Crosstalk between cellular pathways is often mediated through scaffold proteins that function as platforms for the assembly of signaling complexes. BIX 01294 clinical trial Based on yeast two-hybrid analysis, we report here the interaction between two complex scaffold proteins, CREB-binding protein (CBP) and the Ras GTPase-activating-like protein 1 (IQGAP1). Dissection of the interaction between the two proteins reveals that the central, thus far uncharacterized, region of IQGAP1 interacts with the HAT domain and the C-terminal intrinsically disordered region of CBP (termed ID5). Structural analysis of ID5 by solution NMR spectroscopy and SAXS reveals the presence of two regions with pronounced helical propensity. The ID5 region(s) involved in the interaction of nanomolar affinity were delineated by solution NMR titrations and pull-down assays. Moreover, we found that IQGAP1 acts as an inhibitor of the histone acetyltransferase (HAT) activity of CBP. In in vitro assays, the CBP-binding region of IQGAP1 positively and negatively regulates the function of HAT proteins of different families including CBP, KAT5 and PCAF. As many signaling pathways converge on CBP and IQGAP1, their interaction provides an interface between transcription regulation and the coordination of cytoskeleton. Disruption or alteration of the interaction between these scaffold proteins may lead to cancer development or metastatic processes, highlighting the importance of this interaction.An amendment to this paper has been published and can be accessed via a link at the top of the paper.Household air pollution has adverse effects on cardiovascular health. One of the major sources of household air pollutants is the combustion of cooking oils during cooking. Trans, trans-2,4-decadienal (tt-DDE) is a type of dienaldehyde that is present in a wide range of food and food products. It is a byproduct of the peroxidation of linoleic acid following the heating of oil during cooking. The mechanisms of the associations between household air pollution and cardiac arrhythmias are currently unclear. The purpose of this study was to determine effects of tt-DDE on the ion currents in H9c2 cells. The IK and ICa,L in H9c2 cells treated with and without tt-DDE were measured using the whole-cell patch clamp method. Expressions of Kv2.1 and Cav1.2 in H9c2 cells treated with and without tt-DDE were measured by western blot analysis. After the H9c2 cells had been exposed to tt-DDE, the IK and ICa,L were significantly decreased. The expression of Kv2.1, unlike that of Cav1.2, was also significantly decreased in these cells. These changes in IK and ICa,L that were induced by tt-DDE may help to explain the association between cardiac arrhythmogenesis and cooking-oil fumes.Social calls have the function to coordinate the behavior of animals. In the presence of conspecifics foraging Common pipistrelle bats (P. pipistrellus) emitted, in addition to typical echolocation signals, two types of social calls complex social calls and an as-of-yet undescribed, short, frequency-modulated call type with high terminal frequency, which we term "high frequency social call". By recording the flight and acoustic behavior of free flying pairs of foraging P. pipistrellus with an array of four microphones we were able to determine their three-dimensional flight paths and attribute emitted calls to particular behavioral situations. Complex social calls were emitted at further inter-individual distances and at large bearing angles to conspecifics, whereas high frequency social calls were produced at significantly shorter distances and at smaller bearing angles. These calls were associated with chasings and the eviction of the intruder. We assume that the emission of both types of social calls by foraging bats reflects a two-stage-process of the occupation and defense of a food patch. Common pipistrelle bats use complex social calls to claim a food patch and switch to agonistic behaviors, including chasings and high frequency social call emissions, when they defend their foraging territory against an intruder.Gout is a chronic arthritis caused by the deposition of poorly soluble monosodium urate monohydrate (MSU) crystals in peripheral joints. Resident macrophages initiate inflammation in response to MSU mediated by NF-κB nuclear translocation and NLRP3 inflammasome activation. We investigated the role of CD44, a transmembrane receptor, in mediating MSU phagocytosis by macrophages. We used an antibody that sheds the extracellular domain (ECD) of CD44 to study the role of the receptor and its associated protein phosphatase 2A (PP2A) in macrophage activation. We also studied the significance of CD44 in mediating MSU inflammation in-vivo. Cd44-/- BMDMs showed reduced MSU phagocytosis, LDH release, IL-1β expression and production compared to Cd44+/+ BMDMs. Elevated CD44 staining was detected intracellularly and CD44 colocalized with α-tubulin as a result of MSU exposure and ECD-shedding reduced MSU phagocytosis in murine and human macrophages. Anti-CD44 antibody treatment reduced NF-κB p65 subunit nuclear levels, IL-1β expression, pro-IL-1β and IL-8 production in MSU stimulated THP-1 macrophages (p  less then  0.01). The effect of the antibody was mediated by an enhancement in PP2A activity. CD44 ECD-shedding reduced the conversion of procaspase-1 to active caspase-1, caspase-1 activity and resultant generation of mature IL-1β in macrophages. Neutrophil and monocyte influx and upregulated production of IL-1β was evident in wildtype mice. MSU failed to trigger neutrophil and monocyte recruitment in Cd44-/- mice and lower IL-1β levels were detected in peritoneal lavages from Cd44-/- mice (p  less then  0.01). Anti-CD44 antibody treatment reduced neutrophil and monocyte recruitment and resulted in reduced lavage IL-1β levels in the same model. CD44 plays a biologically significant role in mediating phagocytosis of MSU and downstream inflammation and is a novel target in gout treatment.The widespread applications of high-throughput sequencing technology have produced a large number of publicly available gene expression datasets. However, due to the gene expression datasets have the characteristics of small sample size, high dimensionality and high noise, the application of biostatistics and machine learning methods to analyze gene expression data is a challenging task, such as the low reproducibility of important biomarkers in different studies. Meta-analysis is an effective approach to deal with these problems, but the current methods have some limitations. In this paper, we propose the meta-analysis based on three nonconvex regularization methods, which are L1/2 regularization (meta-Half), Minimax Concave Penalty regularization (meta-MCP) and Smoothly Clipped Absolute Deviation regularization (meta-SCAD). The three nonconvex regularization methods are effective approaches for variable selection developed in recent years. Through the hierarchical decomposition of coefficients, our methods not only maintain the flexibility of variable selection and improve the efficiency of selecting important biomarkers, but also summarize and synthesize scientific evidence from multiple studies to consider the relationship between different datasets.
