors and according to genomic, transcriptomic, and proteomic alterations. This study underlines the role of microRNA-21 (miR-21) in metastatic MDA-MB-231 breast cancer cells. Following the knockout of miR-21 from MDA-MB-231 cells, which have the highest miR-21 expression levels compared to MCF-7 and SK-BR-3 BCa cells, a decrease in epithelial-mesenchymal transition (EMT) via downregulation of mesenchymal markers was observed. Wnt-11 was a critical target for miR-21, and the Wnt-11 related signaling axis was altered in the stable miR-21 knockout cells. miR-21 expression was associated with a significant increase in mesenchymal markers in MDA-MB-231 BCa cells. Furthermore, the release of extracellular vesicles (EVs) was significantly reduced in the miR-21 KO cells, alongside a significant reduction in relative miR-21 export in EV cargo, compared with control cells. We conclude that miR-21 is a leading factor involved in mesenchymal transition in MDA-MB-231 BCa. Future therapeutic strategies could focus on its role in the treatment of metastatic breast cancer.Acoustic emission (AE) phenomenon has a direct relationship with the interaction of tool and material which makes AE the most sensitive one among various process variables. However, its prominent sensitivity also means the characteristics of random and board band. Feature representation is a difficult problem for AE-based monitoring and determines the accuracy of monitoring system. It is knottier for the situation of using diamond wheel grinding optical components, not only because of the complexity of grinding process but also the high requirement on surface and subsurface quality. This paper is dedicated to AE-based condition monitoring of diamond wheel during grinding brittle materials and feature representation is paid more attention. AE signal of brittle-regime grinding is modeled as a superposition of a series of burst-type AE events. Theory analysis manifested that original time waveform and frequency spectrum are all suitable for feature representation. Considering the convolution form of b-AE in time domain, a convolutional neural network with original time waveform of AE signals as the input is built for multi-class classification of wheel state. Detailed state division in a wheel's whole life cycle is realized and the accuracy is over 90%. Different from the overlapping in time domain, AE components of different crack mechanisms are probably separated in frequency domain. From this point of view, AE spectrums are more suitable for feature extraction than the original time waveform. In addition, the time sequence of AE samples is essential for the evaluation of wheel's life elapse and making use of sequential information is just the idea behind recurrent neural network (RNN). Therefore, long short-term memory (LSTM), a special kind of RNN, is used to build a regression prediction model of wheel state with AE spectrums as the model input and satisfactory prediction accuracy is acquired on the test set.Chikungunya (CHIKV) and Dengue (DENV) viruses cause an acute febrile illness which is hard to clinically differentiate and treat since both exhibit similar symptoms. Hence, this study was aimed at identifying the expression profiles of cytokines on co-infected samples and compare with CHIKV and DENV mono-infected samples. Serum samples of 292 suspected patients during 2009-2011 were analyzed. The presence of primary (IgM)/secondary (IgG) antibodies and early NS1 Dengue antigens were confirmed by capture ELISA. Molecular diagnosis and serotypes were discriminated by RT-PCR, confirmed by sequencing. All the plasma samples were assayed for cytokine expression by BDTM cytometry bead array (CBA) and compared with independent mono-infection viral load. Among the tested samples, 82 were confirmed as Dengue positive; 52 through IgM (17.8%), and 30 through IgG (10.2%). Additionally, 186 samples were confirmed as Chikungunya, 96 through IgM (32.6%) and 92 through IgG (31.5%) ELISA, respectively. Interestingly, 19 patients were co-infection positive in which, only 6 were confirmed for CHIKV and 7 for DENV by RT-PCR. Among 8 cytokines, IL-2, IL-8, IFNα, IFN γ, and IL-12 were found to be significantly different between co-infected and CHIKV mono-infected patients and correlated with viral load. DENV viral load was correlated with cytokine expression and a significant difference in IL-2 and IL-12 was observed between DENV mono-infected and DENV and CHIKV co-infected patients. Results indicated that apart from serological and molecular confirmation, cytokines could be used as a specific biomarker for the diagnosis of DENV and CHIKV. In the future, the role of independent cytokines can be determined to understand the pathogenesis and etiology of these dreadful diseases.Cardiovascular fitness is linked to better executive functions, preserved gait speed, and efficient cortical activity. Older adults with cardiovascular risk factors (CVRFs) typically show poor cognitive performance, low physical fitness, and altered brain functioning compared with healthy individuals. In the current study, the impact of regular physical activity on cognition, locomotion, and brain functions was explored in a cohort of older adults with low or high CVRFs. Cortical activation of the frontal areas was investigated using functional Near-Infrared Spectroscopy (fNIRS) at baseline, at 6 months and at 12 months. Evoked cortical response and behavioral performance were assessed using the dual-task walking paradigm, consisting of three conditions single cognitive task (2-back task), single walking task (walking), and dual-task (2-back whilst walking).
