SEC1A's presence in the pollen tube cytoplasm was widespread, but its density peaked at the pollen tube's distal end. Our co-immunoprecipitation-mass spectrometry-based protein interaction analysis, combined with in vivo microscopy, demonstrated that the exocyst subunit SEC6 interacts with SEC1A, impacting both their distribution and secretion rate at the tip of the pollen tube. Concurrently, the loss of SEC1A and SEC6 functionality substantially altered the localization of the SNARE (soluble N-ethylmaleimide-sensitive factor attachment protein receptor) complex protein SYP125 at the pollen tube tip, resulting in irregularities within the pollen tube cell wall structure. Compared to their respective single mutants, the sec1a sec6 double mutant displayed a noticeably more pronounced pollen tube-related phenotype, as determined by genetic analysis.
